Microbial remediation is a viable and eco-friendly approach for decontaminating pollution. However, its effectiveness can be limited by the microorganisms’ survival and growth in changing ...environments. Hydrochar materials have been utilized in this study to increase the growth and atrazine degradation capabilities of Paenarthrobacter sp. KN0901, a strain capable of atrazine biodegradation. Acid-modified hydrochars exhibited a higher carbonation rate, specific surface area, and number of defect sites compared to raw hydrochar. Following three days of incubation at 15 °C, the atrazine degradation rate increased from 90.7 % to 98.2 % when utilizing H3PO4-modified hydrochar (PHC). Additionally, the addition of PHC resulted in an increase in both bacterial concentration and cell viability of strain KN0901, by 1.6 and 1.4 times, respectively. Under various conditions, including temperatures of 4 ºC and 35 ºC, as well as pH levels of 5 and 9, and dd·H2O media, PHC exhibited a significant enhancement in atrazine degradation and cell viability of strain KN0901. Furthermore, PHC demonstrated the ability to sustain high proliferation and viability of strain KN0901 over five cycles, indicating its remarkable stability and biocompatibility. This study offers a new perspective on the development and application of bioremediation approaches in restoring atrazine-polluted environments, even under challenging conditions.

Cancer drug development has been riddled with high attrition rates, in part, due to poor reproducibility of preclinical models for drug discovery. Poor experimental design and lack of scientific ...transparency may cause experimental biases that in turn affect data quality, robustness and reproducibility. Here, we pinpoint sources of experimental variability in conventional 2D cell-based cancer drug screens to determine the effect of confounders on cell viability for MCF7 and HCC38 breast cancer cell lines treated with platinum agents (cisplatin and carboplatin) and a proteasome inhibitor (bortezomib). Variance component analysis demonstrated that variations in cell viability were primarily associated with the choice of pharmaceutical drug and cell line, and less likely to be due to the type of growth medium or assay incubation time. Furthermore, careful consideration should be given to different methods of storing diluted pharmaceutical drugs and use of DMSO controls due to the potential risk of evaporation and the subsequent effect on dose-response curves. Optimization of experimental parameters not only improved data quality substantially but also resulted in reproducible results for bortezomib- and cisplatin-treated HCC38, MCF7, MCF-10A, and MDA-MB-436 cells. Taken together, these findings indicate that replicability (the same analyst re-performs the same experiment multiple times) and reproducibility (different analysts perform the same experiment using different experimental conditions) for cell-based drug screens can be improved by identifying potential confounders and subsequent optimization of experimental parameters for each cell line.

A viable cell count is essential to evaluate the kinetics of cell growth. Since the hemocytometer was first used for counting blood cells, several variants of the methodology have ...been developed towards reducing the time of analysis and improving accuracy through automation of both sample preparation and counting. The successful implementation of automated techniques relies in the adjustment of cell staining, image display parameters and cell morphology to obtain equivalent precision, accuracy and linearity with respect to the hemocytometer. In this study we conducted the validation of three trypan blue exclusion-based methods: manual, semi-automated, and fully automated; which were used for the estimation of density and viability of cells employed for the biosynthesis and bioassays of recombinant proteins. Our results showed that the evaluated attributes remained within the same range for the automated methods with respect to the manual, providing an efficient alternative for analyzing a huge number of samples.

