The Miocene is a relatively recent epoch of the Earth's history with warmer climate than today, particularly during the middle Miocene Climatic Optimum (MMCO, approximately 17–15Ma). Although the ...cause of the warming is probably not only attributable to CO2, but also to changes in orography and configuration of ocean gateways, this time interval represents an ideal case study to test the ability of climate models to simulate warm climates comparable to those that the Earth may experience in the near future. However, even with higher than present-day CO2 concentrations, the MMCO warming inferred from terrestrial proxy data has been difficult to reproduce in climate models.
Since fossil flora do not provide direct information on climate, but on flora and vegetation, climate model results are generally compared to climate reconstructions obtained from the fossil flora. In this study, we apply an alternative method by simulating palaeovegetation from the outputs of the climate model, using a dynamic vegetation model. Model vegetation reconstruction can then be compared to the vegetation cover indicated by the fossil flora record at the various localities, provided that a common classification of plant functional types (PFTs) is used for the data and the model. Here, we reconstruct the vegetation of the middle Miocene with the global dynamic vegetation model CARAIB, using the climatologies derived from five atmospheric general circulation models. The reliability of the simulations is examined on a presence/absence basis of PFTs by comparison of vegetation reconstructions to palaeoflora data recorded in the Northern Hemisphere and the Tropics.
This comparison provides an overall agreement around 60% between model and data, when all sites and tree types are considered. Three model simulations out of five show to be better at predicting the absence than the presence. The presence of warm-temperate mixed forests in the middle latitudes, dominated by broadleaved deciduous warm temperate and subtropical trees is generally well reproduced in CARAIB simulations. However, poor agreement is obtained for the presence of tropical PFTs out of the Tropics and for warm PFTs at latitudes northward of 50°N, where climate models remain too cold to produce assemblages of trees consistent with the data. Nevertheless, the model–data comparison performed here highlights several mismatches that could result not only from missing feedbacks in the climate simulations, but also from the data. The results of the likelihood analysis on presence/absence of PFTs illustrate the uncertainties in the PFT classification of the Neogene floral records. The coexistence of some PFTs in the palaeovegetation data is impossible to reproduce in the vegetation model simulations because of the climatic definition of the modern PFTs. This result indicates either a bias in the identification of modern analogues for fossil plant taxa, or a possible evolution of environmental requirements of certain plants.

Simulating gross primary productivity (GPP) of terrestrial ecosystems has been a major challenge in quantifying the global carbon cycle. Many different light use efficiency (LUE) models have been developed recently, but our understanding of the relative merits of different models remains limited. Using CO2 flux measurements from multiple eddy covariance sites, we here compared and assessed major algorithms and performance of seven LUE models (CASA, CFix, CFlux, EC-LUE, MODIS, VPM and VPRM). Comparison between simulated GPP and estimated GPP from flux measurements showed that model performance differed substantially among ecosystem types. In general, most models performed better in capturing the temporal changes and magnitude of GPP in deciduous broadleaf forests and mixed forests than in evergreen broadleaf forests and shrublands. Six of the seven LUE models significantly underestimated GPP during cloudy days because the impacts of diffuse radiation on light use efficiency were ignored in the models. CFlux and EC-LUE exhibited the lowest root mean square error among all models at 80% and 75% of the sites, respectively. Moreover, these two models showed better performance than others in simulating interannual variability of GPP. Two pairwise comparisons revealed that the seven models differed substantially in algorithms describing the environmental regulations, particularly water stress, on GPP. This analysis highlights the need to improve representation of the impacts of diffuse radiation and water stress in the LUE models.
