The efficient global optimization method (EGO) based on kriging surrogate model and expected improvement (EI) has received much attention for optimization of high-fidelity, expensive functions. ...However, when the standard EI method is directly applied to a variable-fidelity optimization (VFO) introducing assistance from cheap, low-fidelity functions via hierarchical kriging (HK) or cokriging, only high-fidelity samples can be chosen to update the variable-fidelity surrogate model. The theory of infilling low-fidelity samples towards the improvement of high-fidelity function is still a blank area. This article proposes a variable-fidelity EI (VF-EI) method that can adaptively select new samples of both low and high fidelity. Based on the theory of HK model, the EI of the high-fidelity function associated with adding low- and high-fidelity sample points are analytically derived, and the resulting VF-EI is a function of both the design variables

Through maximizing the VF-EI, both the sample location and fidelity level of next numerical evaluation are determined, which in turn drives the optimization converging to the global optimum of high-fidelity function. The proposed VF-EI is verified by six analytical test cases and demonstrated by two engineering problems, including aerodynamic shape optimizations of RAE 2822 airfoil and ONERA M6 wing. The results show that it can remarkably improve the optimization efficiency and compares favorably to the existing methods.

. Here we determined the crystal structure of the receptor-binding domain (RBD) of the spike protein of SARS-CoV-2 (engineered to facilitate crystallization) in complex with ACE2. In comparison with the SARS-CoV RBD, an ACE2-binding ridge in SARS-CoV-2 RBD has a more compact conformation; moreover, several residue changes in the SARS-CoV-2 RBD stabilize two virus-binding hotspots at the RBD-ACE2 interface. These structural features of SARS-CoV-2 RBD increase its ACE2-binding affinity. Additionally, we show that RaTG13, a bat coronavirus that is closely related to SARS-CoV-2, also uses human ACE2 as its receptor. The differences among SARS-CoV-2, SARS-CoV and RaTG13 in ACE2 recognition shed light on the potential animal-to-human transmission of SARS-CoV-2. This study provides guidance for intervention strategies that target receptor recognition by SARS-CoV-2.
Wildfires are widespread disasters and are concurrently influenced by global climatic drivers. Due to the widespread and far-reaching influence of climatic drivers, separate regional wildfires may ...have similar climatic cause mechanisms. Determining a suite of global climatic drivers that explain most of the variations in different homogeneous wildfire regions will be of great significance for wildfire management, wildfire prediction, and global wildfire climatology. Therefore, this study first identified spatiotemporally homogeneous regions of burned area worldwide during 2001-2019 using a distinct empirical orthogonal function. Eight patterns with different spatiotemporal characteristics were identified. Then, the relationships between major burned area patterns and sixteen global climatic drivers were quantified based on wavelet analysis. The most significant global climatic drivers that strongly impacted each of the eight major wildfire patterns were identified. The most significant combinations of hotspots and climatic drivers were Atlantic multidecadal Oscillation-East Pacific/North Pacific Oscillation (EP/NP)-Pacific North American Pattern (PNA) with the pattern around Ukraine and Kazakhstan, El Niño/Southern Oscillation-Arctic Oscillation (AO)-East Atlantic/Western Russia Pattern (EA/WR) with the pattern in Australia, and PNA-AO-Polar/Eurasia Pattern-EA/WR with the pattern in Brazil. Overall, these results provide a reference for predicting wildfire and understanding wildfire homogeneity.
Exosomes, extracellular vesicles with diameters ranging from 30 to 150 nm, are widely present in various body fluids. Recently, microRNAs (miRNAs) have been identified in exosomes, the biogenesis, ...release, and uptake of which may involve the endosomal sorting complex required for transport (ESCRT complex) and relevant proteins. After release, exosomes are taken up by neighboring or distant cells, and the miRNAs contained within modulate such processes as interfering with tumor immunity and the microenvironment, possibly facilitating tumor growth, invasion, metastasis, angiogenesis and drug resistance. Therefore, exosomal miRNAs have a significant function in regulating cancer progression. Here, we briefly review recent findings regarding tumor-derived exosomes, including RNA sorting and delivering mechanism. We then describe the intercommunication occurring between different cells via exosomal miRNAs in tumor microenvironmnt, with impacts on tumor proliferation, vascularization, metastasis and other biological characteristics. Finally, we highlight the potential role of these molecules as biomarkers in cancer diagnosis and prognosis and tumor resistance to therapeutics.
