Proteins and peptides of animal and human origin, when expressed in plants are usually correctly folded and glycosylated, ... unlike in bacterial expression systems, where this frequently presents a problem. Cathepsin S is highly active lysosomal cysteine proteinase. The involvement of cathepsin S in the pathogenesis of degenerative diseases in humans renders it a possible target for therapy. Katepsin S also plays an immportant role in normal physiological processes. For studying its function and structure larger amounts of enzyme are needed. In order to overcome the problem of the limited amounts of chatepsin S available from tissues, its cloning and expression were undertaken. We used a plant expression system as an alternative to a bacterial expression system, where cathepsin S is deposited in an insoluble and inactive form as inclusion bodies. We transformed potato plants cv. Desireé with the procathepsin S gene under regulation of the CaMV promoter. The procathepsin S gene could be detected in all the tissues of potato plants in vitro and in vivo. We determined the number of procathepsin genes in different clones of transformed potato plants.
