Cuba is struggling with a growing environmental problem: the uncontrolled spread of the allochthonous weed species marabou (Dichrostachys cinerea) throughout the country. Over the last 70 years, ...marabou has become a formidable invasive species that poses a threat to Cuban biodiversity and agricultural productivity. In this paper, we present a free and affordable method for regularly mapping the spatial distribution of the marabou based on the Google Earth Engine platform and ecological surveys. To test its accuracy, we develop an 18-year remote sensing analysis (2000–2018) of marabou dynamics using the Valle de los Ingenios, a Cuban UNESCO World Heritage Site, as an experimental model. Our spatial analysis reveals clear patterns of marabou distribution and highlights areas of concentrated growth. Temporal trends illustrate the aggressive nature of the species, identifying periods of expansion and decline. In addition, our system is able to detect specific, large-scale human interventions against the marabou plague in the area. The results highlight the urgent need for remedial strategies to maintain the fragile ecological balance in the region.
4-anilino quinazolines have been identified as inhibitors of HCV replication. The target of this class of compounds was proposed to be the viral protein NS5A, although unequivocal proof has never ...been presented. A 4-anilino quinazoline moiety is often found in kinase inhibitors, leading us to formulate the hypothesis that the anti-HCV activity displayed by these compounds might be due to inhibition of a cellular kinase. Type III phosphatidylinositol 4-kinase α (PI4KIIIα) has recently been identified as a host factor for HCV replication. We therefore evaluated AL-9, a compound prototypical of the 4-anilino quinazoline class, on selected phosphatidylinositol kinases. AL-9 inhibited purified PI4KIIIα and, to a lesser extent, PI4KIIIβ. In Huh7.5 cells, PI4KIIIα is responsible for the phosphatidylinositol-4 phosphate (PI4P) pool present in the plasma membrane. Accordingly, we observed a gradual decrease of PI4P in the plasma membrane upon incubation with AL-9, indicating that this agent inhibits PI4KIIIα also in living cells. Conversely, AL-9 did not affect the level of PI4P in the Golgi membrane, suggesting that the PI4KIIIβ isoform was not significantly inhibited under our experimental conditions. Incubation of cells expressing HCV proteins with AL-9 induced abnormally large clusters of NS5A, a phenomenon previously observed upon silencing PI4KIIIα by RNA interference. In light of our findings, we propose that the antiviral effect of 4-anilino quinazoline compounds is mediated by the inhibition of PI4KIIIα and the consequent depletion of PI4P required for the HCV membranous web. In addition, we noted that HCV has a profound effect on cellular PI4P distribution, causing significant enrichment of PI4P in the HCV-membranous web and a concomitant depletion of PI4P in the plasma membrane. This observation implies that HCV--by recruiting PI4KIIIα in the RNA replication complex--hijacks PI4P metabolism, ultimately resulting in a markedly altered subcellular distribution of the PI4KIIIα product.
Alternative splicing of terminal exons increases transcript and protein diversity. How physiological and pathological stimuli regulate the choice between alternative terminal exons is, however, ...largely unknown. Here, we show that Brahma (BRM), the ATPase subunit of the hSWI/SNF chromatin-remodeling complex interacts with BRCA1/BARD1, which ubiquitinates the 50 kDa subunit of the 3' end processing factor CstF. This results in the inhibition of transcript cleavage at the proximal poly(A) site and a shift towards inclusion of the distal terminal exon. Upon oxidative stress, BRM is depleted, cleavage inhibition is released, and inclusion of the proximal last exon is favoored. Our findings elucidate a novel regulatory mechanism, distinct from the modulation of transcription elongation by BRM that controls alternative splicing of internal exons.
The effective and regular remote monitoring of agricultural activity is not always possible in developing countries because the access to cloud-based geospatial analysis platforms or expensive ...high-resolution satellite images are not always available. Herein, using paid high-resolution satellite images first and then free medium-resolution satellite images, we aimed to develop a cost-free, affordable method for regularly mapping the spatial distribution of sicklebush (Dichrostachys cinerea), an archetypal allochthonous invasive plant in Cuba that is becoming impossible to control owing to its rapid growth in areas planted with sugar cane in the Trinidad-Valle de los Ingenios area (Cuba), a UNESCO World Heritage Site. Two types of images were used (WorldView-2 and Landsat-8); these were subjected to supervised classification, with accuracy values of 88.7% and 93.7%, respectively. Vegetation cover was first derived from the purchased WorldView-2 image, and this information was then used as the training field to obtain spectral signatures from the Landsat-8 free image so that Landsat images may be regularly used to monitor D. cinerea infestations. The results obtained in the spatial distribution map for sicklebush in the Landsat-8 images had an overall reliability of 93.7% and a Kappa coefficient reliability of 91.9%. These values indicate high confidence in the results, which suggests that sicklebush has invaded 52.7% of the total 46,807.26-ha area of the Trinidad-Valle de los Ingenios. This process proved extremely effective and demonstrated the benefits of using high-resolution spatial images from which information can be transferred to free satellite images with a larger pixel size.