The purpose of this study is to evaluate the eight ISIMIP2a biome models against independent estimates of long-term net carbon fluxes (i.e. Net Biome Productivity, NBP) over terrestrial ecosystems ...for the recent four decades (1971-2010). We evaluate modeled global NBP against 1) the updated global residual land sink (RLS) plus land use emissions (ELUC) from the Global Carbon Project (GCP), presented as R + L in this study by Le Quéré et al (2015), and 2) the land CO2 fluxes from two atmospheric inversion systems: Jena CarboScope s81_v3.8 and CAMS v15r2, referred to as FJena and FCAMS respectively. The model ensemble-mean NBP (that includes seven models with land-use change) is higher than but within the uncertainty of R + L, while the simulated positive NBP trend over the last 30 yr is lower than that from R + L and from the two inversion systems. ISIMIP2a biome models well capture the interannual variation of global net terrestrial ecosystem carbon fluxes. Tropical NBP represents 31 ± 17% of global total NBP during the past decades, and the year-to-year variation of tropical NBP contributes most of the interannual variation of global NBP. According to the models, increasing Net Primary Productivity (NPP) was the main cause for the generally increasing NBP. Significant global NBP anomalies from the long-term mean between the two phases of El Niño Southern Oscillation (ENSO) events are simulated by all models (p < 0.05), which is consistent with the R + L estimate (p = 0.06), also mainly attributed to NPP anomalies, rather than to changes in heterotrophic respiration (Rh). The global NPP and NBP anomalies during ENSO events are dominated by their anomalies in tropical regions impacted by tropical climate variability. Multiple regressions between R + L, FJena and FCAMS interannual variations and tropical climate variations reveal a significant negative response of global net terrestrial ecosystem carbon fluxes to tropical mean annual temperature variation, and a non-significant response to tropical annual precipitation variation. According to the models, tropical precipitation is a more important driver, suggesting that some models do not capture the roles of precipitation and temperature changes adequately.
Extraction of mouse spinal motor neurons from transgenic mouse embryos recapitulating some aspects of neurodegenerative diseases like amyotrophic lateral sclerosis has met with limited success. ...Furthermore, extraction and long-term culture of adult mouse spinal motor neurons and glia remain also challenging. We present here a protocol designed to extract and purify high yields of motor neurons and glia from individual spinal cords collected on embryos and adult (5-month-old) normal or transgenic mice. This method is based on mild digestion of tissue followed by gradient density separation allowing to obtain two millions motor neurons over 92% pure from one E14.5 single embryo and more than 30,000 from an adult mouse. These cells can be cultured more than 14 days in vitro at a density of 100,000 cells/cm(2) to maintain optimal viability. Functional astrocytes and microglia and small gamma motor neurons can be purified at the same time. This protocol will be a powerful and reliable method to obtain motor neurons and glia to better understand mechanisms underlying spinal cord diseases.
Atmospheric carbon dioxide concentrations are believed to drive climate changes from glacial to interglacial modes, although geological and astronomical mechanisms have been invoked as ultimate ...causes. Additionally, it is unclear whether the changes between cold and warm modes should be regarded as a global phenomenon, affecting tropical and high-latitude temperatures alike, or if they are better described as an expansion and contraction of the latitudinal climate zones, keeping equatorial temperatures approximately constant. Here we present a reconstruction of tropical sea surface temperatures throughout the Phanerozoic eon (the past approximately 550 Myr) from our database of oxygen isotopes in calcite and aragonite shells. The data indicate large oscillations of tropical sea surface temperatures in phase with the cold-warm cycles, thus favouring the idea of climate variability as a global phenomenon. But our data conflict with a temperature reconstruction using an energy balance model that is forced by reconstructed atmospheric carbon dioxide concentrations. The results can be reconciled if atmospheric carbon dioxide concentrations were not the principal driver of climate variability on geological timescales for at least one-third of the Phanerozoic eon, or if the reconstructed carbon dioxide concentrations are not reliable.
