Discriminative Correlation Filters (DCF) have been shown to achieve impressive performance in visual object tracking. However, existing DCF-based trackers rely heavily on learning regularised ...appearance models from invariant image feature representations. To further improve the performance of DCF in accuracy and provide a parsimonious model from the attribute perspective, we propose to gauge the relevance of multi-channel features for the purpose of channel selection. This is achieved by assessing the information conveyed by the features of each channel as a group, using an adaptive group elastic net inducing independent sparsity and temporal smoothness on the DCF solution. The robustness and stability of the learned appearance model are significantly enhanced by the proposed method as the process of channel selection performs implicit spatial regularisation. We use the augmented Lagrangian method to optimise the discriminative filters efficiently. The experimental results obtained on a number of well-known benchmarking datasets demonstrate the effectiveness and stability of the proposed method. Spinal cord injury (SCI) is a severe neurological disease; however, few drugs have been proved to treat SCI effectively. Neuroinflammation is the major pathogenesis of SCI secondary injury and ...considered to be the therapeutic target of SCI. Salidroside (Sal) has been reported to exert anti‐inflammatory effects in airway, adipose and myocardial tissue; however, the role of Sal in SCI therapeutics has not been clarified. In this study, we showed that Sal could improve the functional recovery of spinal cord in rats as revealed by increased BBB locomotor rating scale, angle of incline, and decreased cavity of spinal cord injury and apoptosis of neurons in vivo. Immunofluorescence double staining of microglia marker and M1/M2 marker demonstrated that Sal could suppress M1 microglia polarization and activate M2 microglia polarization in vivo. To verify how Sal exerts its effects on microglia polarization and neuron protection, we performed the mechanism study in vitro in microglia cell line BV‐2 and neuron cell line PC12. The results showed that Sal prevents apoptosis of PC12 cells in coculture with LPS‐induced M1 BV‐2 microglia, also the inflammatory secretion phenotype of M1 BV‐2 microglia was suppressed by Sal, and further studies demonstrated that autophagic flux regulation through AMPK/mTOR pathway was involved in Sal regulated microglia polarization after SCI. Overall, our study illustrated that Sal could promote spinal cord injury functional recovery in rats, and the mechanism may relate to its microglia polarization modulation through AMPK‐/mTOR‐mediated autophagic flux stimulation.
Laser cladding is an advantaged repairing technology due to its low heat input and high flexibility. With preheating wire by resistance heat, laser hot wire cladding shows better process stability ...and higher deposition efficiency compared to laser cold wire/powder cladding. Multi-pass layer were cladded on the surface of martensite precipitation hardening stainless steel FV520B by fiber laser with ER410NiMo wire. Wire feed rate and preheat current were optimized to obtain stable wire transfer, which guaranteed good formation quality of single pass cladding. Response surface methodology (RSM) was used to optimize processing parameters and predict formation quality of multi-pass cladding. Laser power P, scanning speed Vs, wire feed rate Vf and overlap ratio η were selected as the input variables, while flatness ratio, dilution and incomplete fusion value as the responses. Optimal clad layer with flat surface, low dilution and no incomplete fusion was obtained by appropriately reducing Vf, and increasing P, Vs and η. No defect like pore or crack was found. The tensile strength and impact toughness of the clad layer is respectively 96% and 86% of those of the substrate. The clad layer showed nonuniform microstructure and was divided into quenched areas with coarse lath martensite and tempered areas with tempered martensite due to different thermal cycles in adjacent areas. The tempered areas showed similar hardness to the substrate.

