    Background. Classical approach of preparation of biological specimens produces different artefacts in the final image of the cell. As an alternative, cryofixation and freeze substitution are taking ... places in laboratories for electron microscopy. The advantage of these methods is quick immobilization of samples, no chemical reactions that could alter the specimen and better preserved morphology and antigenicity. Methods. The aim of our work was to test and introduce the optimal combination of cryofixation and cryodehydration into our laboratory. We tested and compared three methods, Flat Embedding, Progressive Lowering of Temperature (PLT) and Cryofixation combined with Freeze Substitution. Results. The best preservation of ultrastructure was achieved with cryo methods. Flat embedding turned out to be inferior to cryomethods, while the PLT method produced poorly preserved cells. Conclusions. Our decision to introduce the combination of fixation in liquid propane and freeze substitution was based on the results we achieved with these methods.
