  • The regeneration capability of different explants for the transformation of potato Solanum tuberosum L. cv. Desirée and cv. Igor

    The transformation of potato plants depends, among other fasctors, on a good regeneration capability of the explants used. In our study we investigated the regeneration efficiency of different ... explants of the potao Solanum tuberosum L. cv. Desirée and Slobenian cv. Igor Leaf pieces, internodes, and petioles were used for testing of different regeneration media. Shooy regeneration was better in the potato cultivar Desirée than in cultivar Igor. The most efficient media contained combinations of the growth regulators zeatin riboside (ZR), naphalenatic acid (NAA), and gibberelic acid (GA[sub]3) described by Webster et al. (1994). The optimal concentration ((micro) M) usedfor cv. Desirée was 6 ZR, 0.1 NAA, and 0.1 GA[sub ]3, and 7 ZR, 1 NAA, and 0.1 GA[sub]3, and 7 ZR, 1 NAA, and 0.1 GA[sub]3 for cv. Igor.These media were used in further transformation experiments. Explants were cocultivated with Agrobacterium tumefacients carrying the plasmid pGA482 with NPT II gene. The suitability for transformation purposes is discussed.
