Mycobacterium leprae and Mycobacterium tuberculosis antimicrobial resistance has been followed with great concern during the last years, while the need for new drugs able to control leprosy and ...tuberculosis, mainly due to extensively drug-resistant tuberculosis (XDR-TB), is pressing. Our group recently showed that M. leprae is able to induce lipid body biogenesis and cholesterol accumulation in macrophages and Schwann cells, facilitating its viability and replication. Considering these previous results, we investigated the efficacies of two statins on the intracellular viability of mycobacteria within the macrophage, as well as the effect of atorvastatin on M. leprae infections in BALB/c mice. We observed that intracellular mycobacteria viability decreased markedly after incubation with both statins, but atorvastatin showed the best inhibitory effect when combined with rifampin. Using Shepard's model, we observed with atorvastatin an efficacy in controlling M. leprae and inflammatory infiltrate in the BALB/c footpad, in a serum cholesterol level-dependent way. We conclude that statins contribute to macrophage-bactericidal activity against Mycobacterium bovis, M. leprae, and M. tuberculosis. It is likely that the association of statins with the actual multidrug therapy effectively reduces mycobacterial viability and tissue lesion in leprosy and tuberculosis patients, although epidemiological studies are still needed for confirmation.

Um experimento foi conduzido com pintos de corte macho para o estudo dos efeitos dos níveis de 20 e 23% de PB combinados com 0, 50, 100, 150, 200, 250 mEq/kg de balanço dietético eletrolítico (BDE) ...sobre a atividade da alfa-amilase pancreática de frangos de corte de 1 a 21 dias de idade. O delineamento utilizado foi o inteiramente casualizado. Dietas e água foram fornecidas ad libitum. Aos 1, 7, 14 e 21 dias, três aves de cada tratamento foram sacrificadas por deslocamento cervical para remoção do pâncreas, o qual foi removido, homogeneizado, congelado em nitrogênio líquido e liofilizado. Uma alíquota de cada amostra foi solubilizada em água deionizada e centrifugada a 7500 x g por 3 minutos a 4ºC, para determinação da atividade da alfa-amilase no sobrenadante. Aves alimentadas com 20% de PB apresentaram atividades específicas (U/mg de proteína) superiores à daquelas que receberam 23%, exceto para os níveis de BDE de 100 a 200 mEq/kg, aos 14 dias. Observou-se tendência de aumento da atividade específica dos 7 aos 14 dias e redução dos 14 aos 21 dias. Para maior atividade específica de alfa-amilase, recomendam-se dietas com 20% de PB e 200 mEq/kg de BDE na fase pré-inicial e dietas com 20% de PB e 135 a 250 mEq/kg de BDE para frangos de corte dos 8 aos 21 dias de idade.
An experiment was conducted with male broiler chicks to study the effects of 20 and 23% of crude protein (CP) combined with 0, 50, 100, 150, 200, 250 mEq/kg of dietary electrolytic balance (DEB) on the pancreatic alpha-amylase activity from 1 to 21 days. A completely randomized factorial design was used. Feed and water were supplied ad libitum. At days 1, 7, 14 and 21, three birds from each treatment were sacrificed by cervical dislocation, pancreas were removed, homogeneized and frozen in liquid nitrogen which were then freeze-dried. An aliquot of each sample was solubilized with deionized water and centrifuged at 7500 x g for three minutes at 4ºC. Activities of the pancreatic alpha-amylase in the supernatants were determined. Chicks fed 20% of CP showed specific activities (U/mg protein) higher than those fed 23%, except for DEB levels from 100 to 200 mEq/kg at 14 days. A tendency to increase specific activity was observed from 7 to 14 days and to reduce from 14 to 21 days. The highest specific activity of pancreatic alpha-amylase was observed in diets with 20% CP and 200 mEq/kg of DEB from 1 to 7 days, diets with 20% of CP and 135 to 250 mEq/kg of DEB from 8 to 21 days of age.
Foram investigados os efeitos nutricionais de dois níveis de ácido L-glutâmico (L-Glu) combinados com quatro níveis de vitamina K (Vit K) sobre a atividade de alfa-amilase no quimo e pâncreas de aves ...de corte. Frangos de corte machos de um dia foram criados em baterias aquecidas e alimentados, à vontade, com dietas contendo todos L-aminoácidos essenciais, minerais e vitaminas (exceto Vit K) até os 14 dias de idade. O experimento foi realizado em esquema fatorial, em delineamento inteiramente casualizado 2x4, com quatro repetições de oito aves cada. A dieta básica foi suplementada com 6,25 e 12,5% de L-Glu combinados com 0,02; 0,2; 20,0 e 200,0 mg de Vit K/kg de ração. Efeitos significativos de L-Glu e Vit K foram observados no quimo. A atividade específica máxima (1.005,78±245,25 UI/mg de proteína) foi observada nas aves alimentadas com 6,25% de L-Glu e 20,0 mg de Vit K. Houve redução da atividade com a suplementação de 12,5% de L-Glu. No pâncreas, não houve interação entre L-Glu e Vit K, todavia, foi observado efeito de L-Glu sobre as atividades relativas, expressas em UI/g de tecido e UI/100 g de peso corporal. Os resultados sugerem que a associação entre L-Glu e Vit K interfere na atividade enzimática de alfa-amilase em aves de corte submetidas a dietas purificadas. Os resultados indicam que o nível de 12,5% de L-Glu associado aos diferentes níveis de vitamina K reduziu a atividade enzimática.
