Spremljanje kinetike rasti populacije kvasovk v moštu terana s pulzno elektroforezo : diplomska naloga = Population kinetics of yeast in teran must determined by pulsed field gel electrophoresis : graduation thesisPovhe Jemec, KatjaV jeseni 1995 smo skupno odvzeli 6 vzorcev grozdja, 24 mikrobioloških brisov kleti in kletne opreme ter 36 vzorcev mošta med spontano fermentacijo terana v treh različnih vinskih kleteh kraškega ... vinorodnega okoliša. V nalogi so predstavljeni rezultati kvantitativne mikrobiološke analize alkoholne fermentacije v obdobju devetintridesetdnevnega vzorčenja (odvzetih in obdelanih trikrat po 11 vzorcev med fermentacijo, vse v paralelkah) ter kvalitativne mikrobiološke analize v tipičnih (4 vzorčne točke) obdobjih alkoholne fermentacije. Namen raziskave je bil analizirati dinamiko rasti kvasovk, ugotoviti, kateri mikroorganizmi so prisotni med spontano alkoholno fermentacijo, ter proučiti rast posameznih sevov kvasovk. Določili smo nekatere fizikalne - kemijske parametre mošta. Rezultati teh analiz nam pokažejo vpliv etanola, kislin in drugih snovi na zastopanost posameznih sevov kvasovk. Konstruirali smo rastne krivulje kvasovk, iz katerih smo lahko razbrali, da je vrh fermentacij v vseh naših primerih dosežen sedmi dan, kar potrjuje tudi najvišja temperatura fermentacije. Iz mošta in kletarske opreme je bilo izoliranih preko dvesto izolatov kvasovk, od tega iz mošta 182 čistih kultur kvasovk, ki so bile nadeljne kvalitativno določene z elektroforetsko kariotipizacijo, s pomočjo pulzne gelske elektroforeze. Ugotovili smo, da se nekateri sevi ▫{\sl Sacch. cerevisiae}▫ pojavijo le v določeni kleti in samo v določeni fazi fermentacije, drugi pa so geografsko in tudi med celotno fermentacijo bolj razširjeni. Štirje med temi sevi se pojavijo tudi med enako raziskavo fermentacijo refoška, ki je bila opravljena na koprskem vinorodnem območju.Type of material - undergraduate thesis ; adult, seriousPublication and manufacture - Ljubljana : [K.
