Depression is one of the most common psychiatric diseases and the prevalence of depressive symptoms in women is almost twice compared to men, although the reasons of this gender difference are not fully understood yet. Recently, soluble amyloid beta (Aβ)1-42 peptide has been receiving great importance in the development of depression, also considering that depression is highly comorbid with Alzheimer’s disease and other neurodegenerative illnesses. The central role played by Aβ in the development of depressive-like symptoms in rodents has been evidenced in environmental rodent model of depression. Indeed, we have previously found that lifelong exposure to n-3 polyunsaturated fatty acids (PUFA) deficient diet in female rats at 8 weeks of life leads to depressive like- symptoms and higher susceptibility to stress associated with increased Aβ levels. In order to understand if such effects were maintained over time, rats were exposed to the same diet regimen until 6 or 21 weeks of life. We found that both timepoints of exposure to n-3 PUFA deficient diet lead to depressive-like phenotype. Furthermore, a significant alteration in brain neurochemistry was retrieved. In particular, in hippocampal area a significant reduction in serotonin (5-HT) and noradrenaline (NA) content was evidenced. Considering the prominent role of NA in counterbalancing neuroinflammatory state, we quantified in the same brain area kynurenine levels, a metabolite of tryptophan implicated in inflammatory state and brought to the fore for its implication in depression. Interestingly, kynurenine levels were significantly increased in hippocampus (HIPP) of female rats exposed to such diet. In addition, lifelong deficiency in n-3 PUFA dietary intake led to systemic increase of corticosterone, hence hypothalamic pituitary adrenal (HPA) axis hyperactivation, and higher proinflammatory cytokine production. Increased production of kynurenine, along with HPA axis hyperactivation, have been associated with immune system modulation, particularly through Toll-like receptor type 2 (TLR2) and Toll-like receptor type 4 (TLR4) involvement. In addition, it has been shown that soluble forms of Aβ1-42 can induced depressive like-phenotype in consequence to a crosstalk between TLR4 and 5-HTergic system. Thus, considering that in this model we have previously reported increased plasma Aβ1-42 level, we quantified TRL2 and 4 expression in HIPP of treated rats. We found that chronic exposure to a diet characterized by very low n-3 PUFA content led to higher expression of TLR2 and TLR4 in HIPP of female treated rats, indicating an activation of the immune system and was accompanied by increased expression of oligomeric Aβ.
Taken together, our data indicate that the pro-depressive effects induced by a diet poor in n-3 PUFA can be attributable to a shift of hippocampal tryptophan metabolism toward inflammatory metabolite ultimately corresponding to altered immune response and increased Aβ oligomerization.
De nombreuses pathologies psychiatriques, telles que la schizophrénie, les troubles bipolaires ou la dépression majeure, bien que très différentes, ont en commun une dysfonction du système de ...récompense et de motivation en lien avec une altération de la transmission dopaminergique. Par ailleurs, ces pathologies s’accompagnent de modifications du métabolisme lipidique et en particulier d’une diminution des taux en acide docosahexaenoic (DHA), le principal acide gras polyinsaturé (AGPI) n-3 dans le système nerveux central. Cependant, bien que certaines études cliniques décrivent des effets bénéfiques de supplémentations en AGPI n-3 sur certains symptômes psychiatriques, ces résultats restent controversés, et l’implication de la modification du statut lipidique dans l’étiologie de ces pathologies reste très peu étudiée. Le but de ce travail a été d’établir s’il existe un lien causal entre une déficience en AGPI n-3 et certains endophénotypes neurobiologiques et comportementaux caractéristiques de pathologies psychiatriques. En particulier, nous avons fait l’hypothèse que la déficience en AGPI n-3 pourrait conduire à un dysfonctionnement de la transmission dopaminergique mésolimbique. Dans cette étude, des tâches de conditionnement opérant chez la souris nous ont permis de mettre en évidence un déficit motivationnel à l’âge adulte induit par une déficience développementale en AGPI n-3 qui est partiellement prévenu par une supplémentation en AGPI n-3 à la naissance, mais pas au sevrage. Ce déficit motivationnel s’accompagne d’une altération de la transmission dopaminergique comme le suggère la réduction de la sensibilité au psychostimulant, l’amphétamine. Plus précisément, nous avons pu montrer