This study aimed to define progesterone 5β-reductases (P5βR, EC 1.3.99.6, enone 1,4-reductases) as function-associated molecular markers at the plant family level. Therefore cDNAs were isolated from ...25 Brassicaceae species, including two species, Erysimum crepidifolium and Draba aizoides, known to produce cardiac glycosides. The sequences were used in a molecular phylogeny study. The cladogram created is congruent to the existing molecular analyses. Recombinant His-tagged forms of the P5βR cDNAs from Aethionema grandiflorum, Draba aizoides, Nasturtium officinale, Raphanus sativus and Sisymbrium officinale were expressed in E. coli. Enone 1,4-reductase activity was demonstrated in vitro using progesterone and 2-cyclohexen-1-one as substrates. Evidence is provided that functional P5βRs are ubiquitous in the Brassicaceae. The recombinant P5βR enzymes showed different substrate preferences towards progesterone and 2-cyclohexen-1-one. Sequence comparison of the catalytic pocket of the P5βR enzymes and homology modelling using Digitalis lanata P5βR (PDB ID: 2V6G) as template highlighted the importance of the hydrophobicity of the binding pocket for substrate discrimination. It is concluded that P5βR genes or P5βR proteins can be used as valuable function-associated molecular markers to infer taxonomic relationship and evolutionary diversification from a metabolic/catalytic perspective.
The VIP collaboration is performing high precision tests of the Pauli Exclusion Principle for electrons in the extremely low cosmic background environment of the Underground Gran Sasso Laboratories ...of INFN (Italy). The experimental technique consists in introducing a DC current in a copper conductor, searching for the X-rays emission due to a PEP-forbidden atomic transition from the L shell to the K shell of copper when the K shell is already occupied by two electrons. VIP set un upper limit on the PEP violation probability 12β2<4.7×10−29, the aim of the upgraded VIP-2 experiment is to improve this result by two orders of magnitude at least. The experimental setup and the results of preliminary data analyses will be presented.

We investigate atomic transitions with precision X-ray spectroscopy in order to test the Pauli Exclusion Principle (PEP) and therefore the spin-statistics theorem. We present the experimental method for the search for "anomalous" (i.e. Pauli-forbidden) X-ray transitions in copper atoms, produced by "new" electrons, which could have a tiny probability to undergo a Pauli-forbidden transition to the 1s ground state already occupied by two electrons. We describe the VIP-2 experimental setup and its recent optimisations. Presently VIP-2 is taking data at LNGS. The goal of VIP-2 is to test PEP for electrons with unprecedented accuracy, down to a limit in the probability that PEP is violated at the level of 10−31 (and using a more elaborate analysis even 10−40). We present current experimental results.
In the VIP2 VIolation of the Pauli Exlusion Principle (PEP) experiment at the Gran Sasso underground laboratory (LNGS) we are searching for possible violations of standard quantum mechanics ...predictions. With high precision we investigate the Pauli Exclusion Principle and the collapse of the wave function (collapse models). We will present our experimental method of searching for possible small violations of the Pauli Exclusion Principle for electrons, via the search for "anomalous" X-ray transitions in copper atoms, produced by "new" electrons (brought inside a copper bar by circulating current) which could have the probability to undergo Pauli-forbidden transition to the ground state (1 s level) already occupied by two electrons. We will describe the concept of the VIP2 experiment taking data at LNGS presently. The goal of VIP2 is to test the PEP for electrons with unprecedented accuracy, down to a limit in the probability that PEP is violated at the level of 10−31. We will show preliminary experimental results obtained at LNGS and discuss implications of a possible violation.

The KLOE detector was designed primarily for the study of CP violation in neutral kaon decays at DAΦNE, the Frascati φ-factory. The detector consists of a tracker and an electromagnetic calorimeter. ...A lead-scintillating-fiber sampling calorimeter satisfies best the requirements of the experiment, providing adequate energy resolution and superior timing accuracy. We describe in the following the construction of the calorimeter, its calibration and how the calorimeter information is used to obtain energy, point of entry and time of the arrival of photons, electrons and charged particles. With e+e− collision data at DAΦNE for an integrated luminosity of some 2pb−1 we find for electromagnetic showers, an energy resolution of 5.7%/E(GeV) and a time resolution of 54/E(GeV) ps. We also present a measurement of efficiency for low energy photons.
The Pauli Exclusion Principle (PEP) was introduced by the austrian physicist Wolfgang Pauli in 1925. Since then, several experiments have checked its validity. From 2006 until 2010, the VIP ...(Violation of the Pauli Principle) experiment took data at the LNGS underground laboratory to test the PEP. This experiment looked for electronic 2p to Is transitions in copper, where 2 electrons are in the Is state before the transition happens. These transitions violate the PEP. The lack of detection of X-ray photons coming from these transitions resulted in a preliminary upper limit for the violation of the PEP of 4.7 × 10-29. Currently, the successor experiment VIP2 is under preparation. The main improvements are, on one side, the use of Silicon Drift Detectors (SDDs) as X-ray photon detectors. On the other side an active shielding is implemented, which consists of plastic scintillator bars read by Silicon Photomultipliers (SiPMs). The employment of these detectors will improve the upper limit for the violation of the PEP by around 2 orders of magnitude.
