The mixtures of toxins and taste-and-odor compounds present during cyanobacterial blooms are not well characterized and of particular concern when evaluating potential human health risks. ...Cyanobacterial blooms were sampled in twenty-three Midwestern United States lakes and analyzed for community composition, thirteen cyanotoxins by liquid chromatography/mass spectrometry and immunoassay, and two taste-and-odor compounds by gas chromatography/mass spectrometry. Aphanizomenon, Cylindrospermopsis and/or Microcystis were dominant in most (96%) blooms, but community composition was not strongly correlated with toxin and taste-and-odor occurrence. Microcystins occurred in all blooms. Total microcystin concentrations measured by liquid chromatography/mass spectrometry and immunoassay were linearly related (r s = 0.76, p < 0.01) and LC/MS/MS concentrations were lower than or similar to ELISA in most (85%) samples. Geosmin (87%), 2-methylisoborneol (39%), anatoxin-a (30%), saxitoxins (17%), cylindrospermopsins (9%), and nodularin-R (9%) also were present in these blooms. Multiple classes of cyanotoxins occurred in 48% of blooms and 95% had multiple microcystin variants. Toxins and taste-and-odor compounds frequently co-occurred (91% of blooms), indicating odor may serve as a warning that cyanotoxins likely are present. However, toxins occurred more frequently than taste-and-odor compounds, so odor alone does not provide sufficient warning to ensure human-health protection.
This paper applies public policy concepts such as agenda setting, resource allocation, lobbying, and campaign rhetoric to the field of homeland security. The analysis examines the allocation of ...resources among the U.S. federal government’s five broad homeland security priorities during the fiscal years of 2012-2017. An overemphasis on border security has led to a disproportionate allocation of resources among these various priorities. The uneven distribution is the summation of electoral ambitions, campaign contributions, and corporate lobbying. This analysis highlights the negative consequences that are a direct result of an unbalanced allotment, cumulating in a slow and uncoordinated federal response to both the COVID-19 pandemic and a series of domestic terrorist attacks. The article concludes with a series of reforms that can be implemented by policy makers in order to ensure that resources are distributed more evenly in the future. The historical and ongoing lead (Pb) contamination caused by the 20th-century use of leaded gasoline was investigated by an analysis of bottom sediment in eight small rural reservoirs in eastern ...Kansas, USA. For the reservoirs that were completed before or during the period of maximum Pb emissions from vehicles (i.e., the 1940s through the early 1980s) and that had a major highway in the basin, increased Pb concentrations reflected the pattern of historical leaded gasoline use. For at least some of these reservoirs, residual Pb is still being delivered from the basins. There was no evidence of increased Pb deposition for the reservoirs completed after the period of peak Pb emissions and (or) located in relatively remote areas with little or no highway traffic. Results indicated that several factors affected the magnitude and variability of Pb concentrations in reservoir sediment including traffic volume, reservoir age, and basin size. The increased Pb concentrations at four reservoirs exceeded the U.S. Environmental Protection Agency threshold-effects level (30.2 mg kg-1) and frequently exceeded a consensus-based threshold-effects concentration (35.8 mg kg-1) for possible adverse biological effects. For two reservoirs it was estimated that it will take at least 20 to 70 yr for Pb in the newly deposited sediment to return to baseline (pre-1920s) concentrations (30 mg kg-1) following the phase out of leaded gasoline. The buried sediment with elevated Pb concentrations may pose a future environmental concern if the reservoirs are dredged, the dams are removed, or the dams fail.
Neutrinos are fundamental particles in the standard model and play an important role in the current understanding of the universe; however, the mass of the neutrino one of the most fundamental ...parameters for any particle, is currently unknown. This fact represents a gaping hole in our current knowledge of the universe that may provide clues to the energy scale of physics beyond the standard model. This dissertation summarizes research and development as a member of the Project 8 collaboration towards an experiment to measure the neutrino mass with a sensitivity below 50 meV/c2. which is an order of magnitude less than the most sensitive direct measurements of the neutrino mass to date. Project 8 will perform this measurement using Cyclotron Radiation Emission Spectroscopy (CRES) to measure the beta-decay endpoint spectrum of atomic tritium. I present an analysis of the signal reconstruction performance of an antenna array system designed to perform large-scale CRES measurements in cubic-meter volumes. Next, I discuss an approach to calibrating an antenna-based CRES experiment using a unique probe antenna designed to mimic radiation from CRES events. Finally, I present design studies for a resonant cavity that could be used to perform a CRES experiment with atomic tritium at multi-cubic-meter scales.
