Dietary advanced glycation endproducts (AGEs), abundantly present in Westernized diets, are linked to negative health outcomes, but their impact on the gut microbiota has not yet been well ...investigated in humans. We investigated the effects of a 4-week isocaloric and macronutrient-matched diet low or high in AGEs on the gut microbial composition of 70 abdominally obese individuals in a double-blind parallel-design randomized controlled trial (NCT03866343). Additionally, we investigated the cross-sectional associations between the habitual intake of dietary dicarbonyls, reactive precursors to AGEs, and the gut microbial composition, as assessed by 16S rRNA amplicon-based sequencing. Despite a marked percentage difference in AGE intake, we observed no differences in microbial richness and the general community structure. Only the Anaerostipes spp. had a relative abundance >0.5% and showed differential abundance (0.5 versus 1.11%; p = 0.028, after low- or high-AGE diet, respectively). While the habitual intake of dicarbonyls was not associated with microbial richness or a general community structure, the intake of 3-deoxyglucosone was especially associated with an abundance of several genera. Thus, a 4-week diet low or high in AGEs has a limited impact on the gut microbial composition of abdominally obese humans, paralleling its previously observed limited biological consequences. The effects of dietary dicarbonyls on the gut microbiota composition deserve further investigation.
Application of alginate-microencapsulated therapeutic cells is a promising approach for diseases that require a local and constant supply of therapeutic molecules. However most conventional alginate ...microencapsulation systems are associated with low mechanical stability and protrusion of cells which is associated with higher surface roughness and limits their clinical application. Here we have developed a novel multilayer encapsulation system that prevents cells from protruding from capsules. The system was tested using a therapeutic protein with anti-tumor activity overexpressed in mammalian cells. The cell containing core of the multilayer capsule was formed by flexible alginate, creating a cell sustaining environment. Surrounded by a poly-L-lysine layer the flexible core was enveloped in a high-G alginate matrix that is less flexible and has higher mechanical stability, which does not support cell survival. The cells in the core of the multilayer capsule did not show growth impairment and protein production was normal for periods up to 70 days in vitro. The additional alginate layer also lowered the surface roughness compared to conventional cell containing alginate-PLL capsules. Our system provides a solution for two important, often overlooked phenomena in cell encapsulation: preventing cell protrusion and improving surface roughness.
In a prospective, multicenter, open‐label study, de novo liver transplant patients were randomized at day 30±5 to (i) everolimus initiation with tacrolimus elimination (TAC Elimination) (ii) ...everolimus initiation with reduced‐exposure tacrolimus (EVR+Reduced TAC) or (iii) standard‐exposure tacrolimus (TAC Control). Randomization to TAC Elimination was terminated prematurely due to a higher rate of treated biopsy‐proven acute rejection (tBPAR). EVR+Reduced TAC was noninferior to TAC Control for the primary efficacy endpoint (tBPAR, graft loss or death at 12 months posttransplantation): 6.7% versus 9.7% (−3.0%; 95% CI −8.7, 2.6%; p<0.001 for noninferiority 12% margin). tBPAR occurred in 2.9% of EVR+Reduced TAC patients versus 7.0% of TAC Controls (p = 0.035). The change in adjusted estimated GFR from randomization to month 12 was superior with EVR+Reduced TAC versus TAC Control (difference 8.50 mL/min/1.73 m2, 97.5% CI 3.74, 13.27 mL/min/1.73 m2, p<0.001 for superiority). Drug discontinuation for adverse events occurred in 25.7% of EVR+Reduced TAC and 14.1% of TAC Controls (relative risk 1.82, 95% CI 1.25, 2.66). Relative risk of serious infections between the EVR+Reduced TAC group versus TAC Controls was 1.76 (95% CI 1.03, 3.00). Everolimus facilitates early tacrolimus minimization with comparable efficacy and superior renal function, compared to a standard tacrolimus exposure regimen 12 months after liver transplantation.
This prospective, randomized, multicenter, open‐label study in de novo liver transplant patients shows that initiation of everolimus at day 30 posttransplant facilitates tacrolimus minimization but not elimination, and results in comparable efficacy and superior renal function but increased adverse events compared to a standard tacrolimus exposure regimen.

We investigated to what extent tube voltage, scanner model, and reconstruction algorithm affect radiomic feature reproducibility in a single-institution retrospective database of computed ...tomography images of non-small-cell lung cancer patients.
Methods
This study was approved by the Institutional Review Board (UID 2412). Images of 103 patients were considered, being acquired on either among two scanners, at 100 or 120 kVp. For each patient, images were reconstructed with six iterative blending levels, and 1414 features were extracted from each reconstruction. At univariate analysis, Wilcoxon-Mann-Whitney test was applied to evaluate feature differences within scanners and voltages, whereas the impact of the reconstruction was established with the overall concordance correlation coefficient (OCCC). A multivariable mixed model was also applied to investigate the independent contribution of each acquisition/reconstruction parameter. Univariate and multivariable analyses were combined to analyse feature behaviour.
Results
Scanner model and voltage did not affect features significantly. The reconstruction blending level showed a significant impact at both univariate analysis (154/1414 features yielding an OCCC < 0.85) and multivariable analysis, with most features (1042/1414) revealing a systematic trend with the blending level (multiple comparisons adjusted
p
< 0.05). Reproducibility increased in association to image processing with smooth filters, nonetheless specific investigation in relation to clinical endpoints should be performed to ensure that textural information is not removed.
Conclusions
Combining univariate and multivariable models is allowed to identify features for which corrections may be applied to reduce the trend with the algorithm and increase reproducibility. Subsequent clustering may be applied to eliminate residual redundancy.

