Because of the scarcity of meteorological observations, the precipitation climate on the Tibetan Plateau and surrounding regions (TP) has been insufficiently documented so far. In this study, the ...characteristics and basic features of precipitation on the TP during an 11-yr period (2001–11) are described on monthly-to-annual time scales. For this purpose, a new high-resolution atmospheric dataset is analyzed, the High Asia Reanalysis (HAR), generated by dynamical downscaling of global analysis data using the Weather Research and Forecasting (WRF) model. The HAR precipitation data at 30- and 10-km resolutions are compared with both rain gauge observations and satellite-based precipitation estimates from the Tropical Rainfall Measurement Mission (TRMM). It is found that the HAR reproduces previously reported spatial patterns and seasonality of precipitation and that the high-resolution data add value regarding snowfall retrieval, precipitation frequency, and orographic precipitation. It is demonstrated that this process-based approach, despite some unavoidable shortcomings, can improve the understanding of the processes that lead to precipitation on the TP. Analysis focuses on precipitation amounts, type, seasonality, and interannual variability. Special attention is given to the links between the observed patterns and regional atmospheric circulation. As an example of an application of the HAR, a new classification of glaciers on the TP according to their accumulation regimes is proposed, which illustrates the strong spatial variability of precipitation seasonality. Finally, directions for future research are identified based on the HAR, which has the potential to be a useful dataset for climate, glaciological, and hydrological impact studies.
High‐altitude meteorological processes in the Himalaya are influenced by complex interactions between the topography and the monsoon and westerly circulation systems. In this study, we use the ...Weather Research and Forecasting model configured with high spatial resolution to understand seasonal patterns of near‐surface meteorological fields and precipitation processes in the Langtang catchment in the central Himalaya. Using a unique high‐altitude observational network, we evaluate a simulation from 17 June 2012 to 16 June 2013 and conclude that, at 1 km horizontal grid spacing, the model captures the main features of observed meteorological variability in the catchment. The finer representation of the complex terrain and explicit simulation of convection at this grid spacing give strong improvements in near‐surface air temperature and small improvements in precipitation, in particular in the magnitudes of daytime convective precipitation and at higher elevations. The seasonal differences are noteworthy, including a reversal in the vertical and along‐valley distributions of precipitation between the monsoon and winter seasons, with peak values simulated at lower altitudes (~3000 m above sea level (asl)) and in the upper regions (above 5000 m asl) in each season, respectively. We conclude that there is great potential for improving the local accuracy of climate change impact studies in the Himalaya by using high‐resolution atmospheric models to generate the forcing for such studies.

Electroconvulsive seizure therapy (ECS) is a clinically proven treatment for depression and is often effective even in patients resistant to chemical antidepressants. However, the molecular ...mechanisms underlying the therapeutic efficacy of ECS are not fully understood. One theory that has gained attention is that ECS and other antidepressants increase the expression of select neurotrophic factors that could reverse or block the atrophy and cell loss resulting from stress and depression. To further address this topic, we examined the expression of other neurotrophic-growth factors and related signaling pathways in the hippocampus in response to ECS using a custom growth factor microarray chip. We report the regulation of several genes that are involved in growth factor and angiogenic-endothelial signaling, including neuritin, stem cell factor, vascular endothelial growth factor (VEGF), VGF (nonacronymic), cyclooxygenase-2, and tissue inhibitor of matrix metalloproteinase-1. Some of these, as well as other growth factors identified, including VEGF, basic fibroblast growth factor, and brain-derived neurotrophic factor, have roles in mediating neurogenesis and cell proliferation in the adult brain. We also examined gene expression in the choroid plexus and found several growth factors that are enriched in this vascular tissue as well as regulated by ECS. These data suggest that an amplification of growth factor signaling combined with angiogenic mechanisms could have an important role in the molecular action of ECS. This study demonstrates the applicability of custom-focused microarray technology in addressing hypothesis-driven questions regarding the action of antidepressants.