Diabetes mellitus type 1 and type 2 are diseases characterized by impaired regulation of blood glucose due to decreased insulin production and insulin resistance, respectively. Management of diabetes ...mellitus often requires injection of exogenous insulin. Continuous subcutaneous insulin infusion (CSII or insulin pump) is a diabetes treatment modality utilizing a device to aid in regulation of glycemic control. Malfunctions in device components can have rare fatal consequences. Described in this report are six fatalities due to one such malfunction, the failure of plastic cannulas of CSII devices to penetrate the skin and deliver insulin, resulting in fatal diabetic ketoacidosis (DKA). The cases derive from four different death investigation systems. For each case, scene and autopsy findings are presented, as well as selected toxicology and histology findings. These cases illustrate the importance of careful examination of CSII devices in death investigations and introduce a discussion on discrepant manner of death classifications.
Despite the many advancements to forensic DNA analysis adopted by crime laboratories across the country, the most common method for the differential extraction of sexual assault samples has remained ...relatively unchanged since forensic deoxyribonucleic acid (DNA) typing was discovered in 1985. As the quantity and quality of extracted DNA has significant implications on the success of subsequent analysis methods, the development and optimization of effective extraction procedures is vital to progressing the field of forensic DNA analysis. The graduate students and faculty at the Boston University School of Medicine have been developing a differential extraction process that utilizes a multi-enzymatic approach to preferentially lyse and wash the cell types within temperature controlled environments. The overall procedure is less labor-intensive and time-consuming than the conventional method. Through the extraction process, the inhibitory nature of each enzyme on the amplification process is avoided, circumventing the need for an additional purification step. A single centrifugation step is required in order to pellet the sperm while the cumbersome wash steps are replaced with selective digestion in order to remove the residual epithelial cell DNA from the sperm fraction. The three enzyme used (EA1, Benzonase®, and Acrosolv) operate optimally at distinct temperatures which allows for controlled and sequential activation to achieve desired lysis and digestion outcomes. The enzymatic reactions are conducted within a DNA extraction lab thermal cycler to obtain rapid and accurate temperature changes. This novel temperature controlled differential extraction protocol has been developed and optimized for extraction of primarily liquid mixed samples in 0.2 milliliter (mL) tubes. The epithelial cell lysis and sperm cell lysis stages of the extraction contained a final reaction volume of 100 microliters (µL). Slight modifications to this 100 direct-lysis differential extraction method resulted in a similarly efficient method with a high male DNA yield (74-100%) and minimal female carryover among varying ratios of epithelial cells to sperm cells. This sensitive technique provided nearly complete profiles (14/16 loci) of the male contributor in mixed samples containing ~15,200 female epithelial cells and ~500 sperm, with complete profiles observed in mixed samples containing ~1000 sperm. This modified extraction protocol better accommodates sample sizes that may be encountered in forensic casework testing while providing a more concentrated sperm fraction, possibly eliminating the need for an additional concentration step in some dilute samples. The ease of implementation and the rapid processing time of 2-3 hours make it a great candidate for use in forensic DNA laboratories and may help alleviate backlogs of sexual assault kit. However, further work is needed to alter the composition of the sperm lysis buffer to make it compatible with currently used amplification kits. Until such time, caution must be taken in the kit selection used for amplification of extracts produced with this method. This study also demonstrated a sensitivity of the GlobalFiler® PCR Amplification Kit to inhibition by the buffers used in this extraction protocol, particularly the Orange+ Buffer. This inhibition has dramatic effects on the profile quality of the amplified sperm fractions, with extensive allelic drop-out observed even when the Orange+ Buffer concentration was scaled from 1.0X to 0.2X. Amplification using the AmpFℓSTR® Identifiler® Plus PCR Amplification Kit showed marginal recovery in the profile quality. Other expanded-loci STR amplification kits may also demonstrate resistance to this inhibition.
In this work we present a novel application of non-rigid surface detection to enable gestural interaction with applicable surfaces. This method can add interactivity to traditionally passive media ...such as books, newspapers, restaurant menus, or anything else printed on paper. We allow a user to interact with these surfaces in a natural manner and present basic gestures based on pointing and touching. This technique was developed as part of an ongoing effort to create an assisted reading device for the visually impaired. However, it is suited to general applications and can be used as a practical mechanism for interaction with screenless wearable devices. Our key contributions are a unique application of non-rigid surface detection, a basic gesturing paradigm, and a proof of concept system.
