  • Comparison of HPLC columns for the separation of selected xanthophylls appearing in egg yolks [Elektronski vir]

    Xanthophylls lutein and zeaxanthin are important for the prevention of macular degeneration and cataract development in human eyes. Green leafy vegetables are a good source of lutein and less of ... zeaxanthin, however the bioavailability of lutein is the highest from the egg yolks. For the improvement of technological properties of eggs, natural or synthetic xanthophylls are added in hens feed. For egg quality and verification of organic production the content and the type of xanthophylls should be quantified. C18 columns or more hydrophobic C30 columns are mostly used for the separation of xanthophylls. The aim of our study was to compare four different columns from three different producers for the separation of up to eight selected xanthophylls presented in egg yolk. The follow- ing columns were tested: Kinetex C18 core-shell column (100 mm*4.6 mm I.D., particle size 2.6 m, Phenomenex, Torrance, USA), Acclaim C30 column (250 mm*4.6 mm I.D., particle size 5 nm, Dionex, Sunnyvale, CA, USA), Prontosil C30 column (250 mm*4.6 mm I.D., particle size 5 nm, Bischoff, Leonberg, Germany) and Prontosil C30 column, (250 mm*4.6 mm I.D., particle size 3 nm, Bischoff). Solvents composition needed for the separation of xanthophylls on Acclaim C30 column was more similar to the mobile phase needed for the Kinetex C18 column than for the other two Prontosil C30 columns, probably because of the lower carbon load on Acclaim C30 column in comparison to Prontosil C30. Retention time, resolution, asymmetry, theoretical plates, limit of detectionand quantification were also obtained and compared.