There is strong variation in glacier mass balances in High Mountain Asia. Particularly interesting is the fact that glaciers are in equilibrium or even gaining mass in the Karakoram and Kunlun Shan ...ranges, which is in sharp contrast with the negative mass balances in the rest of High Mountain Asia. To understand this difference, an in-depth understanding of the meteorological drivers of the glacier mass balance is required. In this study, two catchments in contrasting climatic regions, one in the central Himalaya (Langtang) and one in the Karakoram (Shimshal), are modeled at 1 km grid spacing with the numerical atmospheric model WRF for the period of 2011–2013. Our results show that the accumulation and melt dynamics of both regions differ due to contrasting meteorological conditions. In Shimshal, 92% of the annual precipitation falls in the form of snow, in contrast with 42% in Langtang. In addition, 80% of the total snow falls above an altitude of 5000 m a.s.l, compared with 35% in Langtang. Another prominent contrast is that most of the annual snowfall falls between December and May (71%), compared with 52% in Langtang. The melt regimes are also different, with 41% less energy available for melt in Shimshal. The melt in the Karakoram is controlled by net shortwave radiation (r = 0.79 ± 0.01) through the relatively low glacier albedo in summer, while net longwave radiation (clouds) dominates the energy balance in the Langtang region (r = 0.76 ± 0.02). High amounts of snowfall and low melt rates result in a simulated positive glacier surface mass balance in Shimshal (+0.31 ± 0.06 m w.e. year−1) for the study period, while little snowfall, and high melt rates lead to a negative mass balance in Langtang (−0.40 ± 0.09 m w.e. year−1). The melt in Shimshal is highly variable between years, and is especially sensitive to summer snow events that reset the surface albedo. We conclude that understanding glacier mass balance anomalies requires quantification and insight into subtle shifts in the energy balance and accumulation regimes at high altitude and that the sensitivity of glaciers to climate change is regionally variable.
Insulin resistance is a major factor in the pathogenesis of type 2 diabetes and is strongly associated with obesity. Increased concentrations of intracellular fatty acid metabolites have been ...postulated to interfere with insulin signaling by activation of a serine kinase cascade involving PKCtheta in skeletal muscle. Uncoupling protein 3 (UCP3) has been postulated to dissipate the mitochondrial proton gradient and cause metabolic inefficiency. We therefore hypothesized that overexpression of UCP3 in skeletal muscle might protect against fat-induced insulin resistance in muscle by conversion of intramyocellular fat into thermal energy. Wild-type mice fed a high-fat diet were markedly insulin resistant, a result of defects in insulin-stimulated glucose uptake in skeletal muscle and hepatic insulin resistance. Insulin resistance in these tissues was associated with reduced insulin-stimulated insulin receptor substrate 1- (IRS-1-) and IRS-2-associated PI3K activity in muscle and liver, respectively. In contrast, UCP3-overexpressing mice were completely protected against fat-induced defects in insulin signaling and action in these tissues. Furthermore, these changes were associated with a lower membrane-to-cytosolic ratio of diacylglycerol and reduced PKCtheta activity in whole-body fat-matched UCP3 transgenic mice. These results suggest that increasing mitochondrial uncoupling in skeletal muscle may be an excellent therapeutic target for type 2 diabetes mellitus.
High Asia has experienced strong environmental changes in recent decades, as evident in records of glaciers, lakes, tree rings, and vegetation. The multiscale understanding of the climatic drivers, ...however, is still incomplete. In particular, few systematic assessments have evaluated to what degree, if at all, the midlatitude westerly circulation modifies local surface climates in the reach of the Indian Summer Monsoon. This paper shows that a southward shift of the upper‐tropospheric westerlies contributes significantly to climate variability in the core monsoon season (July–September) by two prominent dipole patterns at the surface: cooling in the west of High Asia contrasts with warming in the east, while moist anomalies in the east and northwest occur with drying along the southwestern margins. Circulation anomalies help to understand the dipoles and coincide with shifts in both the westerly wave train and the South Asian High, which imprint on air mass advection and local energy budgets. The relation of the variabilities to a well‐established index of midlatitude climate dynamics allows future research on climate proxies to include a fresh hypothesis for the interpretation of environmental changes.

Electroconvulsive seizure has a proven therapeutic application in the treatment of severe depression and treatment‐resistant depression. Despite the efficacy of electroconvulsive seizure as a ...non‐chemical antidepressant treatment, the mechanism of action is unclear. Elevation in hippocampal trophic factor expression and concomitant cellular proliferation are thought to play a role in its action. We examined whether the reported induction of angiogenic factors and endothelial cell proliferation leads to an increase in vascular density. Two hippocampal regions, the dentate gyrus and the stratum lacunosum moleculare (SLM), were examined employing a combination of vascular density quantification, angiogenic gene expression analysis and immunohistochemistry. A 6% increase in vascular density was observed in the dentate gyrus but this did not achieve statistical significance. The SLM of the hippocampus exhibited a robust 20–30% increase in vascular density and was accompanied by an increase in expression of inhibitor of differentiation‐3. There was also an induction of the angiogenesis markers αVβ3 integrin and Del1. Increases in the vascular density of the SLM could be in response to enhanced metabolic activity in this region. This is supported by the induction of glutamine synthetase and the glutamate transporter GLT1.
