Invasive plants have tremendous potential to enrich native food webs by subsidizing net primary productivity. Here, we explored how a potential food subsidy, seeds produced by the aggressive invader ...cheatgrass (Bromus tectorum), is utilized by an important guild of native consumers--granivorous small mammals--in the Great Basin Desert, USA. In a series of field experiments we examined 1) how cheatgrass invasion affects the density and biomass of seed rain at the ecosystem-level; 2) how seed resources from cheatgrass numerically affect granivorous small mammals; and 3) how the food preferences of native granivores might mediate the trophic integration of cheatgrass seeds. Relative to native productivity, cheatgrass invasion increased the density and biomass of seed rain by over 2000% (P < 0.01) and 3500% (P < 0.01), respectively. However, granivorous small mammals in native communities showed no positive response in abundance, richness, or diversity to experimental additions of cheatgrass seeds over one year. This lack of response correlated with a distinct preference for seeds from native grasses over seeds from cheatgrass. Our experiments demonstrate that increased primary productivity associated with exotic plant invasions may not necessarily subsidize consumers at higher trophic levels. In this context, cheatgrass invasion could disrupt native food webs by providing less-preferred resources that fail to enrich higher trophic levels.
Insects have an open circulatory system in which the heart pumps blood (hemolymph) into the body cavity, where it directly bathes the internal organs and epidermis. The blood contains free and ...tissue-bound immune cells that function in the inflammatory response. Here, we use live imaging of transgenic Drosophila larvae with fluorescently labeled blood cells (hemocytes) to investigate the circulatory dynamics of larval blood cells and their response to tissue injury. We find that, under normal conditions, the free cells rapidly circulate, whereas the tissue-bound cells are sessile. After epidermal wounding, tissue-bound cells around the wound site remain sessile and unresponsive, whereas circulating cells are rapidly recruited to the site of damage by adhesive capture. After capture, these cells distribute across the wound, appear phagocytically active, and are subsequently released back into circulation by the healing epidermis. The results demonstrate that circulating cells function as a surveillance system that monitors larval tissues for damage, and that adhesive capture, an important mechanism of recruitment of circulating cells to inflammatory sites in vertebrates, is shared by insects and vertebrates despite the vastly different architectures of their circulatory systems.
The lateral hypothalamic area (LHA) lies at the intersection of multiple neural and humoral systems and orchestrates fundamental aspects of behavior. Two neuronal cell types found in the LHA are ...defined by their expression of hypocretin/orexin (Hcrt/Ox) and melanin-concentrating hormone (MCH) and are both important regulators of arousal, feeding, and metabolism. Conflicting evidence suggests that these cell populations have a more complex signaling repertoire than previously appreciated, particularly in regard to their coexpression of other neuropeptides and the machinery for the synthesis and release of GABA and glutamate. Here, we undertook a single-cell expression profiling approach to decipher the neurochemical phenotype, and heterogeneity therein, of Hcrt/Ox and MCH neurons. In transgenic mouse lines, we used single-cell quantitative polymerase chain reaction (qPCR) to quantify the expression of 48 key genes, which include neuropeptides, fast neurotransmitter components, and other key markers, which revealed unexpected neurochemical diversity. We found that single MCH and Hcrt/Ox neurons express transcripts for multiple neuropeptides and markers of both excitatory and inhibitory fast neurotransmission. Virtually all MCH and approximately half of the Hcrt/Ox neurons sampled express both the machinery for glutamate release and GABA synthesis in the absence of a vesicular GABA release pathway. Furthermore, we found that this profile is characteristic of a subpopulation of LHA glutamatergic neurons but contrasts with a broad population of LHA GABAergic neurons. Identifying the neurochemical diversity of Hcrt/Ox and MCH neurons will further our understanding of how these populations modulate postsynaptic excitability through multiple signaling mechanisms and coordinate diverse behavioral outputs.
