In this study we evaluated temoporfin-loaded PEGylated PLGA nanoparticles (NPs) as a new formulation for potential use in cancer treatment. Materials & Methods: NPs were characterized for their photophysical properties, temoporfin release, and cellular uptake and intracellular localization of temoporfin by using A549, MCF10AneoT andU937 cell lines. Dark- and photo-cytotoxicities were tested on A549 cells. Bioluminiscence and fluorescence imaging was performed on athymic nude-Foxn1 mice. Results: Compared to non-PEGylated PLGA NPs, the PEGylation significantly reduced the cellular uptake of NPs by all the selected cell lines, especially by the differentiated pro-monocytic U937 cells, revealing the stealth properties of the delivery system. Temoporfin delivered by NPs was localized in the perinuclear region, corresponding to the endoplasmic reticulum and Golgi apparatus, and in lysosomal-endosomal compartments. As shown by fluorescence quenching, temoporfin is highly aggregated within the NPs, and the release of monomers is faster from PEGylated PLGA NPs than from non-PEGylated ones. Dark cytotoxicity of temoporfin delivered by NPs was less than that of free temoporfin in standard solution (FoscanŽ). The photo-toxicity of internalized temoporfin after red light irradiation was, however, not reduced. Temoporfin delivered to mice by PEGylated PLGA NPs exhibits therapeutically favorable tissue distribution.
