), which is a marker protein for autophagy and protein conversion from LC3-I to LC3-II. Inhibiting autophagy with chloroquine increased significantly the LDs content and decreased fatty acid β-oxidation and esterification activities in the ZFL cells cultured in the fed state. Furthermore, inhibiting autophagy function downregulated the mRNA expression of the genes and their proteins related to lipid metabolism. Altogether, the present study verified the existence of lipophagy and its essential regulatory roles in lipid metabolism in fish cells.
We present the first example of charged imidazolium functionalized porphyrin‐based covalent organic framework (Co‐iBFBim‐COF‐X) for electrocatalytic CO2 reduction reaction, where the free anions ...(e.g., F−, Cl−, Br−, and I−) of imidazolium ions nearby the active Co sites can stabilize the key intermediate *COOH and inhibit hydrogen evolution reaction. Thus, Co‐iBFBim‐COF‐X exhibits higher activity than the neutral Co‐BFBim‐COF, following the trend of F−<Cl−<Br−<I−. Particularly, the Co‐iBFBim‐COF‐I− showed nearly 100 % CO2 selectivity at a low full‐cell voltage of 2.3 V, and achieved a high CO2 partial current density of 52 mA cm−2 with a turnover frequency of 3018 h−1 at 2.4 V in the anion membrane electrode assembly, which is 3.57 times larger than that of neutral Co‐BFBim‐COF. This work provides new insight into the importance of free anions in the stabilization of intermediates and decreasing the local binding energy of H2O with active moiety to enhance CO2 reduction reaction.
Charged imidazolium struts were integrated into two‐dimensional cobalt porphyrin‐based covalent organic frameworks (Co‐iBFBim‐COF‐X) creating catalytic pockets with 5,10,15,20‐tetrakis(4‐aminophenyl)‐porphinatocobalt centers and halogen counter ions (e.g., F−, Cl−, Br−, and I−). The free imidazolium anions in the proximity of the active Co sites in the Co‐iBFBim‐COF‐X can stabilize the key intermediate *COOH and inhibit the occurrence of HER through hydrogen bonding.
We discuss the contribution of right-handed neutrinos (RHNs) to the effective neutrino mass of the neutrinoless double beta decay within the minimal type-I seesaw model using the intrinsic seesaw ...relation of neutrino mass and mixing parameters and the relative mass dependence of the nuclear matrix elements. In the viable parameter space, we find the possibilities of both the enhancement and cancellation to the effective neutrino mass from RHNs. The bounds on the parameter space of the RHNs can be determined with the effective neutrino mass extracted from neutrinoless double beta decay experiments.
The purpose of this study was to identify microRNAs (miRNAs) involved in the pathology of colorectal cancer (CRC) liver metastasis and investigate their underlying mechanisms. A total of 39 miRNAs ...were identified to be differentially expressed between 16 primary CRC tissues with liver metastases and 16 CRC tissues without liver metastases from 32 patients by Affymetric miRNA microarrays. A panel of eight miRNAs were confirmed to be significantly and differentially expressed between CRC tissues with and without liver metastases through quantitative reverse‐transcription polymerase chain reaction (RT‐PCR) analysis in the 32 patients. In a validated cohort of 99 CRC patients (44 with and 55 without liver metastases), only miR‐214 was validated to be significantly down‐regulated in CRC with liver metastases, which was associated with an unfavorable prognosis. Ectopic expression of miR‐214 suppressed proliferation, migration, and invasion in vitro, tumor growth and liver metastasis in an in vivo xenograft mouse model, whereas miR‐214 knockdown promoted proliferation, migration, and invasion in CRC cell lines. Further studies indicated that fibroblast growth factor receptor 1 (FGFR1) was a potential target of miR‐214. Restoring miR‐214 expression in CRC cells decreased endogenous FGFR1 messenger RNA (mRNA) and protein levels. FGFR1 knockdown mimicked the tumor suppressive effect of miR‐214 on CRC cells, while reintroduction of FGFR1 abolished the tumor suppressive effect of miR‐214 on CRC cells. Moreover, miR‐214 expression levels were inversely correlated with FGFR1 in CRC patients. Conclusion: Down‐regulation of miR‐214 expression was correlated with increased FGFR1 expression levels, which may contribute to increased CRC liver metastasis. miR‐214 may serve as a potential marker to predict survival, and the miR‐214‐FGFR1 axis may be a therapeutic target in CRC patients. Peroxisome proliferation activated receptor α (PPARα) is an important transcriptional regulator of lipid metabolism and is activated by high-fat diet (HFD) and fibrates in mammals. However, whether ...nutritional background affects PPARα activation and the hypolipidemic effects of PPARα ligands have not been investigated in fish. In the present two-phase study of Nile tilapia (Oreochromis niloticus), fish were first fed a HFD (13% fat) or low-fat diet (LFD; 1% fat) diet for 10 weeks, and then fish from the first phase were fed the HFD or LFD supplemented with 200 mg/kg body weight fenofibrate for 4 weeks. The results indicated that the HFD did not activate PPARα or other lipid catabolism-related genes. Hepatic fatty acid β-oxidation increased significantly in the HFD and LFD groups after the fenofibrate treatment, when exogenous substrates were sufficiently provided. Only in the HFD group, fenofibrate significantly increased hepatic PPARα mRNA and protein expression, and decreased liver and plasma triglyceride concentrations. This is the first study to show that body fat deposition and dietary lipid content affects PPARα activation and the hypolipidemic effects of fenofibrate in fish, and this could be due to differences in substrate availability for lipid catabolism in fish fed with different diets.