que la déficience en AGPI n-3 conduit à une dysfonction des propriétés électrophysiologiques des neurones épineux moyens (medium spinyneurons ou MSN) dans le noyau accumbens (NAc), population neuronale centrale pour la modulation de la motivation. En effet la carence en AGPI n-3 induit une réduction de l’excitabilité des MSNs de la voie directe (dMSNs) qui expriment le récepteur dopaminergique de type D1, associée à une augmentation de la transmission inhibitrice reçue par ces neurones. Ces modifications sont restaurées par l’application de l’agoniste des récepteurs dopaminergiques D2 (RD2), le quinpirole. Ces données nous ont amené à faire l’hypothèse que la diminution d’excitabilité des dMSNs sous déficience en AGPI n-3 résulte de l’augmentation de la transmission GABA issue des MSNs de la voie indirecte (iMSNs) exprimant le RD2. En accord avec ces résultats, par utilisation d’un transgène Cre-dépendant permettant l’expression de l’acide gras désaturase FAT1, nous démontrons que la normalisation des taux d’AGPI dans les iMSN sélectivement, est suffisante pour restaurer les propriétés électrophysiologiques des dMSNs. Par ailleurs, l’expression de la FAT1 dans les neurones exprimant le D2R – mais pas ceux exprimant le D1R - est suffisante pour normaliser le déficit motivationnel en situation de carence. Nos résultats montrent donc un lien causal entre des modifications de taux d’AGPI dans une sous-population neuronale spécifique et une altération comportementale. Par ailleurs, notre étude suggère que la diminution des taux d’AGPI décrite dans plusieurs pathologies psychiatriques pourrait directement participer à l’expression de certains symptômes tels que l’avolition ou l’apathie.
Various, though distinct psychiatric disorders, such as Schizophrenia, bipolar disorder or major depression are associated with a dysfunction of the reward system linked to an alteration of dopamine transmission. Furthermore, these pathologies are also accompanied by changes in lipid metabolism and in particular a decrease in the brain content of docosahexaenoic acid (DHA), the main n-3 polyunsaturated fatty acid (PUFA) in the nervous system. However, despite that n-3 PUFA supplementation seems to improve or prevent some psychiatric symptoms, these results are still controversial and the implication of brain lipid composition in the etiology of psychiatric endophenotypes has been overlooked. The aim of this study was to investigate a potential causal link between n-3 PUFA deficiency and common neurobiological and behavioral endophenotypes of psychiatric disorders. In particular, the hypothesis was that n-3 PUFA deficiency could lead to dysfunctions of mesolimbic dopamine transmission and associated behaviors. Using operant conditioning tasks in mice, we showed that developmental n-3 PUFA deficiency leads to a motivational deficit at adulthood, that is partially reversed by n-3 PUFA supplementation starting at birth, but not at weaning. This motivational deficit was associated with an alteration of dopaminergic transmission as revealed by the reduced sensitivity to the psychostimulant amphetamine. More precisely, we showed that n-3 PUFA deficiency leads to alterations in electrophysiological properties of medium spiny neurons (MSNs) in the nucleus accumbens (NAc), main actors in motivational processes. Indeed, MSNs from the direct pathway (dMSNs), that express dopaminergic D1 receptors, displayed a decrease in neuronal excitability in parallel with an increase of inhibitory input onto these neurons. These alterations were reversed by the dopaminergic D2 receptor (D2R) agonist quinpirole. These data led us to hypothesize that the decreased dMSN excitability induced by n-3 PUFA deficiency could result from an increase of the inhibitory input of MSNs from the indirect pathway (iMSNs that expresses D2R), called lateral inhibition. Accordingly, using a transgenic approach that allows the expression of the fatty acid desaturase FAT1 in a credependent manner, we showed that rescuing appropriate PUFA levels in D2R-expressing neurons selectively (including iMSNs), was sufficient to reverse alterations in electrophysiological properties of MSNs induced by n-3 PUFA deficiency. Moreover, the selective expression of FAT1 in D2-expressing neurons – but not in D1-expressing neurons – reversed the motivational deficit observed in n-3 PUFA deficient mice. We demonstrated the existence of a causal link between modifications in PUFA levels in a discrete neuronal population and behavioral alterations. Overall, this study suggests that altered PUFA levels, observed in some psychiatric disorders, could directly participate in the development of symptoms such as avolition or apathy.