The quality assessment of tropical soils and sediments is still under discussion, with efforts being made on the part of governmental agencies to establish reference values. Energy dispersive X-ray ...fluorescence (EDXRF) is a potential analytical technique for quantifying diverse chemical elements in geological material without chemical treatment, primarily when it is performed at an appropriate metrological level. In this work, analytical curves were obtained by means of the analysis of geological reference materials (RMs), which allowed for the researchers to draw a comparison among the sources of analytical uncertainty. After having determined the quality assurance of the analytical procedure, the EDXRF method was applied to determine chemical elements in soils from the state of Pernambuco, Brazil. The regression coefficients of the analytical curves used to determine Al, Ca, Fe, K, Mg, Mn, Ni, Pb, Si, Sr, Ti, and Zn were higher than 0.99. The quality of the analytical procedure was demonstrated at a 95% confidence level, in which the estimated analytical uncertainties agreed with those from the RM’s certificates of analysis. The analysis of diverse geological samples from Pernambuco indicated higher concentrations of Ni and Zn in sugarcane, with maximum values of 41 mg kg
− 1
and 118 mg kg
− 1
, respectively, and agricultural areas (41 mg kg
− 1
and 127 mg kg
− 1
, respectively). The trace element Sr was mainly enriched in urban soils with values of 400 mg kg
− 1
. According to the results, the EDXRF method was successfully implemented, providing some chemical tracers for the quality assessment of tropical soils and sediments.
One of the main natural uranium deposits in Brazil is located in the municipality of Espinharas, in the State of Paraíba. This area may present high levels of natural radioactivity due to the ...presence of these radionuclides. Since this is a populated area, there is need for a radioecological dosimetry assessment to investigate the possible risks to the population. Based on this problem, the objective of this study was to estimate the environmental effective dose outdoors in inhabited areas influenced by the uranium deposit, using the specific activities of equivalent uranium, equivalent thorium and 40K and conversion factors. The environmental assessment was carried using gamma spectroscopy in sixty-two points within the municipality, with a high-resolution gamma spectrometer with HPGe semiconductor detector and Be window. The results obtained ranged from 0.01 to 19.11 mSv y−1, with an average of 2.64 mSv y−1. These levels are, on average, 23 times higher than UNSCEAR reference levels and up to 273 times the reference value of the earth's crust for primordial radionuclides. Therefore, given the high radioactivity levels found, we conclude that there is need for further investigation to evaluate the levels of radioactivity in indoor environments, which will reflect more closely the risks of the local population.

Paraquat (PQ) is a neurotoxic herbicide that induces superoxide formation. Although it is known that its toxic properties are linked to ROS production, the cellular response to PQ is still poorly ...understood. We reported previously that treatment with PQ induced genome-wide changes in pre-mRNA splicing. Here, we investigated the molecular mechanism underlying PQ-induced pre-mRNA splicing alterations. We show that PQ treatment leads to the phosphorylation and nuclear accumulation of SRPK2, a member of the family of serine/arginine (SR) protein-specific kinases. Concomitantly, we observed increased phosphorylation of SR proteins. Site-specific mutagenesis identified a single serine residue that is necessary and sufficient for nuclear localization of SRPK2. Transfection of a phosphomimetic mutant modified splice site selection of the E1A minigene splicing reporter similar to PQ-treatment. Finally, we found that PQ induces DNA damage and vice versa that genotoxic treatments are also able to promote SRPK2 phosphorylation and nuclear localization. Consistent with these observations, treatment with PQ, cisplatin or γ-radiation promote changes in the splicing pattern of genes involved in DNA repair, cell cycle control, and apoptosis. Altogether, our findings reveal a novel regulatory mechanism that connects PQ to the DNA damage response and to the modulation of alternative splicing via SRPK2 phosphorylation.