Oxidative stress-related apoptosis and autophagy play crucial roles in the development of osteoarthritis (OA), a progressive cartilage degenerative disease with multifactorial etiologies. Here, we ...determined autophagic flux changes and apoptosis in human OA and tert-Butyl hydroperoxide (TBHP)-treated chondrocytes. In addition, we explored the potential protective effects of trehalose, a novel Mammalian Target of Rapamycin (mTOR)-independent autophagic inducer, in TBHP-treated mouse chondrocytes and a destabilized medial meniscus (DMM) mouse OA model. We found aberrant p62 accumulation and increased apoptosis in human OA cartilage and chondrocytes. Consistently, p62 and cleaved caspase-3 levels increased in mouse chondrocytes under oxidative stress. Furthermore, trehalose restored oxidative stress-induced autophagic flux disruption and targeted autophagy selectively by activating BCL2 interacting protein 3 (BNIP3) and Phosphoglycerate mutase family member 5 (PGAM5). Trehalose could ameliorate oxidative stress-mediated mitochondrial membrane potential collapse, ATP level decrease, dynamin-related protein 1 (drp-1) translocation into the mitochondria, and the upregulation of proteins involved in mitochondria and endoplasmic reticulum (ER) stress-related apoptosis pathway. In addition, trehalose suppressed the cleavage of caspase 3 and poly(ADP-ribose) polymerase (PARP) and prevented DNA damage under oxidative stress. However, the anti-apoptotic effects of trehalose in TBHP-treated chondrocytes were partially abolished by autophagic flux inhibitor chloroquine and BNIP3- siRNA. The protective effect of trehalose was also found in mouse OA model. Taken together, these results indicate that trehalose has anti-apoptotic effects through the suppression of oxidative stress-induced mitochondrial injury and ER stress which is dependent on the promotion of autophagic flux and the induction of selective autophagy. Thus, trehalose is a promising therapeutic agent for OA.
In Arabidopsis, two AUXIN RESPONSE FACTORS (ARF7 and ARF19) and several Aux/IAAs regulate auxin-induced lateral root (LR) formation. As direct targets of ARF7 and ARF19, LATERAL ORGAN BOUNDARIES ...DOMAIN 16 (LBD16), LBD29, and LBD18 have a biological function in the formation of lateral roots (LRs). However, the details of the functions of these three LBDs have remained unclear. Each single T-DNA insert mutant has been shown to have slightly fewer LRs than the wild type. We then created a triple mutant, which exhibited a dramatic defect in the LR formation. Our results show that the Ibd mutations can lead to impairment in auxininduced pericycle cell division and in the expression levels of some D-type cyclins (CYCDs). Simultaneously, PLETHORA (PLT) and PIN-FORMED (PIN), which have been well documented to promote cell mitotic activity and are required for auxin response effects, were down-regulated by these Ibd mutations. Our results so far indicate that CYCDs, PLT, and PINs are the main targets of the LBDs. We believe that these three LBDs are involved in cell cycle progression of the pericycle in response to auxin. Overexpression of any of these three LBD genes in the triple mutant was found incapable of completely replacing the other two LBDs. The phenotypes of Ibd29 mutants were not completely consistent with Ibdl6 or Ibd18 mutants. This indicates that LBD29 may play a distinctive role compared with LBD 16 or LBD 18 and LBDs might play partially independent roles during the formation of LRs.
Site-1 protease (S1P) is a Golgi-located protein that activates unique membrane-bound latent transcription factors, and it plays an indispensable role in endoplasmic reticulum stress, lipid ...metabolism, inflammatory response and lysosome function. A patient with S1P mutation exhibits severe skeletal dysplasia with kyphoscoliosis, dysmorphic facial features and pectus carinatum. However, whether S1P regulates bone remodeling by affecting osteoclastogenesis remains elusive. Here, we show that S1P is indeed a positive regulator of osteoclastogenesis. S1P ablation in mice led to significant osteosclerosis compared with wild-type littermates. Mechanistically, S1P showed upregulated during osteoclastogenesis and was identified as a direct target of miR-9-5p. S1P deletion in bone marrow monocytes (BMMs) inhibited ATF6 and SREBP2 maturation, which subsequently impeded CHOP/SREBP2-complex-induced LC3 expression and autophagy flux. Consistently, transfection of LC3 adenovirus evidently rescued osteoclastogenesis in S1P-deficient BMMs. We then identified the interaction regions between CHOP and SREBP2 by Co-immunoprecipitation (Co-IP) and molecular docking. Furthermore, S1P deletion or inhibitor efficaciously rescued ovariectomized (OVX)- and LPS-induced bone loss in vivo. Collectively, we showed that S1P regulates osteoclast differentiation in a LC3 dependent manner and so is a potential therapy target for osteoporosis.