Cancer is a group of diseases in which cells divide continuously and excessively. Cell division is tightly regulated by multiple evolutionarily conserved cell cycle control mechanisms, to ensure the ...production of two genetically identical cells. Cell cycle checkpoints operate as DNA surveillance mechanisms that prevent the accumulation and propagation of genetic errors during cell division. Checkpoints can delay cell cycle progression or, in response to irreparable DNA damage, induce cell cycle exit or cell death. Cancer-associated mutations that perturb cell cycle control allow continuous cell division chiefly by compromising the ability of cells to exit the cell cycle. Continuous rounds of division, however, create increased reliance on other cell cycle control mechanisms to prevent catastrophic levels of damage and maintain cell viability. New detailed insights into cell cycle control mechanisms and their role in cancer reveal how these dependencies can be best exploited in cancer treatment.
The aim of our study was to investigate the effects of cyanobacterial metabolites: microcystin-LR (MC-LR) anabaenopeptin-A (ANA-A), cylindrospermopsin (CYL), their binary and ternary mixtures on ...rainbow trout (Oncorhynchus mykiss) gill (RTgill-W1) cell line. We determined the following cell parameters: Hoechst and propidium iodide (PI) double staining, intracellular ATP level with luminometric assay, glutathione level with ThiolTracker Violet®- glutathione detection reagent and cytoskeletal F-actin fluorescence. The results showed that although reduction of Hoechst fluorescence was observed in both binary and ternary combinations of cyanobacterial metabolites, the mixture of MC-LR + ANA-A + CYL was the most potent inhibitor (EC50 = 148 nM). PI fluorescence and ATP levels were more increased in the cells exposed to the mixtures than those exposed to the individual metabolites with synergistic toxic changes suggesting apoptosis as the mechanism of cell death. Reduced glutathione level was also decreased in cells exposed both to single metabolites and their mixtures with the highest decrease and synergistic effects at 334 nM MC-LR+334 nM ANA-A+ 334 nM CYL suggesting induction oxidative stress by the tested compounds. Reduction of F-actin fluorescence was found in the cells from all of the groups exposed to individual metabolites and their mixtures, however the highest level of inhibition showed the binary MC-LR + CYL and the ternary MC-LR + ANA-A + CYL with synergistic interactions. The study suggests that in natural conditions fish gill cells may be very sensitive to individual cyanobacterial metabolites and more prone to their binary and ternary mixtures.

As a kind of newly emerging pollutant, nanoplastics are easily to be ingested by organisms, and cause severe damage to biological functions because of their small size, high specific surface area, ...and strong biological penetration. Recently, there are increasing reports of numerous airborne microplastics, including polystyrene (PS), being detected in atmospheric samples, which implies a potential risk to the human respiratory system. In this work, we evaluated the effects of polystyrene nanoparticles of two different sizes (PS-NP25: 25 nm diameter and PS-NP70: 70 nm diameter) on the human alveolar epithelial A549 cell line including internalization, cell viability, cell cycle, apoptosis, and associated gene transcription and protein expression. Results showed that PS-NP25 was internalized more rapidly and efficiently into the cytoplasm of A549 than PS-NP70. PS-NPs significantly affected the cell viability, caused cell cycle S phrase arrest, activated inflammatory gene transcription, and changed the expression of proteins associated with cell cycle and pro-apoptosis. PS-NPs induced significant up-regulation of pro-inflammatory cytokines such as IL-8, NF-κB, and TNF-α, as well as pro-apoptotic proteins such as DR5, caspase-3, caspase-8, caspase-9, and cytochrome c, which revealed that PS-NPs triggered a TNF-α-associated apoptosis pathway. This study suggests that exposure duration, diameter, and concentration are the key factors for evaluating the toxicological effects of PS-NPs on alveolar epithelial cells. More attention must be focused on the risk of nanoplastic-related air pollution and the environmental toxicological effects of nanoplastics on humans and other terrestrial mammals.

Reaction between the polymeric RuCl2(CO)2n and the N,N-bidentate ligand, 8-amino-quinoline (Quin), in methanol, afforded the photoactivated CO releasing molecule with the formula of trans-(Cl,Cl)-RuCl2(CO)2Quin. In the presence of biomolecules or in solvents with varying polarity and coordinating abilities, the solvatochromic characteristics and dark stability were investigated. A new board band emerged in the visible spectrum during the illumination, and its position varies according to the type of solvent used, indicating the role of the solvent in controlling the nature of the CO-depleted species. Spectral methods were used in combination with density functional theory simulations to get insight into the local minimum structure and the electronic properties of the Ru(II) complex. The results of the myoglobin assay showed that within the first two hours of illumination, one of the two CO molecules was released. The cytotoxic properties of the Ru(II)-based complex were investigated against normal mice bone marrow stromal cells and malignant human acute monocytic leukaemia cells.
