    Reliable diagnosis of malaria requires laboratory confirmation of the presenceof malaria parasites in the blood of the patient. Even if we are entering in the 21st century, the laboratory techniques ... of the 19th century remain in routine use and continue to be the gold standard: microscopic examination of the Giemsa stained thick blood film for diagnosing malaria (andother blood parasites) and the Pappenheim stained thin blood film for the detection of the characteristic stages of plasmodium parasites in the peripheral blood and the determination of their quantity and species. The detection a small percentage of parasitised cells requres a high quality microscope and experienced microscopist. A new technology is available nowadays for low level infections: the Quantitative Buffy Coat method based onthe hematocrit principle, the fluorochrome acridine orange dye for DNA and RNA of the plasmodium and fluorescent microscopy.
