Due to its environmental persistence and bioaccumulative properties, mercury is considered highly toxic to humans, ecosystems, and wildlife. The present study aimed to investigate the total mercury ...(Hg) concentrations in hair samples of 40 wild cats belonging to eight different species collected from various provinces of Iran and to characterize their risk of Hg exposure. Total Hg levels in Iranian wild cats ranged from 62 to 3670 ng/g dw hair, with a median value of 488 ng/g dw hair. The lowest median Hg concentration was found in west Iran (251 ng/g dw hair), while the highest median level was measured in the north-east of the country (736 ng/g dw hair), likely related to its past use in agriculture and potential contamination of industrial effluents. The overall contamination of Iranian wild cats with mercury can be considered generally low, but 20% of the samples reached levels above 1100 ng/g dw hair, set as indicative of an environmental Hg concern according to the U.S. Fish and Wildlife Service. Our data are comparable with other investigations on wildlife worldwide, confirming the suitability of museum collection specimens to assess the environmental levels of Hg. We showed that, while diet and habitat can influence the mercury accumulation in wild cats, complementary factors, such as age, size, and interspecific metabolic differences, should be considered as well in future investigations.
Heavy metals in coastal waters are a great environmental concern in the North Sea since the middle of the 20th century. Regulatory efforts have led to a significant reduction in atmospheric and ...water-transported heavy metals. Still, high concentrations of these in sediments remain a risk for ecosystems, requiring close monitoring. Here, we investigated the applicability of Nucella lapillus museum collections as a tool for targeted tracking of chronic anthropogenic heavy metal pollution. We analysed the concentration ratios of the common heavy metals Cu, Cd, Pb, and Zn in relation to Ca in N. lapillus shells collected from the Dutch and Belgian intertidal zone over the last 130 years. We found that shell Cu/Ca and Zn/Ca concentration ratios remained remarkably constant, whereas Pb/Ca concentration trends were closely aligned with emissions of leaded petrol in Europe. Our results suggest that N. lapillus provides a suitable Pb pollution archive of the intertidal zone.

Amber, a natural polymer, is fossil tree resin derived from diverse botanical sources with varying chemical compositions. As such, all amber is susceptible to the effects of light, temperature, ...relative humidity, and oxygen, as well as exposure to certain chemicals, and will deteriorate over time in collections if left unprotected. Here we review approaches for the conservation, preparation, and imaging of amber specimens and their inclusions, and address indications and causes of amber degradation, as well as recommendations for a suitable storage environment. We also provide updated preparation and embedding protocols, discuss several techniques for imaging inclusions, and address digitization efforts. A stable storage environment is essential to mitigate or avoid deterioration of amber, which often manifests as crazing, spalling, breaking and colour changes. Based on previous conservation studies of fossil resins, we generally recommend storage in a climate-monitored environment with a relative humidity of ca. 50%, 18 °C, and stress that light exposure must be kept to a minimum. For stabilization and anoxic sealing, amber specimens should ideally be embedded in an artificial epoxide resin (EpoTek 301-2 or similar is currently recommended). Amber should not be treated with or stored in vegetable or mineral oils (even for a short time for examination or photography), or come into contact with alcohol, disinfecting agents, hydrogen peroxide, or other destructive solvents or mixtures, since any of these materials can irreversibly damage the amber. Most photography of inclusions for research and digitization purposes can be successfully accomplished using light microscopy. Scanning electron microscopy (SEM) is sometimes used to uncover fine details, but is an invasive method. However, X-ray based methods (utilizing micro computed tomography, or micro-CT) are becoming more frequently used and increasingly indispensable in the examination of minute internal structures of inclusions, and to fully visualize important structures in opaque amber. Micro-CT makes it possible to digitize an inclusion three-dimensionally, and thus enables digital specimen ‘loans’. Light microscopal images are still widely used in the digitization of amber specimens and are an essential alternative to micro-CT imaging when resources or time are limited. Overall, due to the vulnerability of all fossil resins, we recommend that conservation of amber samples and their inclusions be prioritized.
Natural history museum collections provide unique resources for understanding how species respond to environmental change, including the abrupt, anthropogenic climate change of the past century. ...Ideally, researchers would conduct genome‐scale screening of museum specimens to explore the evolutionary consequences of environmental changes, but to date such analyses have been severely limited by the numerous challenges of working with the highly degraded DNA typical of historic samples. Here, we circumvent these challenges by using custom, multiplexed, exon capture to enrich and sequence ~11 000 exons (~4 Mb) from early 20th‐century museum skins. We used this approach to test for changes in genomic diversity accompanying a climate‐related range retraction in the alpine chipmunks (Tamias alpinus) in the high Sierra Nevada area of California, USA. We developed robust bioinformatic pipelines that rigorously detect and filter out base misincorporations in DNA derived from skins, most of which likely resulted from postmortem damage. Furthermore, to accommodate genotyping uncertainties associated with low‐medium coverage data, we applied a recently developed probabilistic method to call single‐nucleotide polymorphisms and estimate allele frequencies and the joint site frequency spectrum. Our results show increased genetic subdivision following range retraction, but no change in overall genetic diversity at either nonsynonymous or synonymous sites. This case study showcases the advantages of integrating emerging genomic and statistical tools in museum collection‐based population genomic applications. Such technical advances greatly enhance the value of museum collections, even where a pre‐existing reference is lacking and points to a broad range of potential applications in evolutionary and conservation biology.
Heritage institutions face major changes in the areas of digitisation, narrative, inclusivity, and participation.
