However,at present, the overall number and quality of digital talents delivered by colleges and universities still have some gaps compared with the high standards of enterprises. There are problems in the talent training mode, such as lagging training system, insufficient innovation in teaching and assessment methods, and the need to strengthen teaching practice. Combining the practical needs of digital business environment for talents, this paper puts forward relevant suggestions for applied universities on how to cultivate digital talents under the big background of digitalization from the perspectives of concept updating, framework construction, diversified governance and so on, in order to accelerate the pace of reform and adjustment and meet the urgent needs of society and enterprises for digital talents in the development trend of the times.
As one of the significant life forms found in biological soil crusts in arid and semiarid environment, actinomycetes are not only important sources of chemical diversity in natural products that are ...of potential clinical value but also vital participators in the material cycle and energy flows in the ecological systems. Taking cyanobacteria crusts and moss crusts that are widely distributed in the southeastern Tengger Desert as research objects, metagenome sequencing was performed to compare and analyze the distribution characteristics, components and potential metabolic functions of the actinomycete population in two biological soil crusts. The results showed that the actinomycete community structure of the soil microbiome was mainly composed of the genera Geodermatophilus, Rubrobacter, Nocardioides, Actinoplanes, Blastococcus, Streptomyces, Modestobacter, Saccharothrix, Solirubrobacter, Pseudonocardia, Micromonospora, Conexibacter, Marmoricola, Microlunatus and Frankia. There were differences in the distribution of genera between the two biological soil crust types. The participation of actinomycetes in amino sugar and nucleotide sugar metabolism, glycolysis/gluconeogenesis, carbon fixation pathways of prokaryotes, butanoate metabolism, propanoate metabolism, alanine metabolism, aspartate/glutamate metabolism, methane metabolism, 2-oxocarboxylic acid metabolism, peptidoglycan biosynthesis, starch/sucrose metabolism, and valine/leucine/isoleucine degradation was significantly higher in moss crusts than that in cyanobacteria crusts. However, the contributions of Geodermatophilus and Rubrobacter to the top ten most abundant metabolic functions were obviously lower in moss crusts than that in cyanobacteria crusts. In contrast, the relative contributions of Nocardioides, Blastococcus, Modestobacter, Actinoplanes, Streptomyces, Pseudonocardia, and Saccharothrix to these functions was of great significance in moss crusts. The results provide the scientific data for a comprehensive and in-depth understanding of the diversity of actinomycete resources and potential functions in algae and moss crusts in the southeastern Tengger Desert and for understanding the ecological functions of actinomycetes in different types of biological soil crusts.
Abstract
Immune-related adverse events (irAEs), caused by anti-PD-1/PD-L1 antibodies, can lead to fulminant and even fatal consequences and thus require early detection and aggressive management. ...However, a comprehensive approach to identify biomarkers of irAE is lacking. Here, we utilize a strategy that combines pharmacovigilance data and omics data, and evaluate associations between multi-omics factors and irAE reporting odds ratio across different cancer types. We identify a bivariate regression model of LCP1 and ADPGK that can accurately predict irAE. We further validate LCP1 and ADPGK as biomarkers in an independent patient-level cohort. Our approach provides a method for identifying potential biomarkers of irAE in cancer immunotherapy using both pharmacovigilance data and multi-omics data.
Metabolic interaction between cancer-associated fibroblasts (CAFs) and colorectal cancer (CRC) cells plays a major role in CRC progression. However, little is known about lipid alternations in CAFs ...and how these metabolic reprogramming affect CRC cells metastasis. Here, we uncover CAFs conditioned medium (CM) promote the migration of CRC cells compared with normal fibroblasts CM. CAFs undergo a lipidomic reprogramming, and accumulate more fatty acids and phospholipids. CAFs CM after protein deprivation still increase the CRC cells migration, which suggests small molecular metabolites in CAFs CM are responsible for CRC cells migration. Then, we confirm that CRC cells take up the lipids metabolites that are secreted from CAFs. Fatty acids synthase (FASN), a crucial enzyme in fatty acids synthesis, is significantly increased in CAFs. CAF-induced CRC cell migration is abolished by knockdown of FASN by siRNA or reducing the uptake of fatty acids by CRC cells by sulfo-N-succinimidyloleate sodium in vitro and CD36 monoclonal antibody in vivo. To conclude, our results provide a new insight into the mechanism of CRC metastasis and suggest FASN of CAFs or CD36 of CRC cells may be potential targets for anti-metastasis treatment in the future.
Microbial necromass carbon (MNC) is key to soil organic carbon (SOC) storage. However, mechanisms regulating MNC accumulation on large scales are poorly understood. Here we provide the first batch of ...regional-scale MNC data based on amino sugars for the Qinghai-Tibet Plateau alpine grasslands. We show that Qinghai-Tibet grasslands have similar microbial biomass carbon (MBC) but lower MNC concentrations in SOC than Mongolian and other grasslands. The low contribution of MNC to SOC is mainly attributed to high aridity and low net primary productivity of the Qinghai-Tibet grasslands. Our findings highlight climatic and plant influences on MNC accumulation at regional scales.