Un statut pauvre en Acides Gras Poly-Insaturés ω3 (AGPI ω3), favorisé par une alimentation occidentale comportant un faible ratio en ω3/ω6, semble contribuer au déclin cognitif chez les personnes ...âgées, mais les mécanismes cellulaires impactés sont encore mal connus. Nous avons donc étudié l’influence du statut en ω3 sur l’évolution de la neurotransmission glutamatergique et des fonctions astrocytaires au cours du vieillissement dans l’hippocampe de rats. Ces processus sont impliqués dans la formation de la mémoire et leurs dérégulations participent aux dommages cérébraux conduisant au déclin cognitif. Nous avons comparé 6 groupes de rats agés de 6 et 22 mois nourris avec un régime déficient en ω3, équilibré en ω3/ω6 ou supplémenté en ω3 (huile de poisson) : Jeunes équilibrés (JEq), déficients (JDef) ou supplémentés (JSup) et Agés équilibrés (AEq), déficients (ADef) ou supplémentés (ASup). Nous avons évalué l’efficacité synaptique et la plasticité (enregistrements électrophysiologiques), les fonctions astrocytaires (capture de glutamate et expression de la GFAP), les marqueurs neuronaux (transporteurs et récepteurs du glutamate), les capacités cognitives (Openfield et Labyrinthe de Barnes) et analysé la composition lipidique cérébrale. Les manipulations nutritionnelles d’apport en ω3 modifient efficacement l’incorporation de l’acide docosahexaénoïque (DHA, principale ω3 des membranes cellulaires) dans le cerveau (-50% deficient vs équilibré, +10% supplementé vs équilibré). Le vieillissement induit une diminution de 35% de l’efficacité synaptique en raison d’une baisse de la libération de glutamate pré-synatique, et une diminution de 30% de la capture de glutamate associé à une astrogliose conséquente (+100% GFAP). La déficience en ω3 acentue les effets du vieillissement (rats ADef vs AEq: -35% efficacité synaptique, -15% capture de glutamate, +30% GFAP). Al’inverse, la supplémentation en ω3 améliore l’efficacité synaptique (rats ASup vs AEq +25%) et semble inhiber l’astrogliose chez le rat âgé (ASup vs JEq : pas de modification de la GFAP). Les tests comportementaux montrent que le vieillissement a des effets plus marqués chez les déficients en ω3 et au contraire atténués chez les supplémentés. Nos résultats révèlent des altérations de la synapse glutamatergique de l’hippocampe au cours du vieillissement aggravées par la déficience en ω3 et atténuées par la supplémentation en ω3. Afin d’évaluer l’influence du statut en ω3 sur l’activation astrocytaire, des modèles in vitro d’astrocytes « âgés » et « activés » par des cytokines inflammatoires dont l’augmentation à bas bruit est caractéristique du vieillissement cérébral, ont été développés.