The predatory firefly Photuris elliptica is common throughout the Atlantic Forest and has been proposed as a biomonitor due to the species’ narrow niche and elevational range. However, the species is ...only known from adults, and a more effective monitoring of its populations hinges on the lack of knowledge on their immature stages. Recent sampling in ferruginous caves and inserted in other lithologies, on sites in the Atlantic Forest and Cerrado, have led to the capture of firefly larvae later reared to adults in the lab. Firefly larvae have been reported in South American caves before; however, they have only been identified to family due to the adult-biased taxonomy of Lampyridae. Here, we provide an updated diagnosis of Photuris elliptica , describe its immature stages for the first time, and update the distribution of the species. The larvae of Photuris elliptica were observed to interact with guano of several bat species, including that of vampire bats. These observations are consistent with the less specialized feeding preferences of photurine larvae, unlike most other firefly taxa, which specialize in gastropods and earthworms. It is yet unclear whether P. elliptica are cave specialists. However, since its occurrence outside caves remains unknown, protecting cave environments must be considered in conservation strategies for this important biomonitor species.
The fully printed, hole-transporter-free carbon perovskite solar cell structure incorporating a triple mesoscopic layer has emerged as a possible frontrunner for early industrialisation. It is an ...attractive structure because it can be fabricated by the simple sequential screen printing and sintering of titania, zirconia, and carbon. The device is finalised by manual dropping of a perovskite precursor solution onto the carbon which subsequently infiltrates. This stage in device fabrication is inhomogeneous, ineffective for large areas, and prone to human error. Here we introduce an automated deposition and infiltration system using a robotic dispenser and mesh which delivers the perovskite precursor uniformly to the carbon surface over a large area. It has been successfully used to prepare perovskite solar cells with over 9% efficiency. Cells, prepared by this robotic mesh deposition, showed comparable performance to reference cells, made by standard drop deposition, confirming this approach to be effective and reliable. X-ray diffraction and Raman spectroscopy were used to confirm the uniformity of the deposition over a large area.
OBJECTIVE:To evaluate the chronic effects of a static stretching program on the muscle architecture of biceps femoris (BF) and vastus lateralis (VL) muscles in ultrasound (US) images.

SETTING:Biomechanics Laboratory of Physical Education School of the Army, Rio de Janeiro, Brazil.
PARTICIPANTS:The study included 24 healthy and physically active male volunteers (19.05 ± 1.40 years, 1.73 ± 0.07 m, and 73.15 ± 8.33 kg), randomly allocated to 1 of 2 groupsstretching group (SG, n = 12) and control group (n = 12).
INTERVENTIONS:The SG was submitted to 3 sets of 30 seconds of static stretching 3 times a week during 8 weeks.
MAIN OUTCOME MEASURES:Ultrasound equipment (7.5 MHz) was used for the evaluation of BF and VL muscle architecture variables (pennation angle, fiber length, muscle thickness, and fascicle displacement) before and after training. Knee range of motion (ROM) and isometric flexion and extension torque (TQ) were also measured.
RESULTS:There were no significant changes in muscle architecture, TQ, and maximum knee flexion angle (P > 0.05). However, maximum knee extension angle (MEA) increased significantly in the SG (pretraining159.37 ± 7.27 degrees and posttraining168.9 ± 3.7 degrees; P < 0.05).
CONCLUSIONS:Volume or intensity (or both) of the stretching protocol was insufficient to cause structural changes in the VL and BF muscles. The increase in MEA could not be explained by muscle architecture changes.
CLINICAL RELEVANCE:To describe changes in the VL and BF muscle tendon unit using US after a long-term stretching program to identify which structures are responsible for ROM increase.
Regional nutrient ranges are commonly used to diagnose plant nutrient status. In contrast, local diagnosis confronts unhealthy to healthy compositional entities in comparable surroundings. Robust ...local diagnosis requires well-documented data sets processed by machine learning and compositional methods. Our objective was to customize nutrient diagnosis of peach (Prunus persica) trees at local scale. We collected 472 observations from commercial orchards and fertilizer trials across eleven cultivars of Prunus persica and six rootstocks in the state of Rio Grande do Sul (RS), Brazil. The random forest classification model returned an area under curve exceeding 0.80 and classification accuracy of 80% about yield cutoff of 16 Mg ha−1. Centered log ratios (clr) of foliar defective compositions have appropriate geometry to compute Euclidean distances from closest successful compositions in “enchanting islands”. Successful specimens closest to defective specimens as shown by Euclidean distance allowed reaching trustful fruit yields using site-specific corrective measures. Comparing tissue composition of low-yielding orchards to that of the closest successful neighbors in two major Brazilian peach-producing regions, regional diagnosis differed from local diagnosis, indicating that regional standards may fail to fit local conditions. Local diagnosis requires well-documented Humboldtian data sets that can be acquired through ethical collaboration between researchers and stakeholders.