Achieving moldable energy storage devices will make it possible to power wearable electronics by adapting to body contours for wear comfort. Herein, we present a new approach to create hierarchically ...structured electrodes that enable supercapacitors to retain their capacity under mechanical deformation. The electrodes are made by first growing vertical graphene nanosheets (VGNs) and then depositing manganese oxide (MnO2) on ductile nickel wires. Two such electrodes are made into a moldable supercapacitor using a solid-state electrolyte containing carboxymethylcellulose and sodium sulfate. This foldable supercapacitor achieves a high areal capacitance up to 56 mF cm−2, areal energy density of 7.7 μWh cm−2, and areal power density of 5 mWh cm−2. These exceptional properties originate from the synergy between VGNs and MnO2, where the highly porous VGNs serve two critical functions: a mechanically robust platform of large surface area allowing high mass loading deposition of pseudocapacitive MnO2 and an interconnected conductive network for efficient electron/ion transport. Fiber-shaped supercapacitors made from these electrodes can be molded into different shapes by bending and twisting with little performance loss. The promising results presented in this study provide a new route for fabricating high-performance moldable energy storage devices for wearable electronics and wireless electronic skins.

The aim of this study was to explore the regulatory mechanism of circRNA_100876/ microRNA‐136 (miR‐136) axis in the development and progression of osteosarcoma cancer. Quantitative real‐time ...polymerase chain reaction (RT‐qPCR) was used to evaluate the expression levels of circRNA_100876 and miR‐136 in osteosarcoma cancer samples and the adjacent nontumor tissues. Then, cell proliferation, cell cycle, apoptosis, and migration of circRNA_100876‐knocked down cells were analyzed by in vitro and in vivo experiments, using cell counting kit‐8 (CCK‐8), flow cytometry, and transwell and tumorigenesis assays. The expression of circRNA_100876 was found to be significantly upregulated in osteosarcoma, and was closely correlated with the tumor size and tumor differentiation degree. In addition, the knockdown of circRNA_100876 could significantly inhibit the tumor growth, both in vitro and in vivo. Flow cytometry and Western blot analysis results showed that the downregulation of circRNA_100876 inhibited osteosarcoma cells proliferation via promoting apoptosis and arresting more cells in the G2/M stage, as suggested by the expression of apoptosis and cell cycle pathway‐related proteins, which changed consistently. Furthermore, the level of miR‐136 was negatively correlated with the expression of circRNA_100876, and miR‐136 inhibitors were able to reverse the suppression of cell proliferation induced by silencing circRNA_100876. Our study demonstrates that the dysregulation of circRNA_100876 could induce apoptosis and arrest the cell cycle at G2/M stage, followed by suppression of cell proliferation in osteosarcoma, while silencing miR‐136 could restore the cell growth. Therefore, circRNA_100876 might serve as a promising biomarker and treatment target for osteosarcoma.

Background BCMA-specific chimeric antigen receptor-T cells (CAR-Ts) have exhibited remarkable efficacy in refractory or relapsed multiple myeloma (RRMM); however, primary resistance and relapse exist ...with single-target immunotherapy. Bispecific CARs are proposed to mitigate these limitations. Methods We constructed a humanized bispecific BM38 CAR targeting BCMA and CD38 and tested the antimyeloma activity of BM38 CAR-Ts in vitro and in vivo. Twenty-three patients with RRMM received infusions of BM38 CAR-Ts in a phase I trial. Results BM38 CAR-Ts showed stronger in vitro cytotoxicity to heterogeneous MM cells than did T cells expressing an individual BCMA or CD38 CAR. BM38 CAR-Ts also exhibited potent antimyeloma activity in xenograft mouse models. In the phase I trial, cytokine release syndrome occurred in 20 patients (87%) and was mostly grade 1-2 (65%). Neurotoxicity was not observed. Hematologic toxicities were common, including neutropenia in 96% of the patients, leukopenia in 87%, anemia in 43% and thrombocytopenia in 61%. At a median follow-up of 9.0 months (range 0.5 to 18.5), 20 patients (87%) attained a clinical response and minimal residual disease-negativity (less than or equai to 10.sup.-4 nucleated cells), with 12 (52%) achieving a stringent complete response. Extramedullary plasmacytoma was eliminated completely in 56% and partially in 33% and of 9 patients. The median progression-free survival was 17.2 months. Two relapsed patients maintained BCMA and CD38 expression on MM cells. Notably, BM38 CAR-Ts cells were detectable in 77.8% of evaluable patients at 9 months and 62.2% at 12 months. Conclusion Bispecific BM38 CAR-Ts were feasible, safe and significantly effective in patient with RRMM. Trial registration: Chictr.org.cn ChiCTR1800018143.