Conversion-type transition-metal phosphide anode materials with high theoretical capacity usually suffer from low-rate capability and severe capacity decay, which are mainly caused by their inferior ...electronic conductivities and large volumetric variations together with the poor reversibility of discharge product (Li3P), impeding their practical applications. Herein, guided by density functional theory calculations, these obstacles are simultaneously mitigated by confining amorphous FeP nanoparticles into ultrathin 3D interconnected P-doped porous carbon nanosheets (denoted as FeP@CNs) via a facile approach, forming an intriguing 3D flake-CNs-like configuration. As an anode for lithium-ion batteries (LIBs), the resulting FeP@CNs electrode not only reaches a high reversible capacity (837 mA h g–1 after 300 cycles at 0.2 A g–1) and an exceptional rate capability (403 mA h g–1 at 16 A g–1) but also exhibits extraordinary durability (2500 cycles, 563 mA h g–1 at 4 A g–1, 98% capacity retention). By combining DFT calculations, in situ transmission electron microscopy, and a suite of ex situ microscopic and spectroscopic techniques, we show that the superior performances of FeP@CNs anode originate from its prominent structural and compositional merits, which render fast electron/ion-transport kinetics and abundant active sites (amorphous FeP nanoparticles and structural defects in P-doped CNs) for charge storage, promote the reversibility of conversion reactions, and buffer the volume variations while preventing pulverization/aggregation of FeP during cycling, thus enabling a high rate and highly durable lithium storage. Furthermore, a full cell composed of the prelithiated FeP@CNs anode and commercial LiFePO4 cathode exhibits impressive rate performance while maintaining superior cycling stability. This work fundamentally and experimentally presents a facile and effective structural engineering strategy for markedly improving the performance of conversion-type anodes for advanced LIBs.
Drug-induced liver injury (DILI) is an important clinical problem, which has received more attention in recent decades. It can be induced by small chemical molecules, biological agents, traditional ...Chinese medicines (TCM), natural medicines (NM), health products (HP), and dietary supplements (DS). Idiosyncratic DILI is far more common than intrinsic DILI clinically and can be classified into hepatocellular injury, cholestatic injury, hepatocellular-cholestatic mixed injury, and vascular injury based on the types of injured target cells. The CSH guidelines summarized the epidemiology, pathogenesis, pathology, and clinical manifestation and gives 16 evidence-based recommendations on diagnosis, differential diagnosis, treatment, and prevention of DILI.
We propose an effective highest occupied d‐orbital modulation strategy engendered by breaking the coordination symmetry of sites in the atomically precise Cu nanocluster (NC) to switch the product of ...CO2 electroreduction from HCOOH/CO to higher‐valued hydrocarbons. An atomically well‐defined Cu6 NC with symmetry‐broken Cu−S2N1 active sites (named Cu6(MBD)6, MBD=2‐mercaptobenzimidazole) was designed and synthesized by a judicious choice of ligand containing both S and N coordination atoms. Different from the previously reported high HCOOH selectivity of Cu NCs with Cu−S3 sites, the Cu6(MBD)6 with Cu−S2N1 coordination structure shows a high Faradaic efficiency toward hydrocarbons of 65.5 % at −1.4 V versus the reversible hydrogen electrode (including 42.5 % CH4 and 23 % C2H4), with the hydrocarbons partial current density of −183.4 mA cm−2. This is the first attempt to regulate the coordination mode of Cu atom in Cu NCs for hydrocarbons generation, and provides new inspiration for designing atomically precise NCs for efficient CO2RR towards highly‐valued products.
Breaking the coordination symmetry of Cu site in atomically precise Cu6 cluster forms Cu‐S2N1 site, which rank the dx2‐y2 orbital as the highest occupied d orbital to favor the specific coordination between C atom of CO2 and Cu−S2N1 site. This binding mode is conductive to the generation of *COOH instead of *OCHO, thereby switching the product of electrocatalytic CO2 reduction reaction to higher‐valued hydrocarbons.
Radiomics is an emerging field in oncological research. In this study, we aimed at developing a radiomics score (rad-score) to estimate postoperative recurrence and survival in patients with solitary ...hepatocellular carcinoma (HCC).
A total of 319 solitary HCC patients (training cohort: n = 212; validation cohort: n = 107) were enrolled. Radiomics features were extracted from the artery phase of preoperatively acquired computed tomography (CT) in all patients. A rad-score was generated by using the least absolute shrinkage and selection operator (lasso) logistic model. Kaplan-Meier and Cox's hazard regression analyses were used to evaluate the prognostic significance of the rad-score. Final nomograms predicting recurrence and survival of solitary HCC patients were established based on the rad-score and clinicopathological factors. C-index and calibration statistics were used to assess the performance of nomograms.
Six potential radiomics features were selected out of 110 texture features to formulate the rad-score. Low rad-score positively correlated with aggressive tumor phenotypes, like larger tumor size and vascular invasion. Meanwhile, low rad-score was significantly associated with increased recurrence and reduced survival. In addition, multivariate analysis identified the rad-score as an independent prognostic factor (recurrence: Hazard ratio (HR): 2.472, 95% confident interval (CI): 1.339-4.564, p = 0.004;survival: HR: 1.558, 95%CI: 1.022-2.375, p = 0.039). Notably, the nomogram integrating rad-score had a better prognostic performance as compared with traditional staging systems. These results were further confirmed in the validation cohort.
The preoperative CT image based rad-score was an independent prognostic factor for the postoperative outcome of solitary HCC patients. This score may be complementary to the current staging system and help to stratify individualized treatments for solitary HCC patients.