Degradation of the vascular basement membrane stimulates angiogenesis and is tightly controlled by balancing the actions of metalloproteases and their inhibitors. Previous work demonstrated that ...electroconvulsive seizure (ECS) elevates angiogenic factors and endothelial proliferation in the hippocampus. The robust induction of tissue inhibitor of matrix metalloprotease 1 (TIMP-1) in the stratum lacunosum moleculare (SLM) corresponds to sites of increased vascular density. This led us to examine the spatial and cellular expression of TIMP-1 and its substrate, matrix metalloprotease 9 (MMP-9). Chronic ECS increased TIMP-1 by 12-fold and MMP-9 by 3-fold in discrete SLM cells. We then characterized the expression of TIMP-1 mRNA in relation to vasculature in the SLM and glial-limiting membrane (GLM). Employing laser microdissection we identified the cell types associated with SLM vasculature and also phenotyped the cells expressing TIMP-1 and MMP-9. We concluded that TIMP-1 is produced by perivascular cells positive for alpha smooth actin and that MMP-9 is expressed by GFAP-positive astrocytes. These studies suggest that ECS-induced remodelling occurs at the vascular basement membrane and facilitates neovascularization.
Accurate knowledge of the impact of internal atmospheric variability is required for the detection and attribution of climate change and for interpreting glacier records. However, current knowledge ...of such impacts in high-mountain regions is largely based on statistical methods, as the observational data required for process-based assessments are often spatially or temporally deficient. Using a case study of Kilimanjaro, 12 years of convection-permitting atmospheric modeling are combined with an 8-yr observational record to evaluate the impact of climate oscillations on recent high-altitude atmospheric variability during the short rains (the secondary rain season in the region). The focus is on two modes that have a well-established relationship with precipitation during this season, El Niño–Southern Oscillation and the Indian Ocean zonal mode, and demonstrate their strong association with local and mesoscale conditions at Kilimanjaro. Both oscillations correlate positively with humidity fluctuations, but the association is strongest with the Indian Ocean zonal mode in the air layers near and above the glaciers because of changes in zonal circulation and moisture transport, emphasizing the importance of the moisture signal from this basin. However, it is found the most anomalous conditions during co-occurring positive events because of the combined effects of the (i) extended positive sea surface temperature anomalies, (ii) enhanced atmospheric moisture capacity from higher tropospheric temperatures, (iii) most pronounced weakening of the subsiding branch of the Indian Ocean Walker circulation over East Africa, and (iv) stronger monsoonal moisture fluxes upstream from Kilimanjaro. This study lays the foundation for unraveling the contribution of climate modes to observed changes in Kilimanjaro’s glaciers.
A better understanding of regional‐scale precipitation patterns in the Himalayan region is required to increase our knowledge of the impacts of climate change on downstream water availability. This ...study examines the impact of four cloud microphysical schemes (Thompson, Morrison, Weather Research and Forecasting (WRF) single‐moment 5‐class, and WRF double‐moment 6‐class) on summer monsoon precipitation in the Langtang Valley in the central Nepalese Himalayas, as simulated by the WRF model at 1 km grid spacing for a 10 day period in July 2012. The model results are evaluated through a comparison with surface precipitation and radiation measurements made at two observation sites. Additional understanding is gained from a detailed examination of the microphysical characteristics simulated by each scheme, which are compared with measurements using a spaceborne radar/lidar cloud product. Also examined are the roles of large‐ and small‐scale forcings. In general, the schemes are able to capture the timing of surface precipitation better than the actual amounts in the Langtang Valley, which are predominately underestimated, with the Morrison scheme showing the best agreement with the measured values. The schemes all show a large positive bias in incoming radiation. Analysis of the radar/lidar cloud product and hydrometeors from each of the schemes suggests that “cold‐rain” processes are a key precipitation formation mechanism, which is also well represented by the Morrison scheme. As well as microphysical structure, both large‐scale and localized forcings are also important for determining surface precipitation.