Autosomal recessive spastic ataxia of Charlevoix-Saguenay (ARSACS) is an early-onset neurodegenerative disease mainly characterized by spasticity in the lower limbs and poor muscle control. The ...disease is caused by mutations in the SACS gene leading in most cases to a loss of function of the sacsin protein, which is highly expressed in motor neurons and Purkinje cells. To investigate the impact of the mutated sacsin protein in these cells in vitro, induced pluripotent stem cell- (iPSC-) derived motor neurons and iPSC-derived Purkinje cells were generated from three ARSACS patients. Both types of iPSC-derived neurons expressed the characteristic neuronal markers β3-tubulin, neurofilaments M and H, as well as specific markers like Islet-1 for motor neurons, and parvalbumin or calbindin for Purkinje cells. Compared to controls, iPSC-derived mutated SACS neurons expressed lower amounts of sacsin. In addition, characteristic neurofilament aggregates were detected along the neurites of both iPSC-derived neurons. These results indicate that it is possible to recapitulate in vitro, at least in part, the ARSACS pathological signature in vitro using patient-derived motor neurons and Purkinje cells differentiated from iPSCs. Such an in vitro personalized model of the disease could be useful for the screening of new drugs for the treatment of ARSACS.
Estimating the future state of air quality associated with transport policies and infrastructure investments is key to the development of meaningful transportation and planning decisions. This paper ...describes the design of an integrated transportation and air quality modelling framework capable of simulating traffic emissions and air pollution at a refined spatio-temporal scale. For this purpose, emissions of Nitrogen Oxides (NOx) were estimated in the Greater Montreal Region at the level of individual trips and vehicles. In turn, hourly Nitrogen Dioxide (NO2) concentrations were simulated across different seasons and validated against observations. Our validation results reveal a reasonable performance of the modelling chain. The modelling system was used to evaluate the impact of an extensive regional transit improvement strategy revealing reductions in NO2 concentrations across the territory by about 3.6% compared to the base case in addition to a decrease in the frequency and severity of NO2 hot spots. This is associated with a reduction in total NOx emissions of 1.9% compared to the base case; some roads experienced reductions by more than half. Finally, a methodology for assessing individuals’ daily exposure is developed (by tracking activity locations and trajectories) and we observed a reduction of 20.8% in daily exposures compared to the base case. The large difference between reductions in the mean NO2 concentration across the study domain and the mean NO2 exposure across the sample population results from the fact that NO2 concentrations dropped largely in the areas which attract the most individuals. This exercise illustrates that evaluating the air quality impacts of transportation scenarios by solely quantifying reductions in air pollution concentrations across the study domain would lead to an underestimation of the potential health gains.

The past decade has seen great advances in unraveling the biological basis of hereditary ataxias. Molecular studies of spinocerebellar ataxias (SCA) have extended our understanding of dominant ...ataxias. Causative genes have been identified for a few autosomal recessive ataxias: Friedreich's ataxia, ataxia with vitamin E deficiency, ataxia telangiectasia, recessive spastic ataxia of Charlevoix-Saguenay and ataxia with oculomotor apraxia type 1 (refs. 6,7) and type 2 (ref. 8). Nonetheless, genes remain unidentified for most recessive ataxias. Additionally, pure cerebellar ataxias, which represent up to 20% of all ataxias, remain poorly studied with only two causative dominant genes being described: CACNA1A (ref. 9) and SPTBN2 (ref. 10). Here, we report a newly discovered form of recessive ataxia in a French-Canadian cohort and show that SYNE1 mutations are causative in all of our kindreds, making SYNE1 the first identified gene responsible for a recessively inherited pure cerebellar ataxia.

Five independent domains reflecting different aspects of the craniofacial structure were examined. We confirmed heritability of sella-nasion-subspinale (38%, P = 0.002), saddle angle (55%, P < 0.0001), mandibular length (24%, P = 0.02) and lower facial height (33%, P = 0.006) previously measured by cephalometry. In addition, the current study added new insights by demonstrating significant heritability of mandibular width (30%, P = 0.005), maxillary width (47%, P < 0.0001), distance from the hyoid bone to the retropogonion (36%, P = 0.0018) and size of the oropharyngeal space (31%, P = 0.004). Finally, our data indicate that heritability of the craniofacial structures is similar in normal patients and those with apnea.
The data support our a priori hypothesis that the craniofacial structures that have been associated with obstructive sleep apnea (OSA) are heritable. We have demonstrated heritability for several intermediate craniofacial phenotypes for OSA. Thus, we believe that future studies should be able to identify genes associated with these intermediate craniofacial phenotypes.
