    Several active pharmaceutical ingredients exhibit low water solubility and poor permeability in the intestinal area. This inevitably affects the bioavailability of the drug and therefore its ... efficiency. The problem of low water solubility of active pharmaceutical ingredients is a central topic and several approaches are studied to overcome this drawback. One of these is represented by the employment of solid dispersions, where the drug is loaded inside an inert matrix, which increases dissolution rate of the drug, leads to it amorphisation and prevents recrystallization. Naproxen is a widely used drug, however it has low water solubility and consequently it has been studied in several approaches. Herein the attention is focused on solid dispersions of naproxen in different types of Syloid, a non-ordered porous silica. A porous carrier may store the active pharmaceutical ingredient inside the pores, decreasing drug dimensions and promoting amorphisation, and silica favours dissolution in aqueous medium thanks to its hydrophilicity. Solid dispersions of naproxen in non-ordered silica have been prepared employing three manufacturing techniques. These are here compared because the drug is loaded in different ways and this influences the release and the final properties of the system. Solid dispersions have been produced through solvent evaporation, using a high volume of drug solution or with an incipient wetness impregnation. Physical mixtures have been prepared also, firstly as specimen to confirm solid dispersions, secondly as drug delivery systems themselves. The attention was focused on how the properties of the excipients and the dimension of the drug loaded influence the dissolution from physical mixtures. The incipient wetness impregnation method results the best option to load naproxen in non-ordered porous silica Syloid. Moreover, physical mixtures show to be promising drug delivery systems and not only used as specimens. In addition, Syloid has been confirmed as an appropriate excipient in manufacturing of solid dispersions of naproxen.
