Utjecaj ozona na planktonske bakterije i biofilm Legionella pneumophila [Elektronski vir]Dobrić, Dorotea ; Kovač, Bruno ; Gobin, IvanaLegionella pneumophila is an aerobic, gram-negative bacterium found naturally in the aquatic environment, and accordingly, due to improper maintenance, it can be detected in the water supply system ... of humans in the form of biofilm. By forming a biofilm, bacteria resist water flow and thus provide themselves with the nutrients necessary for reproduction and greater resistance to disinfectants. The purpose of the study was to investigate the impact of ozone on the planktonic bacteria L. pneumophila and their biofilm. For the cultivation of biofilms, ceramic tiles and stainless-steel tiles were used. Planktonic Legionella were prepared in a bacterial suspension in sterile tap water and direct exposure to ozone. Biofilm cultivation was carried out for these materials over 24 hours and 5 days. Stainless steel tiles were added to the bacterial suspension inside the 12-well, while ceramic tiles were placed in glass Petri dishes with the addition of agar in such a way that it did not cover the upper side of the tiles and were topped with a prepared bacterial suspension. The treatment for both materials was the same; the tiles were washed with sterile tap water and then treated with an ozone concentration of 25 ppm within an hour of exposure. They were placed in an ultrasonic bath for 1 minute, and a series of tenfold dilutions was made. After planting on BCYE agar and cultivation at (35±1) °C for 4 days, the CFU/mL was determined by counting colonies. For each treated material, a control sample has been prepared that has undergone equal treatment, excluding ozonization.
