A layered organization of cells is a common architectural feature of many neuronal formations. Mutations of the zebrafish gene nagie oko (nok) produce a severe disruption of retinal architecture, ...indicating a key role for this locus in neuronal patterning. We show that nok encodes a membrane-associated guanylate kinase-family scaffolding protein. Nok localizes to the vicinity of junctional complexes in retinal neuroepithelium and in the photoreceptor cell layer. Mosaic analysis indicates that the nok retinal patterning phenotype is not cell-autonomous. We propose that nok function in patterning of postmitotic neurons is mediated through neuroepithelial cells and is necessary for guiding neurons to their proper destinations in retinal laminae.
In the past, craftspeople contributed to a great extent to the economic development of Bjelovar and the towns in the today small, but once rather large county, which included wider areas of Bjelovar ...and Križevci. Since Bjelovar was the centre of the Varaždin Military Border, which first started with the demilitarisation in 1871, Bjelovar and its accompanying towns of Koprivnica and Križevci, as well as Daruvar at the turn of the century, benefited the most from economic development. Craftspeople were through their multifold activities a part of this development in the period 1871–1941. Limited to family business, livelihood from crafts was very insecure and of brief duration. Without joining in an association, they could not have survived at all, since as of the rule of Khuen Héderváry, the attitude towards craftpeople was rather inferior in comparison with industry and tradespeople. In the period 1884–1931, Croatian craftspeople had to operate pursuant to the Crafts Act, which was infavourable to them. Subsequently, as of 1932, pursuant to the Trades Act of that year, they even had to change the name of their profession to tradespeople. They were joined within the Chamber of Trades in Zagreb and Osijek, or rather within Yugoslav Chamber of Trades. Though craftspeople from Bjelovar, Daruvar, Grubišno Polje, Garešnica and Čazma shared a similar history and opened their shops to meet the needs of the local population, it was only the town of Bjelovar that as early as in 1891 had craftspeople of nearly all professions. It thereby met the needs of other smaller fair centres, municipalities, and towns as well, except for Koprivnica and Križevci, which were included in the county until 1918, but developed parallel with Bjelovar thanks to their position on the railway line. In early capitalism, craftspeople succeeded the guilds and took over their role in between trade and industry. However, in developed monopolistic capitalism, they became a surplus, as standardised goods produced in factories were sold by wholesale. Craftspeople thus became a surplus that only educated workpower for shops and factories. The ruin of craftspeople had been cut to the very core of the regulations and dependent on executive orders of the regime.
Although microtubule-dependent motors are known to play many essential functions in eukaryotic cells, their role in the context of the developing vertebrate embryo is less well understood. Here we ...show that the zebrafish ale oko (ako) locus encodes the p50 component of the dynactin complex. Loss of ako function results in a degeneration of photoreceptors and mechanosensory hair cells. Additionally, mutant Müller cells lose apical processes and their perikarya translocate rapidly towards the vitreal surface of the retina. This is accompanied by the accumulation of the apical determinants Nok and Has/aPKC in their cell bodies. ako is required cell-autonomously for the maintenance of the apical process but not for cell body positioning in Müller glia. At later stages, the retinotectal projection also degenerates in ako mutants. These results indicate that the p50 component of the dynactin complex is essential for the survival of sensory neurons and the maintenance of ganglion cell axons, and functions as a major determinant of apicobasal polarity in retinal radial glia.
The zebrafish MAGUK protein Nagie oko is a member of the evolutionarily conserved Crumbs protein complex and functions as a scaffolding protein involved in the stabilization of multi-protein ...assemblies at the tight junction. During zebrafish embryogenesis, mutations in nagie oko cause defects in both epithelial polarity and cardiac morphogenesis. We used deletion constructs of Nagie oko in functional rescue experiments to define domains essential for cell polarity, maintenance of epithelial integrity and cardiac morphogenesis. Inability of Nagie oko to interact with Crumbs proteins upon deletion of the PDZ domain recreates all aspects of the nagie oko mutant phenotype. Consistent with this observation, apical localization of Nagie oko within the myocardium and neural tube is dependent on Oko meduzy/Crumbs2a. Disruption of direct interactions with Patj or Lin-7, two other members of the Crumbs protein complex, via the bipartite L27 domains produces only partial nagie oko mutant phenotypes and does not impair correct junctional localization of the truncated Nagie oko deletion protein within myocardial cells. Similarly, loss of the evolutionarily conserved region 1 domain, which mediates binding to Par6, causes only a subset of the nagie oko mutant epithelial phenotypes. Finally, deletion of the C-terminus, including the entire guanylate kinase and the SH3 domains, renders the truncated Nagie oko protein inactive and recreates all features of the nagie oko mutant phenotype when tested in functional complementation assays. Our observations reveal a previously unknown diversity of alternative multi-protein assembly compositions of the Crumbs-Nagie-oko and Par6-aPKC protein complexes that are highly dependent on the developmental context.
