In ticks, the digestion of blood occurs intracellularly and proteolytic digestion of hemoglobin takes place in a dedicated type of lysosome, the digest vesicle, followed by transfer of the heme ...moiety of hemoglobin to a specialized organelle that accumulates large heme aggregates, called hemosomes. In the present work, we studied the uptake of fluorescent metalloporphyrins, used as heme analogs, and amitraz, one of the most regularly used acaricides to control cattle tick infestations, by Rhipicephalus (Boophilus) microplus midgut cells. Both compounds were taken up by midgut cells in vitro and accumulated inside the hemosomes. Transport of both molecules was sensitive to cyclosporine A (CsA), a well-known inhibitor of ATP binding cassette (ABC) transporters. Rhodamine 123, a fluorescent probe that is also a recognized ABC substrate, was similarly directed to the hemosome in a CsA-sensitive manner. Using an antibody against conserved domain of PgP-1-type ABC transporter, we were able to immunolocalize PgP-1 in the digest vesicle membranes. Comparison between two R. microplus strains that were resistant and susceptible to amitraz revealed that the resistant strain detoxified both amitraz and Sn-Pp IX more efficiently than the susceptible strain, a process that was also sensitive to CsA. A transcript containing an ABC transporter signature exhibited 2.5-fold increased expression in the amitraz-resistant strain when compared with the susceptible strain. RNAi-induced down-regulation of this ABC transporter led to the accumulation of metalloporphyrin in the digestive vacuole, interrupting heme traffic to the hemosome. This evidence further confirms that this transcript codes for a heme transporter. This is the first report of heme transport in a blood-feeding organism. While the primary physiological function of the hemosome is to detoxify heme and attenuate its toxicity, we suggest that the use of this acaricide detoxification pathway by ticks may represent a new molecular mechanism of resistance to pesticides.
O milho (Zea mays L.), é uma cultura de grande importância para o mundo. Dentre os grandes produtores dessa comoditie, destaca-se os Estados Unidos, China e Brasil. Visto o alto valor comercial do ...milho, tecnologias vem sendo empregadas no processo de produção, como a utilização de bioestimulantes. Nesse sentido, esse estudo objetivou revisar o conhecimento atual sobre os efeitos dos bioestimulantes na cultura do milho, em especial, o extrato de algas e extrato pirolenhoso, destacando a resposta direta ou indireta do vegetal quando utilizado esses tipos de substâncias. Diversos estudos de pesquisa já enfatizam a relevância do potencial de algas para uma melhor eficiência na produção agrícola, sendo fortes influenciadores ativos no crescimento da planta, visto sua aptidão de produzir ou interagir com os fitohormônios da planta, proporcionando ainda, um melhor desenvolvimento do sistema, maior vigor e aumento da produtividade. Por sua vez, o extrato pirolenhoso é um produto natural obtido da queima da madeira e utilizado como condicionador do solo, indutor de enraizamento, repelente de insetos pragas e outras funções que diminuem a utilização de defensivos agrícola no sistema de cultivo, sendo um bioestimulante com forte tendência no mercado. Os bioestimulantes apresenta-se como um potencial insumo alternativo para o incremento no crescimento e desenvolvimento de plantas de milho, contudo, entende-se que novas pesquisas são necessárias, com o intuito de fornecer melhores informações de interesses agronômicos.
Mycobacterium leprae and Mycobacterium tuberculosis antimicrobial resistance has been followed with great concern during the last years, while the need for new drugs able to control leprosy and ...tuberculosis, mainly due to extensively drug-resistant tuberculosis (XDR-TB), is pressing. Our group recently showed that M. leprae is able to induce lipid body biogenesis and cholesterol accumulation in macrophages and Schwann cells, facilitating its viability and replication. Considering these previous results, we investigated the efficacies of two statins on the intracellular viability of mycobacteria within the macrophage, as well as the effect of atorvastatin on M. leprae infections in BALB/c mice. We observed that intracellular mycobacteria viability decreased markedly after incubation with both statins, but atorvastatin showed the best inhibitory effect when combined with rifampin. Using Shepard's model, we observed with atorvastatin an efficacy in controlling M. leprae and inflammatory infiltrate in the BALB/c footpad, in a serum cholesterol level-dependent way. We conclude that statins contribute to macrophage-bactericidal activity against Mycobacterium bovis, M. leprae, and M. tuberculosis. It is likely that the association of statins with the actual multidrug therapy effectively reduces mycobacterial viability and tissue lesion in leprosy and tuberculosis patients, although epidemiological studies are still needed for confirmation.

