  • Urokinazni plazminogenski aktivator in njegova inhibitorja PAI-1 in PAI-2 pri primarnem kožnem malignem melanomu : magistrska naloga

    Urokinase plasminogen activator and its inhibitors PAI-1 and PAI-2 are included in the degradation of extracellular matrix and formation of tumor stroma, which are condition for invasion and ... metastatic spread of tumor cells.Our study was aimed to find out the existence of eventual differences between the measured levels of urokinase plasminogen activator (uPA) and its inhibitors type1 and 2 (PAI-1ž2) in clinically suspected nevi, primary cutaneous malignant melanoma and healthy skin in the tumor surroundings. We also wanted to find out what is the correlation between levels of these factors in the tumors and healthy skin, and within subgroups formed on the basis of the clinical and histopatological prognostic factors of primary malignant melanoma. In our study 51 patients with clinical confirmed primary cutaneous malignant melanoma were enrolled, 27 of them were women and 24 men aged from 17 to 84 years. The clinical diagnosis was confirmed histological in43 patients. The histological diagnosis in 8 patients was dysplastic nevus. We determined the histopatological factors in melanomasČ Breslow tumor thickness, Clark level of invasion, microscopic ulceration on the tumor surface, vascular invasion, type of vertical growth, number of tumor infiltrating lymphocytes, and microscopic satellites. The tissue levels of theurokinase system components (uPA, PAI-1 and PAI-2) were quantified by enzyme-linked immunosorbent assay (ELISA) and were termed in ngžmg proteins. The levels of uPA were determined in tissue extracts of 36 melanomas and 5 dysplastic nevi and healthy skin, and the levels of PAI-1 and PAI-2 in a cytosol of all melanomas and dysplastic nevi and healthy skin.
