    Background: Type II phospholipase A2 (PLA2) is an enzyme important not only inthe process of malignant transformation of the cells but also in the processof invasion. The aim of the present study was ... to investigate the expression of type II PLA2 in the carcinomas of the colorectum, peritumoral mucosa and in the mucosa distant from the tumor. Methods: Resection specimens from 57 patients with colorectal carcinoma (caecum 10, ascending colon 10, transverse colon 10, sigmoid colon 9, and rectum 18) were analysed immunohistochemically. Histological slides from paraffin blocks were stained by the human monoclonal type II PLA2 antibody (ĆUpstate BiotechnologyĆ, Lake Placid, NY 12946, USA. Antibody ClassČ IgGlk. ImunogenČ HPC purifed human sperm phospholipase A2 - 14 kDa enzyme) using the standard DAKO peroxidase labelled Streptavidin-Biotin method and TechMate 500 stainer. Type II PLA2 expression was evaluated semiquantitatively according to the extensivity and intensivity of staining. For statistical evaluation the Kruskal-Wallis one wayanalysis of variance on ranks and the Mann-Whitney rank sum tests were used. ResultsČ The highest expression of type II PLA2 was found in the peritumoral mucosa (median 4.00). It was much lower in the mucosa distant fromthe tumor (median 0.70) and almost absent in the tumor itself (median 0.00). All differences were statistically significant (Kruskal-Wallis test, all pairwise multiple comparison procedures - Dunnćs MethodČ p<0.05). The expression of type II PLA2 was higher in the left colon and rectum than in theright colon (Mann-Whitney rank sum testČ p<0.0`). ConclusionsČ Our results suggest that there is variation of the type II PLA2 expression throughout the mucosa and tumors of the colorectum, which might reflect the progression of neoplastic process.
