For many years, the identification has relied on investigation of culture properties and biochemical ... characteristics. Conventional biochemical methods are timeconsuming. It was therefore decided to investigate an identitication system based on DNA technology. There are many molecular mcthods for identification of mycobacteria, but none of thcm are suitable for identification of rarely isolated species of non-tuberculous mycobacteria. For rapid identification of this kind of species a newly developed assay GenoType Mycobacterium AS (Hain Lifescience, Nehren, Germany) was evaluated. The assay is able to identify 16 more rarely isolated mycobacterial species. The test is based on polymerse chain reaction followed by hybridisation to specific oligonucleotide probes immobilized on membrane strips. The conventional biochemical identification methods are currently onlymethod used in Slovenia for identification of rarely isolated non-tuberculous mycobacterial species. fhe aim of our study was comparative evaluation of two methods for identification of mycobacterial speciesČ conventional biochemical methods versus molccular assay GenoType MyeobacteriumAS. In the study we included 47 samples, all from Slovenia. Firstwe tried to identify samples with conventional biochemical methods. Then we repeated identification of samples with molecular assay. The results were compared. The molecular assay identified morc samplcs to the species level then conventional biochemical methods, 6 (13%) versus 1 (2%). The advantages of the assay is that provides identification of a large variety of rarely isolated nontubercuolus mycobacterial species in a single test, it can identify multiple mycobacterial species within the samc sample, is rapid and the results are easy to interpret.Type of material - undergraduate thesis ; adult, seriousPublication and manufacture - Ljubljana : [J.
