  • Adipogenic and osteogenic potential in mesenchymal stem cells derived from osteoarthritic donors. = Osteogen och adipogen potential i mesenchymala stamceller hos patienter med artros : [projektna naloga]
    Andersson, Lina
Osteoarthritis is one of the most common diseases in the western world, with a prevalence of 1:4 in the population >45 years old, and twice as common in women than men. Osteoarthritis is ... characterized by joint degeneration and there are no cures for osteoarthritis but joint replacement surgery, and treatment usually consists of pain and symptomatic relieving combined with physical activity. Mesenchymal stem cells (MSCs) are multipotent stem cells existing in several tissues, such as cartilage, bone, muscles and adipose tissue. It has been proven that the MSCs are disturbed in several conditions and diseases and that their multipotency and tissue regeneration are affected. It has been proven that bone-derived MSCs in osteoporosis have a shift from osteogenesis to adipogenesis which could explain the mechanism of low bone density in this disorder. MSCs are also a promising tool in tissue engineering to repair and treat degenerative disorders, but before treatment can be applied to a disease, the multipotency of MSCs needs to be evaluated. MSCs derived from trabecular bone and skeletal muscle from a total of 13 donors with primary osteoarthritis, divided to 29 samples, were cultivated in vitro and differentiated to adipocytes and osteoblasts. To evaluate their adipogenic and osteogenic potential, adipogenic and osteogenic specific gene expressions were measured with quantitative real-time polymerase chain reaction. We discovered that muscle-derived MSCs had a statistically significant higher osteogenic potential compared to bone-derived MSCs, but a similar adipogenic potential. However, to fully evaluate the clinical significance of these results, a larger study needs to be executed.