A poor ω3 polyunsaturated fatty acids (ω3 PUFA) status, favored by the low ω3/ω6 ratio in western diets, seems to contribute to cognitive decline in the elderly, but mechanistic evidence is lacking. We therefore explored the impact of ω3 status on the evolution of glutamatergic transmission and astrocytic functions in the hippocampus during ageing in rats. These processes are involved in memory formation and their dysregulation participates to the age-related brain damage leading to cognitive decline. We have compared 6 groups of rats aged 6 to 22 months fed ω3-deficient, ω3/ω6-balanced, or ω3 (fish oil) supplemented diets: Young ω3 Balanced (YB), Deficient (YD) or Supplemented (YS), and Old ω3 Balanced (OB), Deficient (OD) or Supplemented (OS) rats. We have evaluated synaptic efficacy and plasticity (electrophysiological recording), astroglial regulations (glutamate uptake and GFAP expression), neuronal markers (glutamate transporters and receptors), cognitive abilities (Barnes maze and Openfield) and analyzed brain fatty acids composition. Dietary modulation of ω3 intakes efficiently modified the incorporation of docosahexaenoic acid (DHA, the main ω3 in cell membranes) in brain (-50% deficient vs balanced, +10% supplemented vs balanced). Ageing induced a 35% reduction of synaptic efficacy due to decreased pre-synaptic glutamate release, and a 30% decrease in the astroglial glutamate uptake associated to a marked astrogliosis (+100% GFAP). ω3 deficiency further decreased these hallmarks of ageing (OD vs OB rats: -35% synaptic efficacy, -15% glutamate uptake, +30% GFAP). On the opposite, ω3 supplementation increased synaptic efficacy (+25% OS vs OD) and seems to abolish astrogliosis (OS vs YS : no change in GFAP). Behavioural tests showed some increased effects of age in deficient rats and attenuated effects in supplemented ones. Our results characterize some specific age-related alterations of the glutamatergic synapse in the hippocampus that are aggravated by a dietary deficit in ω3 and attenuated by ω3 supplementation. In order to explore ω3 status on astrocytic activation, in vitro models of “old” astrocytes and “activated” by inflammatory cytokines which characterize the low-grade inflammation in brain aging, have been developed.
Stabilità ossidativa e caratteristiche qualitative di carne di pollo arricchita con PUFA n-3 - 360 broiler, femmine ceppo Cobb 500, hanno ricevuto da 21 d fino alla macellazione (46 d) una dieta ...commerciale addizionata con 0, 0,5 o 1% dell'alga marina Schizochitrium sp. apportatrice di PUFA n-3. Le carni con un più alto contenuto di PUFA n-3 presentano una ridotta stabilità ossidativa, particolarmente evidente nel petto, mentre le proprietà organolettiche valutate nella coscia non risultano influenzate dal trattamento dietetico. Il colore del petto appare più intenso nel gruppo che ha ricevuto la più alta integrazione di alga.
Proprietà tecnologiche della carne di coniglio arricchita in PUFA n-3 - La ricerca ha avuto lo scopo di valutare alcune caratteristiche tecnologiche di prodotti trasformati (impasti di carne macinata ...e hamburger) provenienti da conigli alimentati con diete prive o contenenti l'8% di semi di lino. L'inclusione di semi di lino nella dieta ha determinato un significativo aumento della suscettibilità ossidativa (TBARS) degli impasti sia a 3 che a 6 mesi di conservazione a -20°C, mentre l'analisi sensoriale ha fatto rilevare la comparsa di sapore di rancido solo dopo 3 mesi di conservazione. Le variazioni di colore degli hamburger durante la conservazione a 2-4°C in atmosfera ordinaria o sottovuoto sono risultate comparabili nei due gruppi sperimentali.
Les acides gras polyinsaturés (AGPI) incorporés dans les phospholipides membranaires représentent une proportion importante (environ 20 %) de la matière sèche cérébrale. Ils sont apportés par ...l’alimentation sous forme de précurseurs (acide a-linolénique pour la série n-3, acide linoléique pour la série n-6) ou de dérivés à longues chaînes (notamment l’acide docosahexaénoïque, ou 22 :6n-3 et l’acide arachidonique, ou 20 :4n-6). Des données récentes indiquent que le statut en AGPI constitue l’un des facteurs environnementaux capables d’affecter le fonctionnement du système nerveux central (SNC). Les résultats principaux sont, d’une part, la mise en évidence d’anomalies comportementales chez des animaux recevant des régimes déséquilibrés en AGPI et, d’autre part, la détection de taux anormaux d’AGPI chez des humains souffrant de maladies du SNC. Au cours de ces dernières années, nous avons apporté des arguments établissant que ces dysfonctionnements peuvent être reliés à des altérations des processus de neurotransmission induites par le déséquilibre en AGPI.
