Collateral axon sprouting in the peripheral nervous system is a branching outgrowth of new axon terminals from noninjured axons into adjacent denervatedregions of a target tissue. Axonal sprouts ...regularly grow within theabandoned neurilemmal tubes from the Schwann cells pertaining to degraded axons. Collateral sprouts of nociceptive axons in the mammalian skin stop growing after a few weeks although there is more denervated skin available to them in their vicinity. We decided to check the hypothesis that this limitation might be due to inability of the axons sprouts to grow outside of their parent dermatomes. In order to explain this, we further assumed that Schwann cell tubes, pertaining to axons innervating a foreign dermatome, represent a poor growth substratum for axon sprouts from the parent dermatome.Experiments were performed on the hind limb and foot of the rat. Autonomous innervation region of a dermatome was determined by transection of an individual dorsal spinal root. Maximal innervation region was demonstrated by cutting one dorsal root above and one below the studied spinal nerve. The latter animals were thereafter monitored for 6 months for possible expansion of a dermatomal area because of axonal sprouting into adjacent denervated dermatomes. Maximal innervation area of the saphenous nerve in the foot and its expansion due to sprouting were studied after cutting and ligating the sciatic nerve. After two months, either the dorsal root L3 or L4 were severed to determine which area did the sprouts originating from those two segments occupy. Ability of sprouting axons to grow along the neurilemmal tubes of Schwann cells pertaining to axons from different spinal chord segments was investigated by making an end-to-side anastomosis of distal stumps of either peroneal or saphenous nerve to the sural nerve.
