/mass of sample), cosolvent percentage (0, 1, 2 and 4%) and global yield isotherms as a function of different pressures (250, 350 and 450 bar) and temperatures (31.7, 40 and 50 °C). Within the investigated parameters, the best conditions found were an S/F of 131 and the use of ethanol at the highest concentration (4%

), which resulted in higher extract yields and higher content of antioxidant compounds. Formononetin, the main biomarker of red propolis, was the compound found at the highest amounts in the extracts. As expected, the temperature and pressure conditions also influenced the process yield, with 350 bar and 40 °C being the best conditions for obtaining bioactive compounds from a sample of red propolis. The novel results for red propolis found in this study show that it is possible to obtain extracts with high antioxidant potential using a clean technology under the defined conditions.

To map OTA/AO type 43C3 tibial pilon fractures by means of computed tomography and analyze the difference between varus and valgus fractures. We hypothesized that valgus fractures are less frequent than varus, and the affected zones of the tibial pilon are different among the patterns.
Retrospective, cross-sectional, observational study, using images of 73 computed tomographies of patients who had presented OTA/AO type 43C3 tibial pilon fractures. The radiographs and computed tomography were subdivided into two groups: varus and valgus fractures. Also, the presence or absence of fibula fractures. Both groups were subdivided into eight zones: four lateral to the pilon and four medial.
The distribution of total affected zones is significantly different in the male and female subgroups (p = 0.027). The incidence of cases in zone 1 is significantly different in varus and valgus displacement subgroups (p = 0.002). In the patients with valgus displacements, 61.9% of the fractures affect zone 1; in the patients with varus displacements, only 25.8% of the fractures affect zone 1. In the patients without fibula fracture, 58.3% of the fractures affect zone 2; in the patients with fibula fracture, 24.6% of the fractures affect zone 2.

The main challenge in the control of malaria has been the emergence of drug-resistant parasites. The presence of drug-resistant Plasmodium sp. has raised the need for new antimalarial drugs. ...Molecular modelling techniques have been used as tools to develop new drugs. In this study, we employed virtual screening of a pyrazol derivative (Tx001) against four malaria targets: plasmepsin-IV, plasmepsin-II, falcipain-II, and PfATP6. The receiver operating characteristic curves and area under the curve (AUC) were established for each molecular target. The AUC values obtained for plasmepsin-IV, plasmepsin-II, and falcipain-II were 0.64, 0.92, and 0.94, respectively. All docking simulations were carried out using AutoDock Vina software. The ligand Tx001 exhibited a better interaction with PfATP6 than with the reference compound (-12.2 versus -6.8 Kcal/mol). The Tx001-PfATP6 complex was submitted to molecular dynamics simulations in vacuum implemented on an NAMD program. The ligand Tx001 docked at the same binding site as thapsigargin, which is a natural inhibitor of PfATP6. Compound TX001 was evaluated in vitro with a P. falciparum strain (W2) and a human cell line (WI-26VA4). Tx001 was discovered to be active against P. falciparum (IC50 = 8.2 µM) and inactive against WI-26VA4 (IC50 > 200 µM). Further ligand optimisation cycles generated new prospects for docking and biological assays.
Sickle cell disease (SCD) incurs vaso-occlusive episodes and organ damage, including nephropathy. Despite displaying characteristics of vascular dysfunction, SCD patients tend to present relatively ...lower systemic blood pressure (BP), via an unknown mechanism. We investigated associations between BP and renin-angiotensin-system (RAS) components in SCD and determined whether an inhibitor of angiotensin converting enzyme (ACE; often used to slow SCD glomerulopathy) further modulates BP and RAS components in a murine model of SCD.
BP was compared in human subjects and mice with/without SCD. Plasma angiotensin II, ACE and renin were measured by immunoassay. BP was reevaluated after treating mice with enalapril (25 mg/kg, 5x/week) for 5 weeks; plasma and organs were stored for angiotensin II and ACE activity measurement, and quantitative real-time PCR.
Diastolic BP and systolic BP were significantly lower in patients and mice with SCD, respectively, compared to controls. Reduced BP was associated with increased plasma renin and markers of kidney damage (mice) in SCD, as well as significantly decreased plasma ACE concentrations and ACE enzyme activity. As expected, enalapril administration lowered BP, plasma angiotensin II and organ ACE activity in control mice. In contrast, enalapril did not further reduce BP or organ ACE activity in SCD mice; however, plasma angiotensin II and renin levels were found to be significantly higher in enalapril-treated SCD mice than those of treated control mice.
Relative hypotension was confirmed in a murine model of SCD, in association with decreased ACE concentrations in both human and murine disease. Given that ACE inhibition has an accepted role in decreasing BP, further studies should investigate mechanisms by which ACE depletion, via both Ang II-dependent and alternative pathways, could contribute to reduce BP in SCD and understand how ACE inhibition confers Ang II-independent benefits on kidney function in SCD.
The present work aimed to evaluate the wood of fourteen genetic materials (nine species, between native and exotic, and five clones of Eucalyptus) cultivated under two post-planting irrigation ...regimes in the Brazilian semiarid region. For each genotype, six trees (11 years old) were selected and subjected to two initial irrigation regimes (up to 12 months and up to 36 months) after planting. Discs of different stem heights were taken: 0% (base), breast height (DBH), 20%, 40%, 60%, 80%, and 100%. Samples were extracted along the radial axis of each disk at three regions (the pith-adjacent region, intermediate region, and bark-adjacent region). Samples were subsequently saturated in water to determine their density via the water displacement method. The irrigation systems caused changes in the pattern of radial and longitudinal variation in the clones regarding the proportion of woody material in the stem. Among the native species, Angico stood out with a high density and little juvenile wood, and Pau d’arco, with a low density and a high rate of juvenile wood. Among the exotic species, Nim presented a high density and a low rate of juvenile wood, and Chichá presented a high rate of juvenile wood and a low density. In the Eucalyptus hybrids, VE38 stood out with a high density. Overall, there was an influence from irrigation management, observed with greater intensity in exotic species for Mahogany and Acacia and for the VE41 and AEC1528 clones of Eucalyptus. The values of basic densities in the trees varied from 0.35 to 0.85 g·cm−3. There was good adaptation of native and exotic species and clones to the planting area in the Semiarid region.