The effects of two levels of L-glutamic acid (L-Glu) combined with four levels of vitamin K (Vit K) were studied with the objective of evaluating the activities of alpha-amylase in the chyme and pancreas of chicks. Day-old male broilers were reared in eletrically heated batteries, fed with purified amino acids diets, minerals and vitamins (except Vit K) at the requirement levels, for 14 days. Feed and water were provided ad libitum. The experimental design was a factorial 2x4 with four replicates with eight chicks each The experimental diet was supplemented with 6.25 and 12.5% of L-Glu combined with .02; .2; 20; 200 mg Vit K/kg diet. Effects of L-Glu and Vit K were observed in the chyme with maximum specific activity (1,005.78±245.25 IU/g tissue) observed in chicks fed with 6.25% of L-Glu and 20 mg of Vit K. Reduction in activity was observed with 12.5% L-Glu. In pancreas, there was no significant interaction between L-Glu and Vit K for alpha-amylase activity, but significant effect of L-Glu was observed (IU/g tissue and IU/100 g body weight). Results suggest that alpha-amylase activity is affected by an association between L-Glu and Vit K in chickens fed purified diets. In general, 12.5% L-Glu regardless of Vit K levels led to reduction in alpha-amylase activity.
O complexo HLA tem sido amplamente estudado, na tentativa de elucidar os mecanismos que levam ao direcionamento da forma clínica na hanseníase. Foram observadas associações positivas dos alelos ...HLA-DR2 e HLA-DR3, com a forma tuberculóide (HT) e do alelo HLADQ1, com a forma virchoviana (HV). Em relação ao HIV os alelos de classe I, HLA-B35 e HLA-Cw4 parecem estar mais fortemente associados com a deterioração imunológica e com a aceleração da progressão para a aids e os alelos HLA-A1, HLA-B8, HLA-B27, HLA-Cw7 e os de classe II, HLADR3 e HLA-DQ2 com a progressão lenta da doença. Por não haver nenhum dado na literatura descrevendo a participação dos alelos HLA em indivíduos co-infectados com hanseníase e HIV, o presente estudo teve como objetivo avaliar a freqüência dos alelos HLA de Classe I e II (locus A, B, C, DR e DQ) em pacientes co-infectados, atendidos no Instituto Lauro de Souza Lima de Bauru. Foi possível observar que a presença de alelos descritos na literatura como associados com rápida progressão da aids parece não infl uenciar no espectro clínico da hanseníase. Embora a infecção pelo HIV cause profundos danos no sistema imune, não houve direcionamento para a forma virchoviana multibacilar como se poderia esperar. Estudos abrangendo maior casuística devem ser conduzidos para que os resultados sejam mais informativos uma vez que a co-infecção HIV/M.leprae é um evento importante em área endêmica para ambas as doenças.

In cattle, in vitro embryo production (IVEP) is an important reproductive biotechnology responsible for the rapid expansion of the Senepol breed in our country. This breed has shown important results ...when used in crossbreeding and estimate IVEP in Senepol based on seminal analysis would be valuable for the semen cryopreservation industry, research institutes and breeders. Combining the evaluation of sperm subpopulations with analysis of other sperm attributes may help to improve fertility predictions in cattle. Therefore, the objectives of the present study were to: 1) identify and characterize motile sperm subpopulations in cryopreserved Senepol semen following the washing process carried out before in vitro fertilization, and 2) to determine an model for estimate IVEP based on sperm subpopulations in conjunction with other sperm quality analyzes. Samples of 38 cryopreserved semen from 28 Senepol bulls, chosen based on retrospective data from 386 IVEP routines, underwent the semen washing and were evaluated by the computer-assisted sperm analysis system. Sperm morphology was evaluated by wet preparation technique, and plasma and acrosomal membranes integrity, mitochondrial potential, oxidative status and chromatin resistance were analyzed by flow cytometry. After multivariate analysis of principal components and grouping, three sperm subpopulations were identified: SBP1 (fast and progressive motility), SBP2 (hyperactivated motility) and SBP3 (slow non-progressive motility). After categorization of IVEP in high, medium and low embryo yield, logistic regression analysis was applied to associate the results of subpopulations and other sperm quality variables with IVEP. The SBP1 and SBP2 variables affected embryo production, and an IVEP estimation model was generated for Senepol bulls based on these two subpopulations: embryo yield = 0.1563 + 0.0328 (SBP1) + 0.0173 (SBP2). SBP1 and SBP2 represents the absolute value of the percentage of subpopulations in semen. If the calculated value (by this equation) is close to 1, the embryo yield will be low; if is close to 2, will be medium; if is close to 3, will be high. In conclusion, three subpopulations were found for Senepol semen and, despite all analyzed variables, only SBP1 and SBP2 were included in the model to estimate IVEP in this breed.
