In Pinus peuce zygotic embryo culture grown on Gresshoff and Doy (1972; GD) basal medium, 2.22 μM benzyladenine (BA) was superior in promoting adventitious bud induction during 4 weeks comparing to ...kinetin or BA + kinetin. Shoot elongation was achieved on half-strength GD medium devoid of plant growth regulators and containing activated charcoal. Pulse treatment with 1 mM indole-3-butyric acid (IBA) for 2 h, followed by transfer to half-strength GD medium, produced the most efficient rooting. Rooted shoots were transplanted to the greenhouse and plantlets continued to grow and developed into phenotypically normal plants. Up to 10 plants per explant can be obtained within 36 weeks from culture initiation.
The chemical composition and antimicrobial activity of the essential oils isolated from twigs with needles (T+N) and from twigs without needles (T-N) from wild Pinus peuce Griseb. (Pinaceae), from ...three different locations in R. Macedonia, were investigated. Essential oil yields of T+N ranged from 7.5 mL/kg to 12.5 mL/kg and for T-N from 13.8 mL/kg to 17.3 mL/kg. GC/FID/MS analysis of the essential oils revealed eighty-four components, representing 93.7–95.7% and 91.2–92.0% of the T+N and T-N oils, respectively. The major components in T+N and T-N oils were monoterpenes: α-pinene (23.8–39.9%, 21.2–23.3%), camphene (2.2–5.5%, 0.7–2.0%), β-pinene (10.1–17.1%, 8.2–16.4%), myrcene (1.2–1.41%, 1.6–2.5%), limonene+β-phellandrene (6.8–14.0%, 8.8–23.6%) and bornyl acetate (2.3–6.9%, 1.1–3.4%), followed by the sesquiterpenes: trans-(E)-caryophyllene (3.6–4.3%, 3.2–7.3%), germacrene D (7.1–9.5%, 5.0–10.3%) and δ-cadinene (2.1–3.1%, 3.3–4.2%, respectively). Antimicrobial screening of the essential oils was made by disk diffusion and broth dilution methods against 13 bacterial isolates of Gram-positive and Gram-negative bacteria and one strain of Candida albicans. T-N essential oils showed antimicrobial activity toward Streptococcus pneumoniae, Staphylococcus aureus, S. epidermidis and Candida albicans as well as Streptococcus agalactiae, Acinetobacter spp. and Haemophilus influenzae. The antimicrobial activity of T+N essential oils was greater, especially against Streptococcus agalactiae, S. pyogenes, Enterococcus and Candida albicans, followed by Haemophilus influenzae, Acinetobacter spp., Escherichia coli, Salmonella enteritidis, Staphylococcus aureus and S. epidermidis. Minimal inhibitory concentrations (MICs) of all tested essential oils ranged from 15–125 μL/mL. Summarizing the obtained results, the antimicrobial activity of Pinus peuce T+N and T-N essential oils collected from different localities in R. Macedonia varied considerably. These alterations in the antimicrobial activity can be attributed to the differences in the quantitative composition and percentage amounts of the components present in the respective essential oils, although it was evident that there were no differences in the qualitative composition of the essential oils, regardless of the locality of collection, or the type of plant material (T+N or T-N).
To gain insight into the question of which vegetation characteristics have the most influence on avian assemblages in late-successional forests, the habitat preferences of bird-guilds in old-growth ...endemic forests of Macedonian pine were studied over 3 years in the Pirin National Park, Bulgaria. Bird–habitat relationships were investigated by comparing vegetation characteristics, and bird species richness, diversity, abundance, and guild structure of birds (determined according to food type, foraging and nesting sites) between mature (60–100 years old) and over-mature (>120 years old) Macedonian pine forest stands. Studied forest age-classes differed mainly by the density, height and diameter of trees, and the amount of dead wood. The first one of these parameters decreased and the latter two parameters increased with the forest succession. The difference in the vegetation structure affected the abundance of bird-guilds and thus, the overall bird abundance and the structure of avian assemblages within Macedonian pine forests. There was no significant difference in bird diversity among studied forest age-classes, but the overall bird abundance increased with forest maturation. Analyzed by study plots, species richness was higher in over-mature forests, but at cluster level, there was no significant difference between mature and over-mature forest age-classes. Half of the studied (insectivorous, hole- and ground-nesters, bark- and canopy-foraging bird species) guilds were more abundant in over-mature forests, while there was no bird-guild exhibiting a preference for mature forest stands. The abundances of bird-guilds were correlated with tree height, diameter at breast height and the amount of dead wood between the studied forest age-classes and this might explain their preferences for over-mature pine forests. Therefore, for future sustainable management of these endemic forests and the conservation of their avifauna, efforts should focus on protecting the remaining native old-growth forest stands and the importance of the structure of Macedonian pine forests on their bird assemblages should be considered in forestry practices.
Terpenes are often used as ecological and chemotaxonomic markers of plant species, as well as for estimation of geographic variability. Essential oils of relic and Balkan endemic/subendemic conifers, ...Picea omorika, Pinus heldreichii, and P. peuce, in central part of Balkan Peninsula (Serbia and Montenegro), on the level of terpene classes and common terpene compounds were investigated. In finding terpene combinations, which could show the best diversity between species and their natural populations, several statistical methods were applied. Apart from the content of different terpene classes (P. omorika has the most abundant O‐containing monoterpenes and sesquiterpenes; P. heldreichii and P. peuce have the largest abundance of sesquiterpene and monoterpene hydrocarbons, resp.), the species are clearly separated according to terpene profile with 22 common compounds. But, divergences in their populations were established only in combination of several compounds (specific for each species), and they were found to be the results of geomorphologic, climatic, and genetic factors. We found similarities between investigated species and some taxa from literature with respect to terpene composition, possibly due to hybridization and phylogenetic relations. Obtained results are also important regarding to chemotaxonomy, biogeography, phylogeny, and evolution of these taxa.