A retrospective cohort study was conducted using data collected from medical records of women who had breast cancer and underwent treatment between 2005 and 2015 at the Hospital da Fundação de Assistencial da Paraíba in Campina Grande, State of Paraiba, Brazil. Survival curves were estimated using the Kaplan-Meier method, Cox regression, and conditional decision tree.
Women with triple-negative molecular subtypes had a shorter survival time compared to women with positive hormone receptors. The addition of hormone therapy reduced the risk of a patient dying by 5.5%, and the risk of a HER2-positive patient dying was 34.5% lower compared to those who were negative for this gene. Patients undergoing hormone therapy had a median survival time of 4 753 days.
This paper shows a favorable scenario for the use of immunotherapy for patients with HER2 overexpression. Further studies could assess the effectiveness of immunotherapy in patients with other conditions, to favor the prognosis and better quality of life for the patient.
Excess of adipose tissue increases the concentration of proinflammatory cytokines, triggering a subclinical inflammatory condition. This inflammatory profile contributes to retina damage, which can ...lead to retinal dysfunction and reduced vision. Regularly practicing both aerobic and strength exercises is well known for promoting anti-inflammatory effects on different organs in the peripheral and central regions. However, the effects of combined physical exercise (CPE; strength + aerobic) on the inflammatory process in the retina tissue are not yet known. This study aimed to investigate the effects of CPE on the inflammatory profile of the retina in obese mice. Swiss mice were distributed into control, sedentary obese, and trained obese groups. The trained obese group was subjected to short-term CPE, 1 h/day, for 7 days. The CPE was composed of aerobic and strength exercises in the same exercise session. The strength exercise protocol consisted of 10 climbing series, with 12 ± 1 dynamic climbing movements at 70% of the maximum voluntary carrying capacity (MVCC), and the aerobic exercise protocol consisted of 30 min of treadmill running, with an intensity of 75% of the exhaust velocity. Subsequently, the retina was excised and analyzed by Western blot. Obese animals presented impairment on glucose homeostasis and elevated levels of proinflammatory proteins in the serum and retina; however, CPE was effective in reversing these parameters, independently of changes in body adiposity. Therefore, for the first time, we have shown that short-term CPE can be an important strategy to treat an inflammatory profile in the retina.

The determination of measurement reliability in weighing lysimeters via error analysis is essential for scientific research and irrigation management. The objective of this study was to evaluate four different weight measuring systems (MSs) applied to load cell weighing lysimeters and compare the results with the expected uncertainty values obtained from data provided by manufacturers. A weighing lysimeter with an area of 0.385 m2 and a volume of 0.289 m3 was used, installed on three load cells. In MS1, the load cells were connected to a junction box and the box to a weighing indicator module in a six-wire configuration. In MS2, a four-wire connection was used between the junction box and a datalogger, whereas in MS3, there was a six-wire connection. For MS4, the connection between the load cells and datalogger was direct. The uncertainties of the measurement systems were determined from the calibration results. MS1 presented the lowest measurement errors and uncertainties, resulting in performance superior to those of the other MSs. After MS1, the best performances were obtained by MS2 and MS3, and MS4 presented the worst performance. The effect of the signal measurement uncertainties and the excitation by the datalogger had the greatest effects on the overall uncertainty of the system compared with the influence of temperature on the load cells. The measurement system may be selected according to the technical data supplied by the manufacturer; however, periodic calibration of the effective measuring range is necessary to verify and compensate for systematic errors, which are accentuated during the operation time.
The objective of this study was to evaluate the presence of anti-Sarcocystis spp. specific IgG antibodies in serum samples from precolostral lambs to determine the occurrence of transplacental ...transmission of Sarcocystis spp. in sheep.
Blood samples were collected from 80 ewes and their respective lambs, immediately after lambing and before colostrum ingestion, respectively. The presence of anti-Sarcocystis spp. IgG was evaluated in serum samples using the indirect fluorescent antibody test (IFAT). Positive samples of the lambs were submitted to titration and IFAT to detect anti-T. gondii and anti-N. caninum specific IgG.
Anti-Sarcocystis spp. IgG was detected in 62.5% of the ewes (50/80) and in 4% of the lambs of the seropositive ewes (2/50). None of the lambs from seronegative ewes were positive. The final titers of the positive lambs were 80. No cross reaction was detected among the positive samples to anti-Sarcocystis spp., anti-N. caninum, and anti-T. gondii IgG. The detection of anti-Sarcocystis spp. antibodies in serum samples of lambs deprived of colostrum suggests transplacental transmission of infection. Thus, the vertical transmission may be an alternative route of infection of Sarcocystis spp. also in sheep. Further studies are warranted to confirm transplacental transmission in sheep and to explain the importance of this infection pathway.
