Domuz, insan cerrahisinde yürütülen deneysel çalışmalarda tercih edilen bir hayvan türüdür. Sedatif, ataraktik ve ancstezik ajanlara tepkilerinin değişkenliği ve solunum depresyonuna yatkınlıkları, ...domuzlarda güvenilir anestezi tekniklerinin seçimini zorunlu kılar. 60 dakikadan uzun sürecek cerrahi işlemlerde, inhalasyon anestezisi güvenle kullanılabilecek bir yöntemdir.
Domuzlarda inhalasyon anestezisi maske yöntemi ya da entübasyon uygulamasıyla gerçekleştirilir. Bu çalışmada domuzlarda kas içi yolla 0.03 mg/kg atropin ve 3 mg/kg xylazin premedikasyonu uygulanmıştır. 10 mg/kg i.m. ketamin enjeksiyonu sonrasında, i.v. yolla 10 mg/kg thiopental şeklinde düzenlenen anestezi protokolü sonrasında inhalasyon anestezisi gerçekleştirilmiştir.
Tarafımızdan modifiye edilen entübasyon tüplerinin de kullanıldığı çalışmada, 6 olguda entübasyon, 4 olguda maske yöntemiyle halotan ya da isofluranla inhalasyon anestezisi uygulanmıştır.Çalışma sonucunda, domuzlarda yapılacak her türlü majör cerrahi operasyonlarda entübasyonla gerçekleştirilen inhalasyon anestezisi yönteminin, anestezi kalitesi ve güvenilirliği açısından oldukça tatminkar bulunduğu belirlenmiştir.
Pig is an animal species that preferred in experimental studies for human surgery. The variation of the reaction to the sedative, ataractic and anesthetic agent and the inclination to respiratory depression makes it necessary to choose reliable anesthetic methods for the pigs. Inhalations anesthesia is a reliable method in surgery procedures more than 60 minutes.
Inhalations anesthesia in pig is realized by mask technique or intubation application in pig. In this study, pigs were premedicated by intramuscular administration of 0.03 mg/kg atropine and 3 mg/kg xylazin. The anesthesia protocol was scheduled to be contiuned by 10 mg/kg intramuscular ketamine then 10 mg/kg iniravenoz thiopental and followed by inhalation anesthesia.
Intubation tubes modified by us have been used in this study. Inhalation anesthesia has been applied by the mask technique in 4 cases and by intubation in 6 cases. Halothane or isoflurane has been used in inhalation anesthesia.
As a result of research, concerning anesthesia quality and reliability inhalation anesthesia method which is used in all types surgery is found to be the satisfactory one.
Çalışmada, günlük, aşı uygulanmamış, 170 adet, erkek, Ross-PM3 ırkı, etçi civciv kullanılmıştır ve biri kontrol 4'ü deneme 5 grup oluşturulmuştur. Kontrol grubuna aflatoksin içermeyen, deneme ...gruplarına ise sırasıyla 0,05 ppm (Grup II), 0,1 ppm (Grup III), 0,5 ppm (Grup IV) ve l ppm (Grup V) total aflatoksin içeren yem otuz gün süreyle verilmiştir. Denemenin 15 ve 30. gününde alınan kanlarda glikoz, alanin aminotransferaz (ALT), aspartat aminotransferaz (AST), gamma-glutamil transferaz (GGT), alkalen fosfataz (ALP), laktat dehidrogenaz (LDH), kolesterol, trigliserid ve total protein düzeyleri değerlendirilmiştir. Ayrıca, canlı ağırlık kazancı ile 30. gün karaciğer aflatoksin düzeyleri tespit edilmiştir. Otuzuncu günün sonunda, kontrol ve deneme gruplarından 7'şer hayvan üzerinde damar içi ve kursak içi 10 mg/kg.ca dozunda enrofloksasin verilerek farmakokinetik çalışma yapılmıştır. Sonuç olarak, otuz gün süre ile belirtilen dozlarda verilen aflatoksinin kanatlılarda olumsuz etkilerinin olduğu, yapılan çok yönlü değerlendirmelerle anlaşılmıştır. Farmakokinetik çalışma sonucunda ise, hem kursak hem de damar içi uygulamalarda, özellikle de yüksek dozda (0,5-1 ppm) aflatoksin verilen gruplarda (Grup IV-V) ilacın, biyoyararlanımı düşmüş, atılımı hızlanmış dolayısıyla da vücutta kalış süresi kısalmıştır.
In this study, total 170, daily, unvaccinated, male, Ross-PM3 strain, broiler chicks were used. Five main groups were designed; one of them was control and the others experimental groups. While the control group received normal food (without aflatoxin) during to 30 days, the experimental groups received feed with aflatoxin, orderly, 0.05 ppm (Group II), 0.1 ppm (Group III), 0.5 ppm (Group IV), and 1 ppm (Group V). On the 15th and 30th days of experiments, blood was taken and glucose, alanine aminotranferase (ALT), aspartate aminotransferase (AST), gamma-glytamil transferase (GGT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), cholesterol, triglyceride and total protein levels were evaluated. Beside this fact, body weight gain and their liver aflatoxin levels were determined. At the end of 30th day, 7 animals were taken from control and experimental groups and 10 mg/kg.bw enrofloxacin were administrated by IV and intra-crop and pharmacokinetic parameters calculated. It was conclusion that there were negative effects of aflatoxin in poultry, given doses above, for 30 days. The pharmakokinetic investigations showed that for both IV and intra-crop applications, especially in high dose (0.5-1 ppm) aflatoxin given groups (Groups IV-V), bioavailability of drug was lowered, its excretion become rapid, therefore, its terminal phase body half-life ($t_1$/2ß) was shortened.
