  • Tipizacija hantavirusa s pomoću lančane reakcije polimeraze

    Hemorrhagic Fever with Renal Syndrome (HFRS) is caused by viruses of genus Hantavirus. Various hantavirus isolates are classified in four groups or serotypes: Hantaan, Puumala, Seoul and Prospect ... Hill. It is important to define a hantavirus serotype due to a good relation between the patients humoral immune response, variety of clinical course and the prognosis of the disease. Aim of the study present searching of the possible detection and tipization of hantaviruses using new highly specific polymerase chain reaction (PCR). Two strategies were used for detection and tipization of hantaviruses. First, PCR was performed by genus specific primer pairs followed with restriction enzyme digestion of the PCR products. Second strategy was directed by using serotype specific primers. It was established that both strategies are suitable for classification of hantaviruses. Hantaviruses were successfully amplified and typed when isolated by classical method from material of infected rodents or from material of HFRS patients. Furthermore, PCR method with additional optimization is suitable for detection and tipization of hantaviruses directly from material of HFRS patients. Due to high specificity, sensitivity and fast performance, PCR is very applicable in study of epidemiology and pathogenesis of HFRS. Therefore PCR represent important addition in diagnostic repertoire of this disease.
