    Synpatic communication is a fundamental function of neurons. Mechanisms underlying formation and function of synapses are therefore one of the mayor research fields in modern neurobiology. Due to its ... experimental accessibility,neuromuscular junction is considered a model synapse and virtually all of our current understanding of synaptogenesis derives from studies on this particular synapse. Synaptogenetic studies necessiate an experimental model enabling insight into all stages of this complex process. Especially suitable for such studies are therefore models in which neuromuscular junction formation proceeds under in vitro conditions. In the first part of study we introduced and characacterised the experimental model in which human muscle is innervated in vitro with rat spinal cord. Cell composition of this model was studied and the influence of several growth factors on the neuronal outgrowth under such conditions was tested. In the second part of our study we aimed to prepare homologous in vitro model by coculturing human myotubes with the explants of human spinal cords obtained atabortions. In the third part we studied the influence of nerve-derived factor agrin on the functional innervation in cocultures of human myotubes andrat motor neurons. Three hypotheses were testedČ 1. Nerve growth factors BDNF, GDNF, NT-3, NT-4 and CTNF faciliate neuronal outgrowth from embryonic rat spinal cord explants cocultured with human myotubes. 2. Human myotubes canbe innervated by human spinal cord neurons also under in vitro conditions. 3. Blocjking the effects of nerbe - derived factor agrin by heparin prevents functional innervation of human myotubes in human muscle - rat spinal cord cocultures.