Still, for this compound, molecular dynamics (MD) simulations were performed to determine the stability of the complex with tyrosinase, generating RMSD, RMSF, and interaction plots. Additionally, docking studies were performed to elucidate the binding pose at the tyrosinase, suggesting that the hydroxyl group of coumarin derivative performs coordinate bonds (bidentate) with the copper(II) ions at distances ranging from 2.09 to 2.61 Å. O rótulo dos alimentos são elementos fundamentais de comunicação entre as empresas que produzem alimentos e os consumidores, por isso, é importante que as informações que constam nesses rótulos sejam ...expressas de forma simples e com fácil entendimento. Considerando que um rótulo apresente mais clareza e legibilidade nas informações nutricionais dos produtos alimentícios foram estabelecidas a RDC nº 429 e a Instrução Normativa nº 75 em 8 de outubro de 2020. Nas novas normas regulamentadoras, dentre as modificações da rotulagem incluem-se, a utilização de rotulagem nutricional frontal nas embalagens, fornecendo informações acerca dos nutrientes que são considerados relevantes para a saúde do consumidor e a tabela nutricional que deve apresentar as informações sobre determinados nutrientes. Essas modificações no rótulo nutricional, contribuem para que o consumidor possa realizar escolhas alimentares mais adequadas e saudáveis.
Resumo Fundsamento As sementes de Moringa oleifera , que são utilizadas para clarificação de água, contêm uma lectina chamada WSMoL que tem mostrado atividade antibacteriana e imunomoduladora in ...vitro . Devido ao seu valor nutritivo e potencial terapêutico, as folhas e as sementes dessa árvore são consumidas em algumas comunidades. Algumas lectinas de plantas não são tóxicas para mamíferos, mas tem sido relatado que outras são prejudiciais quando ingeridas ou administradas por outros meios. Objetivo Como um dos passos necessários para determinar a segurança de WSMoL, nós avaliamos os possíveis efeitos cardiotóxicos desta proteína purificada. Métodos Durante 21 dias consecutivos, a WSMoL foi administrada a camundongos por gavagem. Foram investigadas as funções eletrofisiológicas, mecânicas e metabólicas in vivo e ex vivo por meio de registros eletrocardiográficos, ressonância magnética nuclear e respirometria de alta resolução. Resultados O tratamento com WSMoL não induziu alterações nos níveis de glicose no sangue ou peso corporal em comparação com o grupo controle. Adicionalmente, as relações peso cardíaco/peso corporal e peso cardíaco/comprimento tibial estavam semelhantes em ambos os grupos. A ingestão de lectina também não modificou a tolerância à glicose ou resistência à insulina. Não foram observadas alterações nos parâmetros eletrocardiográficos ou na duração do potencial de ação cardíaco. Os corações dos camundongos dos grupos controle e WSMoL mostraram função ventricular esquerda preservada. Além disso, a WSMoL não induziu alterações na função mitocondrial (em todos os casos, p > 0,05). Conclusões A administração de WSMoL demonstrou ter um perfil de segurança cardíaca. Estes resultados contribuem à avaliação de segurança do uso de sementes de M. oleifera para tratar água, visto que essa lectina está presente na preparação empregada por algumas populações com esse fim. The protozoa, Trypanosoma cruzi (etiological agent of Chagas diseases – also named American trypanosomiasis) and T. brucei (causative agent of human African trypanosomiasis – HAT), negatively impact ...public health, being endemic in several countries and leading to thousands of deaths per year. Moreover, the pharmacological treatment of diseases has several limitations, such as parasitic resistance and several side effects in patients, which decrease therapeutic adherence. Two cysteine proteases, cruzain (CRZ) from T. cruzi and a cathepsin L-like enzyme ( Tbr CATL) from T. brucei , are considered promising targets of these protozoa since they are responsible for many key biological processes in their life cycles. Coumarin analogs have been reported in diverse studies targeting the development of trypanocidal agents, and have shown activity against different evolutionary forms of these parasites. In this study, we report a virtual fragment-based drug design (vFBDD) approach to develop