  • Populacijska analiza Y kromosoma na območju Kočevja in Gorskega Kotarja = Population analysis of Y chromosome in Kočevje and Gorski Kotar region : diplomsko delo, univerzitetni študij

    The Kočevje and Gorski Kotar region has a very high prevalence of multiple sclerosis (MS) (190,5/100 000). Two intervals of imigration to Kočevje and Gorski Kotar region are documented from Saxony ... (Germany) where the prevalence of MS is also high (120/100 000). The aim of this study was to test the hypothesis that the population of Kočevje and Gorski Kotar is isolated and hasa higher genetic similarity to the population of Saxony compared to the general Croatian and Slovene population. We analysed 7 Y-chromosomal STR loci in Kočevje and Gorski Kotar region, Slovenia and Croatia. Gene diversities of each locus and haplotype diversities were calculated. The allelic frequency distribution, haplotypes and molecular variance were compared between the analysed populations and the Saxony population. Results indicate isolation andlow genetic diversity of the Kočevje and Gorski Kotar population. Haplotypecomparison and some of the YSTR loci analyses (DYS392) confirmed higher similarity of the population of Kočevje and Gorski Kotar to the population of Saxony. AMOVA analyses confirmed that most of the total genetic variability between populations is due to variability inside populations (>99 %) and not between them. Analysing isolated populations with low genetic variability and high prevalence of a specific disease is optimal for identifying possible causes of multifactorial diseases like MS. We have defined the first such population in Slovenia and Croatia which has great relevance in the identification of previously unknown possible genetic factorsin MS.