The size of animal populations fluctuates with number of births, rate of immigration, rate of emigration, and number of deaths. For many ungulate populations, adult female survival is the most ...important factor influencing population growth. Therefore, increased understanding of survival and causes of mortality for adult females is fundamental for conservation and management. The objectives of our study were to quantify survival rates of female elk (Cervus canadensis) and determine cause-specific mortality. We predicted that hunter harvest would be the leading cause of mortality. Further, we predicted that hunters would harvest animals that were in prime age (2–9 yr) and in better condition than elk predated by mountain lions (Puma concolor). From 2015 to 2017, we captured 376 female elk in central Utah, USA. We assessed body size and condition of captured elk, fitted each animal with a global positioning system-collar, and determined cause of death when we received mortality signals. We estimated survival using Kaplan-Meier estimates and Cox proportional hazard models within an Akaike’s Information Criterion model selection framework to identify covariates that influenced survival. We analyzed differences in size and condition measurements between harvested elk and predated elk using analysis of variance tests. Our best model indicated consistent survival across years; mean survival was 78.3±3.5% (SE) including hunter harvest and 95.5±1.7% without hunter harvest. In decreasing order of importance, elk mortality occurred from hunter harvest (21.2%), mountain lion predation (3.7%), depredation removal (0.5%), automobile collision (0.3%), disease (0.3%), complications during calving (0.3%), and those characterized as undetermined (1.3%). Neck circumference and body length were negatively associated with survival, suggesting that larger animals in good condition had lower survival as a result of hunter harvest. Individuals that died because of cougar predation were smaller and had less loin muscle than the average animal. Hunters removed large, healthy, prime-aged females, individuals that likely have a greater effect on population growth than elk lost to other predators. If the proportion of larger, healthy females in the population begins to decline, hunting practices may require adjustment because hunters may be removing individuals with the greatest reproductive value.
The basal forebrain (BF) plays crucial roles in arousal, attention, and memory, and its impairment is associated with a variety of cognitive deficits. The BF consists of cholinergic, GABAergic, and ...glutamatergic neurons. Electrical or optogenetic stimulation of BF cholinergic neurons enhances cortical processing and behavioral performance, but the natural activity of these cells during behavior is only beginning to be characterized. Even less is known about GABAergic and glutamatergic neurons. Here, we performed microendoscopic calcium imaging of BF neurons as mice engaged in spontaneous behaviors in their home cages (innate) or performed a go/no-go auditory discrimination task (learned). Cholinergic neurons were consistently excited during movement, including running and licking, but GABAergic and glutamatergic neurons exhibited diverse responses. All cell types were activated by overt punishment, either inside or outside of the discrimination task. These findings reveal functional similarities and distinctions between BF cell types during both spontaneous and task-related behaviors.
Environmental factors often limit plant establishment and survival through increased seedling mortality. Understanding plant growth and the causes of mortality can be helpful in developing solutions ...that enhance seeding success and improve restoration monitoring. The purpose of our research was to assess the efficacy of time lapse and motion sensing cameras for monitoring seedling height, density, and fate. We conducted this study in a salt desert shrub community in northwest Utah, USA. In spring 2017, we placed 28 cameras in fenced and unfenced plots seeded with bottlebrush squirreltail and collected hourly images of the seedlings’ development for the initial four months post-seeding. The seedling attributes were recorded in-field and compared with camera images to determine accuracy and reliability. We found that the optimal period for capturing imagery occurred near the sun’s zenith when shadows were minimized. We were able to detect both the timing of the plant emergence, plant height, density, growth rate, and seedling death. The average seedling height and density were underestimated by 14% and 30% between the camera and field estimates, respectively. We recognize that it could be beneficial to adjust for the effect of the date. The reduced seedling density improved the measurement accuracy through a lower visual obscurity. Managers can utilize remote cameras to effectively measure vegetation that can provide an insight into environmental influences.