It is well-established that the nutritional deficiency or inadequacy can impair immune functions. Growing evidence suggests that for certain nutrients increased intake above currently recommended ...levels may help optimize immune functions including improving defense function and thus resistance to infection, while maintaining tolerance. This review will examine the data representing the research on prominent intervention agents n-3 polyunsaturated fatty acids (PUFA), micronutrients (zinc, vitamins D and E), and functional foods including probiotics and tea components for their immunological effects, working mechanisms, and clinical relevance. Many of these nutritive and non-nutritive food components are related in their functions to maintain or improve immune function including inhibition of pro-inflammatory mediators, promotion of anti-inflammatory functions, modulation of cell-mediated immunity, alteration of antigen-presenting cell functions, and communication between the innate and adaptive immune systems. Both animal and human studies present promising findings suggesting a clinical benefit of vitamin D, n-3 PUFA, and green tea catechin EGCG in autoimmune and inflammatory disorders, and vitamin D, vitamin E, zinc, and probiotics in reduction of infection. However, many studies report divergent and discrepant results/conclusions due to various factors. Chief among them, and thus call for attention, includes more standardized trial designs, better characterized populations, greater consideration for the intervention doses used, and more meaningful outcome measurements chosen.
Le métabolisme énergétique cérébral via l’utilisation du glucose est fortement impliqué dans la production d’énergie nécessaire au fonctionnement du neurone en situation basale et d’activation. Des ...travaux précédents ont mis en évidence chez le rat chroniquement déficient en acides gras polyinsaturés (AGPI) de la série n-3 une altération de ce métabolisme en situation basale (diminution de l’utilisation cérébrale du glucose et de la densité des transporteurs de glucose GLUT1). Pour cerner les différentes étapes du métabolisme énergétique pouvant être modifiées par les AGPI n-3, l’expression des gènes clés a été mesurée par approche transcriptomique (cartes microfluidiques) chez l’animal déficient en AGPI n-3 ou supplémenté en acide docosahexaénoïque (DHA, 22 :6n-3). Ces mesures ont été réalisées sur deux zones cérébrales (cortex fronto-pariétal et couche CA1 de l’hippocampe) chez les animaux en situation basale et soumis à un environnement enrichi activant ces deux zones cérébrales. Pour ces 2 situations, le niveau d’utilisation cérébrale de glucose a été quantifié par la technique du fluoro-2-déoxyglucose couplée à l’imagerie de tomographie par émissions de positons (18FDG-TEP) chez les animaux déficients en AGPI n-3. L’analyse de la teneur cérébrale en AGPI membranaire a été réalisée par chromatographie en phase gazeuse et une approche in vitro sur culture primaire d’astrocytes a été développée pour apprécier l’impact du DHA sur les paramètres métaboliques de ces cellules.Les principaux résultats montrent que :- la déficience en n-3 diminue de 67% la teneur membranaire en DHA dans les deux zones cérébrales étudiées. Si la déficience induit principalement une diminution spécifique de l’expression de GLUT1 (-33%) dans le cortex fronto-pariétal en situation basale et d’activation, en revanche elle perturbe la neurotransmission glutamatergique dans l’hippocampe en augmentant l’expression des 2 transporteurs de glutamate (GLAST et GLT1). Par ailleurs, les données d’imagerie TEP mettent en évidence un hypométabolisme général du glucose chez les animaux déficients en n-3 en situation basale. Les données recueillies sur le modèle astrocytes soulignent un effet direct du DHA sur l’utilisation du glucose et l’expression de GLUT1 ;- La supplémentation en DHA ne modifie pas de façon appréciable la teneur membranaire en DHA dans les deux zones cérébrales étudiées. Au contraire de la déficience, il apparaît clairement pour la couche CA1 de l’hippocampe que l’expression de l’ensemble des gènes codant pour les complexes enzymatiques du cycle de krebs et de la voie de phosphorylation oxydative est significativement augmentée.Ces résultats originaux laissent ainsi entrevoir la possibilité que les acides gras de cette famille d’AGPI puissent intervenir sur l’énergétique et le fonctionnement de la synapse glutamatergique en modulant 1) le métabolisme glucidique (captage de glucose) et du glutamate en situation de déficit d’apport et 2) la production d’ATP (phosphorylation oxydative) en situation de supplémentation en DHA. L’altération de ces paramètres métaboliques au cours du vieillissement et dans certains désordres neurologiques, liée à un déficit de statut en DHA, mettent en avant les potentialités nutritionnelles des AGPI n-3 comme facteur préventif.