Life cycle design is understood as 'to develop' (to plan, to calculate, to define, to draw) a holistic concept for the entire life cycle of a product'. Life cycle design means a one time planning ...during the concept phase of a product in which the pathway of a product over the entire life cycle is determined. So e.g. the planning of possible services for a product during its utilization phase, the way of material recycling, how and which parts can be reused, how the logistics for recycling will be organised or how the product can be used afterwards. So it is a conceptual pre-design of all later activities over the life cycle. By this understanding the book delivers a really holistic approach because before a product is physically made a life-long concepts and utilization scenarios with closed material and information cycles have to be developed. This promotes a real 'thinking in product (life) cycles'. The book addresses professionals as well as researchers and students in the field of product life cycle management. Different methods in the field of product design, operation and recycling will be presented and finally merge to an integrated method of product life cycle design. Readers will benefit from the holistic approach which enables them to design successful products by the implementation of closed loop product life cycles.
Deformed conglomeratic clasts exposed along the Neoproterozoic Nakasib Suture and the Oko Shear Zone are used to calculate three-dimensional (3D) tectonic strain associated with the latter to ...quantify strain associated with post-accretionary deformational belts in the Arabian–Nubian Shield. The Nakasib Suture is a NE-trending fold and thrust belt that is sinistrally offset (∼10
km) by the cross-cutting NNW- to NW-trending strike-slip faults of the Oko Shear Zone. The Nakasib Suture was formed as a result of collision between the Haya terrane and the Gebeit terrane at ∼750
Ma ago. The Oko Shear Zone was subsequently formed as a result of an E–W directed shortening of the Arabian–Nubian Shield due to collision between East and West Gondwana at ∼670–610
Ma ago. This analysis indicates the following: (1) The Nakasib Suture is dominated by flattening strain with the flattening plane of the associated strain ellipsoid oriented at 21°/77°SE. This flattening deformation is interpreted to be associated with nappe emplacement from north to south. (2) Some regions along the Nakasib Suture are characterized by constriction strain that might be due to refolding of the early nappes about NE-trending axes. (3) The Oko Shear Zone is characterized by constriction strain, with the
XY plane of the strain ellipsoid oriented at 171°/68°E. The strain ellipsoid associated with the Oko Shear Zone manifests superimposition of E–W shortening on the NE-trending fold and thrust belt associated with the Nakasib Suture. (4) The tectonic strain of the Oko Shear Zone, superimposed over the structures of the Nakasib Suture, is characterized by a strain ellipsoid whose flattening plane is oriented at 21°/49°W. The strain ellipsoid of the tectonic strain has a major axis with a quadratic elongation of 3.6 and an orientation of 357°/25°, an intermediate axis with a quadratic elongation of 1.2 and an orientation of 231°/30°, and a minor axis with a quadratic elongation of 0.25 and an orientation of 115°/18°. This suggests that the post-accretionary deformation of the Arabian–Nubian Shield was superimposed as a NW–SE directed shortening that created early N–S shortening zones and late NW-trending sinistral strike-slip faults.
The differentiation of distinct cell populations in the retina is a multi-step process that involves cell cycle exit, migration, and dramatic changes of cell morphology. All these steps are tightly ...controlled by multiple regulatory pathways, which involve both cell-autonomous networks of transcription factors and cell–cell signaling events. Additional regulatory inputs into cell fate decisions have been recently suggested: accumulating evidence shows that the timing of cell cycle exit, the orientation of the mitotic spindle during the last cell division, and the polarity of neuronal progenitor cells could play important roles in cell fate determination.