Gene silencing with siRNA nanoparticles (si-NPs) is promising but still clinically unrealized for inhibition of tumor driver genes. Ternary si-NPs containing siRNA, a single block NP core-forming ...polymer poly(2-(dimethylamino)ethyl methacrylate)-co-(butyl methacrylate) (DMAEMA-co-BMA, 50B), and an NP surface-forming diblock polymer 20 kDa poly(ethylene glycol)-block-50B (20kPEG-50B) have the potential to improve silencing activity in tumors due to the participation of both 50B and 20kPEG-50B in siRNA electrostatic loading and endosome disruptive activity. Functionally, single block 50B provides more potent endosomolytic activity, while 20kPEG-50B colloidally stabilizes the si-NPs. Here, we systematically explored the role of the molecular weight (MW) of the core polymer and of the core:surface polymer ratio on ternary si-NP performance. A library of ternary si-NPs was formulated with variation in the MW of the 50B polymer and in the ratio of the core and surface forming polymeric components. Increasing 50B core polymer MW and ratio improved si-NP in vitro gene silencing potency, endosome disruptive activity, and stability, but these features also correlated with cytotoxicity. Concomitant optimization of 50B size and ratio resulted in the identification of lead ternary si-NPs 50B4-DP100, 50B8-DP100, and 50B12-DP25, with potent activity and minimal toxicity. Following intravenous treatment in vivo, all lead si-NPs displayed negligible toxicological effects and enhanced pharmacokinetics and tumor gene silencing relative to more canonical binary si-NPs. Critically, a single 1 mg/kg intravenous injection of 50B8-DP100 si-NPs silenced the tumor driver gene Rictor at the protein level by 80% in an orthotopic breast tumor model. 50B8-DP100 si-NPs delivering siRictor were assessed for therapeutic efficacy in an orthotopic HCC70 mammary tumor model. This formulation significantly inhibited tumor growth compared to siControl-NP treatment. 50B8-DP100 si-NPs were also evaluated for safety and were well-tolerated following a multi-dose treatment scheme. This work provides new insight on ternary si-NP structure-function relationships and identifies core polymer optimization strategies that can yield safe si-NP formulations with potent oncogene silencing.

Parturition timing has long been a topic of interest in ungulate research. However, few studies have examined parturition timing at fine scale (e.g., <1 day). Predator activity and environmental ...conditions can vary considerably with diel timing, which may result in selective pressure for parturition to occur during diel times that maximize the likelihood of neonate survival. We monitored parturition events and early‐life survival of elk (Cervus canadensis) and mule deer (Odocoileus hemionus) in Utah, USA to better understand diel timing of parturition in temperate ungulates. Diel timing of parturition was moderately synchronous among conspecifics and influenced by environmental variables on the date of parturition. For elk, parturition events were most common during the morning crepuscular period and generally occurred later (i.e., closer to 12:00) when a relatively large proportion of the moon was illuminated. For mule deer, parturition events were most common during the diurnal period and generally occurred later (i.e., closer to 15:00) on cold, wet dates. Diel timing of parturition did not influence neonate survival, but larger datasets may be required to verify the apparent lack of influence. Although additional work could evaluate alternative variables that might affect parturition timing, our data provide an improved and finer scale understanding of reproductive ecology and phenology in ungulates.

Two-partner secretion systems of gram-negative organisms are utilized in adherence, invasion, and biofilm formation. The HrpAB proteins of Neisseria meningitidis are members of a two-partner ...secretion system, and HrpA is established as being important to adherence and intracellular escape. This study set out to determine the expression pattern of members of the hrpBAS putative operon and to find a functional role for the HrpA protein. The upregulation of these genes was found in situations of anaerobiosis and cell contact. These observations prompted the study of the function of HrpA in biofilms on human bronchial epithelial cells. HrpA mutants in encapsulated and unencapsulated NMB strains demonstrated biofilm growth equivalent to that of the wild-type strain at 6 h but a decreased ability to form biofilms at 48 h. Biofilms formed by hrpA mutants for 48 h on collagen-coated coverslips demonstrated significant reductions compared to those of wild-type strains. Taken together, these observations imply a role for HrpA in the biofilm structure. Further analysis demonstrated the presence of HrpA on the surface of the bacterium.