Cerebral energy metabolism via glucose utilization is heavily involved in the production of energy required to the neuron in basal conditions and activation. Previous work has shown in rats chronically deficient in n-3 polyunsaturated fatty acids (PUFA) altered the metabolism in basal condition (decrease of cerebral glucose use and density of glucose transporters GLUT1). To identify the different stages of energy metabolism may be modified by n-3 PUFA, the expression of key genes was measured by transcriptomic approach (Taqman Low Density Arrays) in animals deficient in n-3 PUFA or docosahexaenoic acid supplementation (DHA, 22:6n-3). These measurements were performed on two brain areas (fronto-parietal cortex and layer CA1 of the hippocampus) in animals in basal condition or submit to an enriched environment. For these two situations, the level of cerebral glucose utilization was quantified by the technique of fluoro-2-deoxyglucose imaging coupled with positron emission tomography (18FDG-PET) only in deficient n-3 PUFA animals. Analysis of brain PUFA content of membrane was performed by gas chromatography and an in vitro approach to primary culture of astrocytes was developed to assess the impact of DHA on metabolic parameters of these cells.The main results show that: - n-3 Deficiency decreases from 67% in membrane DHA content in both brain areas studied. If the deficiency induces mainly a decrease in the specific expression of GLUT1 (-33%) in the fronto-parietal cortex in basal and activation conditions, however it disrupts glutamatergic neurotransmission in the hippocampus by increasing the expression of two glutamate transporters (GLAST and GLT1). In addition, PET data show a general hypometabolism of glucose in animals deficient in n-3 in basal situation. Data collected on the model astrocytes point to a direct effect of DHA on glucose utilization and expression of GLUT1; - DHA supplementation does not alter significantly the membrane content of DHA in both brain areas studied. Unlike the n-3 deficiency, it is clear for the layer CA1 of the hippocampus that the expression of all genes encoding the enzyme complexes of the Krebs cycle and oxidative phosphorylation pathway is significantly increased. These original results suggest the possibility that the fatty acids of the n-3 PUFAs family can act on the energy and functioning of the glutamatergic synapse by modulating 1) glucose metabolism (glucose uptake) and glutamate in deficit intake situation and 2) the production of ATP (oxidative phosphorylation) in DHA supplementation. The alteration of these metabolic parameters during aging and certain neurological disorders, related to a deficit of DHA status, highlight the potential of dietary n-3 PUFA as a preventive factor.
Dietary intake of long-chain n-3 PUFA (n-3 PUFA), particularly EPA and DHA, has been associated with psychological well-being, but little is known about the n-3 PUFA intake of homeless youth. The ...current study determined the association between depression and anxiety symptoms and n-3 PUFA intake and erythrocytes status in homeless youth. Totally, 114 homeless youth aged 18–24 years were recruited from a drop-in centre. n-3 PUFA dietary intake was assessed using an FFQ, and erythrocytes status was determined by gas chromatography (GC). Linear regression models were used to determine the relationship between psychological well-being and n-3 PUFA intake and status. The mean intakes of EPA and DHA for all participants (0·06 ± 0·13 g/d and 0·11 ± 0·24 g/d) were well below recommended levels, and mean erythrocytes EPA + DHA (n-3 index) in the cohort (2·42 %) was lower than reported for healthy, housed adolescents and those with clinical depression. There was no association of n-3 PUFA intake and erythrocytes status with either depression or anxiety. However, the relationships of depression with dietary EPA (P = 0·017) and DHA (P = 0·008), as well as erythrocytes DHA (P = 0·007) and n 3-index (P = 0·009), were significantly moderated by sex even after adjusting for confounders. Specifically, among females, as the intake and status of these n-3 PUFA decreased, depression increased. Our findings show poor dietary intake and low erythrocytes status of n-3 PUFA among homeless youth, which is associated with depressive symptoms among females.