coumarin-based analogs capable of inhibiting these main cysteine proteases. Also, their experimental validation involved enzymatic inhibition, in vitro infected-cell-based, and antitrypomastigote assays. One compound, FN-27, a coumarin-thiosemicarbazone analog, inhibited both CRZ (IC 50 : 14.4 μM ± 0.02) and Tbr CATL (IC 50 : 2.0 μM ± 0.6), and exhibited trypanocidal activity against T. cruzi amastigote-infected cells (EC 50 : 5.5 μM), but had no effect on T. brucei trypomastigotes. These results suggest that FN-27 probably exerts its mechanism of action against the T. cruzi parasite via inhibition of CRZ, although other targets could be involved. In parallel, FN-10, a coumarin-chalcone analog, was active against T. brucei trypomastigotes (EC 50 : 4.8 μM ± 0.15) but it did not inhibit CRZ or Tbr CATL. Accordingly, FN-10 may exhibit its effects via a different macromolecular target(s) in each parasite. For FN-27, molecular dynamics (MD) simulations were performed to gain insights into the stability of its final complexes with both proteases within 200 ns, and the parameters RMSD, RMSF, R g , and SASA were determined, through which it was verified that FN-27 adopts various binding modes in the catalytic site for both proteases, corroborating our experimental data. MM/PBSA calculations suggested that the most relevant stabilizing interactions for the complex formation were van der Waals interactions. Also, it was noted that the binding energy (Δ E MM ) for Tbr CATL is more favorable than for CRZ, again corroborating the enzymatic inhibition assays. Overall, the data generated will be useful in developing novel natural-product-based inhibitors targeting both cysteine proteases.
In this study, an extensive analysis of microsatellite markers (Single Tandem Repeats-STRs) in Penaeus vannamei was conducted at an advanced level. The markers were thoroughly examined, ...characterized, and specific markers located within coding regions were identified. Out of a total of 306 STRs, 117 were classified as perfect markers based on their single repeat motif. Among these perfect markers, 62 were found to be associated with predicted coding genes (mRNA), which were involved in various functions such as binding, catalytic activity, ATP-dependent activity, transcription, structural and molecular regulation. To validate the accuracy of the findings, a sample of nine markers was subjected to in vitro testing, which confirmed the presence of polymorphisms within the population. These results suggest the existence of different protein isoforms within the population, indicating the potential of these markers for application in both population and phenotype-genotype association studies. This innovative approach opens up new possibilities for investigating the impact of genomic plasticity in populations of P. vannamei.
O queijo de coalho é considerado um produto típico da região Nordeste do Brasil e sua representa uma atividade econômica de importante relevância para os agricultores de base familiar. Embora, seja ...de ampla popularidade e influencie significativamente a economia, este tipo de queijo tem sido associado a um produto com baixa qualidade microbiológica, pois seu processo de produção pode apresentar desde a utilização de matéria-prima em condições inadequadas até a ausência da realização das Boas Práticas de Fabricação (BPF). Por isso, o objetivo do estudo foi identificar evidências disponíveis na literatura sobre a qualidade microbiológica de queijo de coalho. Para tanto, foi realizada uma revisão integrativa de literatura com dimensão temporal entre 2016 e 2020, utilizando a base de dados Periódicos CAPES e os descritores: queijo de coalho, coalho, microbiologia, qualidade e contaminação. Após a localização e refinamento das publicações científicas, foram obtidos cinco artigos que versavam sobre a questão em estudo. Verificou-se através dos achados, que os queijos de coalho avaliados eram provenientes de feira-livre, fábricas e vendedores informais e todos os estudos indicaram a contaminação do derivado lácteo por microrganismos patogênicos. Logo, os queijos avaliados apresentavam-se em não conformidade com as exigências da legislação acerca do perfil microbiológico do referido laticínio, o que os tornam inaptos para o consumo.