This article studies a nonlocal dispersal susceptible-infected-susceptible (SIS) epidemic model with Neumann boundary condition in spatially heterogeneous environment, where the spatial movement of ...individuals is described by dσm∫ΩJσ(x−y)u(y)−u(x)dy,σ is the scaling of the size of the support of dispersal kernel J, and m represents the cost parameter. We firstly derive the basic reproduction number R0 for this model, and show that 1−R0 has the same sign as the generalised principal eigenvalue of a linear nonlocal operator. We also investigate asymptotic profiles of R0 with respect to σ,m and diffusion rates. Next, the existence and uniqueness of endemic equilibrium state of this model is obtained as R0>1. Finally, we consider the impact of small dispersal spread on the persistence criteria for disease. Particularly, our results illustrate that, even though ∫Ωβ(x)dx>∫Ωγ(x)dx, there exists the possibility of disease control only when m∈0,2)andσ→0+.
In recent years, deep-learning-based WiFi fingerprinting has been intensively studied as a promising technology for providing accurate indoor location services. However, it still demands a ...time-consuming and labor-intensive site survey and suffers from the fluctuation of wireless signals. To address these issues, we propose a prototypical network-based positioning system, which explores the power of few-shot learning to establish a robust RSSI-position matching model with limited labels. Our system uses a temporal convolutional network as the encoder to learn an embedding of the individual sample, as well as its quality. Each prototype is a weighted combination of the embedded support samples belonging to its position. Online positioning is performed for an embedded query sample by simply finding the nearest position prototype. To mitigate the space ambiguity caused by signal fluctuation, the Kalman Filter estimates the most likely current RSSI based on the historical measurements and current measurement in the online stage. The extensive experiments demonstrate that the proposed system performs better than the existing deep-learning-based models with fewer labeled samples.
With the rise of the Internet and the booming development of information technology, more and more IT projects have been launched on the information infrastructure. However, due to the large ...uncertainty and specificity of IT projects, the project delays are very serious. Both the Critical Path Method (CPM) and Program Evaluation and Review Technique (PERT) can no longer meet the management requirements. This paper has improved the Critical Chain Project Management (CCPM) to strengthen the progress and quality management of IT projects. According to the characteristics of IT projects, an improved Critical Chain Project Management (NS-CCPM) had been proposed, and which was based on social network and software maturity models. Then the application of NS-CCPM has been specifically elaborated in three aspects: critical chain identification, buffer design, and buffer dynamic management. Finally, a case was shown, which used the Monte Carlo simulation method to verify the availability of NS-CCPM.
Fire is one of the most unfavorable conditions that cement-based composites can face during their service lives. The uniaxial tensile and flexural tensile properties of the steel-polyvinyl alcohol ...fiber-calcium carbonate whisker (CW) multi-scale fiber reinforced cement matrix composites (MSFRCs) under high temperatures are studied, including strength, deformation capacity, energy dissipation capacity, and its ability to be assessed through the empirical calculation method. The study showed that with the increase of the treatment temperature, the MSFRC residual bending strength, bending toughness, and tensile strength decreased overall, but the decline was slow at 600 °C. The peak flexural deflection and peak tensile strain of MSFRC first reduced and then increased with the increase of the temperature. As the temperature increased, the nominal stiffness of MSFRC bending and straight gradually reduced, and the rate of decline was faster than that of its strength. However, the uniaxial tensile properties were more sensitive to the temperature and degraded more rapidly. A quantitative relationship was established between MSFRC residual bending, tensile strength, and temperature. A comparison with existing research results shows that MSFRC has achieved an ideal effect of high temperature resistance. The multi-scale hybrid fiber system significantly alleviates the deterioration of cement-based composite's mechanical properties under high temperatures. With the help of an optical microscope and scanning electron microscope (SEM), the high temperature influence mechanism on the uniaxial tensile and flexural properties of MSFRC was revealed.
