) as an alternative behavioral model of formalin-induced nociception. We evaluated the nociceptive effect of 0.1% formalin (3 or 5 μL; intramuscularly i.m.), applied into the tail or lips, on locomotor activity, using as parameter the number of times the fish crossed the lines between the quadrants of a glass Petri dish during the neurogenic stage (0–5 min) and the inflammatory stage (15–30 min). The behavioral model was validated by testing the antinociceptive effect of morphine and indomethacin (standard analgesic drugs used in the formalin test of rodents). We also tested whether the effect of morphine could be modulated by naloxone, an opioid antagonist. The effect of morphine and indomethacin on zebrafish locomotor behavior was evaluated with the open field test. The white/black test was used to rule out the anxiolytic effect of 0.1% formalin injected into the tail on adult zebrafish. Formalin (0.1%; 3 and 5 μL injected into the tail) increased significantly the nociceptive behavior of the adult zebrafish in both stages (
p
< 0.001 vs. control). Morphine and indomethacin (both 0.2 mg/mL; 20 μL; intraperitoneally i.p.) significantly inhibited nociception induced with formalin (5 μL injected i.m. into the tail) in both stages (
p
< 0.001). Naloxone blocked the antinociceptive effect of morphine. No influence on locomotion was observed. Locally administered formalin (injected into the tail) induced nociception, but not anxiety. The results suggest that the adult zebrafish behavioral model is a feasible alternative to more conventional laboratory models used in research on novel compounds with antinociceptive potential.
Introdução: A hérnia de disco é a projeção do núcleo pulposo para além de seus limites normais. A Ressonância Magnética (RM) é a melhor modalidade de imagens para avaliação da coluna vertebral. ...Objetivo: analisar, epidemiologicamente, os indivíduos com hérnias discais avaliadas pelo método de ressonância magnética. Método: foi aplicado um questionário em uma amostra composta por 268 indivíduos que procuraram diagnóstico médico de hérnia discal por meio da Ressonância Magnética. De acordo com os resultados, a média de idade foi de 50,22 ± 11,75 anos, com IMC médio de 27,5 ± 3,63 m2/Kg, 56% dos indivíduos são do sexo masculino, 33,95% só cursaram o ensino fundamental completo, 25,57% trabalham em serviços gerais, 88,80% têm uma jornada de trabalho maior que 6 horas diárias e 40,18% estão afastados de suas atividades laborais. A renda mensal de 66,04% é de até um salário mínimo. O espaço intervertebral mais acometido foi L5-S1 em 63,43% (n = 170). Conclusão: o aumento da idade e do peso corporal contribui para o aparecimento de hérnias discais, sendo os homens os mais acometidos. As profissões também influenciam no surgimento de hérnias, sobretudo as vinculadas a esforço físico e a altas cargas horárias de trabalho, e o nível com maior incidência é na coluna lombar.

Seaweed polysaccharides are increasingly important in the biochemical and medical fields. ...Previous work has already demonstrated the antinociceptive effect of polysaccharides of the red marine macroalga Hypnea pseudomusciformis. This study aimed to evaluate the antinociceptive effect of sulfated polysaccharides from seaweed Hypnea pseudomusciformis (PLS) on experimental models of orofacial pain. The algae were collected, identified and deposited in the Herbarium of the Institute of Marine Sciences - Labomar. The algae PLS were extracted and submitted to chemical characterization by FTIR and RNM H1 and C13. Toxicity tests were performed against A. salina and Vero cells, as well as preliminary antioxidant activity test. Afterwards, formalin-induced acute orofacial nociception, capsaicin, cinnamaldehyde, acid saline or glutamate (cutaneous models) tests were performed. Using the glutamate model, the antinociceptive mechanisms of neuromodulation were investigated by treating the animals with the antagonists red rutinium (TRPV1), glibenclamide (K + ATP), naloxone (opioid), L-NAME (nitric oxide) or blue Of methylene (guanylate cyclase). The test of hypertonic saline induced acute corneal nociception was performed, as well as the tests of temporomandibular and craniofacial nociception induced by formalin and mustard oil, respectively. The motor activity of the PLS treated animals was evaluated with the open field test. The spectrometric data confirmed the structural determination of PLS as being in a higher proportion of ¿-carrageenan, in the infrared spectrum typical absorption bands of PLS were observed with peaks of 811, 875 and 900 cm-1. PLS did not demonstrate cytotoxic potential and had antioxidant activity reducing chelating ions. Pretreatment of animals with PLS significantly reduced nociceptive behavior associated with acute pain. Antinociception was effectively reduced but not inhibited by ruthenium red and ketamine. L-NAME and glibenclamide increased the effect of PLS. The antinociception of PLS was resistant to methylene blue, naloxone and heating - included in the methodology. Our results confirm the potential pharmacological relevance of PLS as an inhibitor of orofacial nociception in acute pain, probably mediated by the glutamatergic and nitrergic systems, and by TRPs and K + -ATP channels.

Os polissacarídeos de algas marinhas são cada vez mais importantes nas áreas bioquímica e médica. Trabalhos anteriores já demonstraram o efeito antinociceptivo de polissacarídeos da macroalga marinha vermelha Hypnea pseudomusciformis. Este estudo teve como objetivo avaliar o efeito antinociceptivo dos polissacarídeos sulfatados da alga marinha Hypnea pseudomusciformis (PLS) em modelos experimentais de dor orofacial. A alga foi coletada, identificada e depositada no Herbário do Instituto de Ciências do Mar - Labomar. Os PLS da alga foram extraídos e submetidos à caracterização química por FTIR e RNM H1 e C13. Foram realizados testes de toxicidade frente a A. salina e células Vero, bem como teste de atividade antioxidante preliminar. Em seguida, foram realizados os testes de nocicepção orofacial aguda induzida por formalina, capsaicina, cinamaldeído, salina ácida ou glutamato (modelos cutâneos). Empregando-se o modelo do glutamato, foram investigados os mecanismos antinociceptivos de neuromodulação tratando-se os animais com os antagonistas vermelho rutêndio (TRPV1), glibenclamida (K+ATP), naloxona (opióide), L-NAME (óxido nítrico) ou azul de metileno (guanilato ciclase). Foi realizado o teste de nociceção corneal aguda induzida por salina hipertônica, bem como os testes de nocicepção temporomandibular e craniofacial induzida por formalina e óleo de mostarda, respectivamente. A atividade motora dos animais tratados com PLS foi avaliada com o teste em campo aberto. Os dados espectrométricos confirmaram determinação estrutural dos PLS como sendo em maior proporção de ¿-carragenana, no espectro de infravermelho foi observado bandas de absorção típica do PLS com picos de 811, 875 e 900 cm-1. PLS não demonstrou potencial citotóxico e apresentou atividade antioxidante redutora de íons quelantes. O pré-tratamento dos animais com PLS reduziu significativamente o comportamento nociceptivo associado à dor aguda. A antinocicepção foi efetivamente reduzida, mas não inibida, pelo vermelho de rutênio e pela cetamina. L-NAME e glibenclamida aumentaram o efeito do PLS. A antinocicepção do PLS foi resistente ao azul de metileno, à naloxona e ao aquecimento ¿ incluir na metodologia. Nossos resultados confirmam a potencial relevância farmacológica dos PLS como inibidor da nocicepção orofacial em dor aguda, provavelmente mediada pelos sistemas glutamatérgico e nitrérgico, e por canais TRPs e K+-ATP.