In this study, a three-dimensional segmented coupled model for continuous casting billets under combined mold and final electromagnetic stirring (M-EMS, F-EMS) was developed. The model was verified ...by comparing carbon segregation in billets with and without EMS through plant experiments. The findings revealed that both M-EMS and F-EMS induce tangential flow in molten steel, impacting solidification and solute distribution processes within the billet. For M-EMS, with operating parameters of 250A-2Hz, the maximum tangential velocity (velocity projected onto the cross-section) was observed at the liquid phase's edge. For F-EMS, with operating parameters of 250A-6Hz, the maximum tangential velocity occurred at fl=0.7. Furthermore, F-EMS accelerated heat transfer in the liquid phase, reducing the central liquid fraction from 0.93 to 0.85. M-EMS intensified the washing effect of molten steel on the solidification front, resulting in the formation of negative segregation within the mold. F-EMS significantly improved the centerline segregation issue, reducing carbon segregation from 1.15 to 1.02. Experimental and simulation results, with and without EMS, were in good agreement, indicating that M+F-EMS leads to a more uniform solute distribution within the billet, with a pronounced improvement in centerline segregation.
Multi-attribute negotiation is essentially a multi-objective optimization (MOO) problem, where models of agent-based emotional persuasion (EP) can exhibit characteristics of anthropomorphism. This ...paper proposes a novel EP model by fusing the strategy of emotion-driven concession with the method of multi-objective optimization (EDC-MOO). Firstly, a comprehensive emotion model is designed to enhance the authenticity of the emotion. A novel concession strategy is then proposed to enable the concession to be dynamically tuned by the emotions of the agents. Finally, a new EP model is constructed by integrating emotion, historical transaction, persuasion behavior, and concession strategy under the framework of MOO. Comprehensive experiments on bilateral negotiation are conducted to illustrate and validate the effectiveness of EDC-MOO. These include an analysis of negotiations under five distinct persuasion styles, a comparison of EDC-MOO with a non-emotion-based MOO negotiation model and classic trade-off strategies, negotiations between emotion-driven and non-emotion-driven agents, and negotiations involving human participants. A detailed analysis of parameter sensitivity is also discussed. Experimental results show that the proposed EDC-MOO model can enhance the diversity of the negotiation process and the anthropomorphism of the bilateral agents, thereby improving the social welfare of both parties.
The quality of the bloom will be impacted by the non-metallic impurities in the molten steel in the tundish, which will reduce the plasticity and fatigue life of the steel. In this research, a ...mathematical model of a six-flow double-channel T-shaped induction heating tundish was established, the effects of induction heating conditions on the removal of inclusions in the tundish were investigated, and the impact of various inclusion particle sizes on the removal effect of inclusions under induction heating was explored. The results show that the Residence Time Distribution (RTD) curve produced through numerical simulation and physical simulation is in good agreement. The reduction of inclusion particles in the channel is made affordable by the dual-channel induction heating technique. As the diameter of inclusion particles increases from 10 μm to 50 μm, the probability of inclusion particles being removed from the channel gradually decreases from 70.9% to 56.1%.
Carbon segregation is the major and classical internal defect in the continuous casting process of carbon steel. Based on the combined electromagnetic stirring equipment for new billet in a steel ...plant, China, the influence of combined electromagnetic stirring (M-EMS + F-EMS) on the carbon segregation of 300 mm × 340 mm special-shaped billet was studied via numerical simulation and on-site industrialization tests. The Lorentz force and carbon solute distribution were simulated under different EMS parameters. The formation mechanism of the carbon segregation of medium carbon steel with different combined electromagnetic stirring processes was analyzed. The results show that: (1) with the combined action of "solute flushing" effect and gravity, the carbon concentration in the loose side of the medium carbon steel casting billet is gradually lower than the fixed side, while the carbon concentration on the fixed side gradually accumulates more; and (2) under the action of combined electromagnetic stirring, the segregation index of casting billet could be controlled to remain between 0.96-1.05 and shows an increasing change in solidification from the skin to the center. When the current and frequency of M-EMS are 250 A and 2.0 Hz and the F-EMS are 180 A and 8.0 Hz, the carbon segregation defects in the special-shaped (300 mm × 340 mm) casting billet can be significantly improved.
