    In this study we are presenting a new forensic identification method for humanidentification and for determining relatedness in the Slovenian population. This method is based on the analysis of 14 ... STR loci and the part of amelogenin gene used for gender identification. We are presenting some DNA extraction methods for isolation of genomic DNA from different biological samples, multiplex PCR (Polymerase Chain Reaction) amplification methods, capillary elecrophoresis method for separation of the amplification products, and the method used for automatic sizing and determining genotypes of the amplification products with 310 Genetic Analyzer (Perkin Elmer). We have analysed 250 urelated Slovenian individuals to determine genetic veriability of STR loci TH01, vWA, FESžFPS, F13A1, D21S11, FGA, D3S1358, TPOX, CSF1PO, D8S1179, D18S51, D5S818, D13S317, and D7S820 respectively. From the results obtained, we have estimated that all loci meet Hardy-Weinberg expectations. For all the 14 loci we have calculated statistical parameters of forensic interest (relative allele frequencies, observed and expected heterozygosity, polymorphism information content, probability of match and power of discrimination). The results obtained from comparison among allele distributions from Slovenian and other Caucasian population samples confirmed homogeneity of Caucasian population samples. For paternity disputes we have estiznated from the population data for all the 14 STR loci the combined paternity exclusion probability (0.9999974), and from data obtained in analyses of 130 paternity cases the practical paternity exclusion probability for separate loci. A practical application of the genetic identification method is shown with examples of confirmation and exclusion of paternity and one identification case. (Abstract truncated at 2000 characters.)
