The aim of this study is to evaluate the efficacy of Baccharis trimera infusion on high-fat diet-induced metabolic disorders in mice and macrophages activation. This study evaluated obesity, insulin ...resistance, dyslipidemia and hepatic steatosis induced by a high-fat diet in Swiss mice. Cellular parameters in macrophages, such as cell viability (MTT), the production and release of nitric oxide (NO) and hydrogen peroxide (H2O2), cell spreading, cell adhesion and phagocytosis were determined. Our results showed that treatment with B. trimera prevented the mentioned conditions, except for the production of hydrogen peroxide. B. trimera prevented the development of obesity and associated comorbidities, as well as activation of macrophages. In conclusion, B. trimera is able to prevent obesity and metabolic disorders and macrophages activation, minimizing inflammation and validating the popular use of this plant tea.
Computational drug repositioning methods have emerged as an attractive and effective solution to find new candidates for existing therapies, reducing the time and cost of drug development. ...Repositioning methods based on biomedical knowledge graphs typically offer useful supporting biological evidence. This evidence is based on reasoning chains or subgraphs that connect a drug to a disease prediction. However, there are no databases of drug mechanisms that can be used to train and evaluate such methods. Here, we introduce the Drug Mechanism Database (DrugMechDB), a manually curated database that describes drug mechanisms as paths through a knowledge graph. DrugMechDB integrates a diverse range of authoritative free-text resources to describe 4,583 drug indications with 32,249 relationships, representing 14 major biological scales. DrugMechDB can be employed as a benchmark dataset for assessing computational drug repositioning models or as a valuable resource for training such models.

. The collection of fish and water was conducted in three distinct stations (S1, S2, S3) in an international protected area in northeast Brazil. The histological alterations index was used to assess changes in the gills and kidneys at a histological level. In addition, micronucleus and nuclear abnormalities tests were employed to examine damage to the genetic material. According to the frequency of Micronucleus, nuclear injuries, and the histopathology of gills and kidneys, it can be inferred that station S1 had the highest impact on the fish, causing significant harm. Based on our findings, the organisms residing in Lake Açu are adversely affected by the poor environmental conditions. This raises doubts about the effectiveness of the Protection Area by Humid Zone of International Interest in Northeast Brazil.

in Wistar rats. At first, a morphoanatomical characterization of
leaves and stems was performed. Then, a purified infusion (ethanol-soluble fraction obtained from
ESAP) was obtained followed by its chemical profile elucidation. Furthermore, an acute toxicity test was performed, and the acute and prolonged diuretic and hypotensive effects were also evaluated in Wistar rats. Finally, the vasodilatory responses of ESAP in mesenteric vascular beds were investigated. The main secondary metabolites identified from ESAP were O-glycosylated flavonoids, chlorogenic acids, and phenylpropanoic acid derivatives. ESAP did not promote any toxic effects in female rats nor increased urinary excretion in male rats after a single exposure. However, ESAP significantly reduced renal elimination of sodium, potassium, and chloride after prolonged treatment. An ESAP highest dose promoted significant acute hypotension without affecting blood pressure levels after prolonged use. Furthermore, its cardiovascular effects seem to be related with the calcium-activated potassium channel activation in resistance vessels.
The geographic structure of Y-chromosome variability has been analyzed in native populations of South America, through use of the high-frequency Native American haplogroup defined by the
DYS199-T ...allele and six Y-chromosome–linked microsatellites (
DYS19, DYS389A, DYS389B, DYS390, DYS391, and
DYS393), analyzed in 236 individuals. The following pattern of within- and among-population variability emerges from the analysis of microsatellite data: (1) the Andean populations exhibit significantly higher levels of within-population variability than do the eastern populations of South America; (2) the spatial-autocorrelation analysis suggests a significant geographic structure of Y-chromosome genetic variability in South America, although a typical evolutionary pattern could not be categorically identified; and (3) genetic-distance analyses and the analysis of molecular variance suggest greater homogeneity between Andean populations than between non-Andean ones. On the basis of these results, we propose a model for the evolution of the male lineages of South Amerindians that involves differential patterns of genetic drift and gene flow. In the western part of the continent, which is associated with the Andean area, populations have relatively large effective sizes and gene-flow levels among them, which has created a trend toward homogenization of the gene pool. On the other hand, eastern populations—settled in the Amazonian region, the central Brazilian plateau, and the Chaco region—have exhibited higher rates of genetic drift and lower levels of gene flow, with a resulting trend toward genetic differentiation. This model is consistent with the linguistic and cultural diversity of South Amerindians, the environmental heterogeneity of the continent, and the available paleoecological data.
ABSTRACT This study evaluated the effects of chromium yeast (Cr yeast) and two apparent metabolizable energy (AME) levels on productive performance, egg quality, and lipid and carbohydrate metabolism ...in laying hens. A total of 192 Bovans White laying hens at 47 weeks of age were randomly assigned to eight dietary treatments (six replicates each) in a 4 × 2 factorial arrangement: four levels of supplemental Cr (0, 0.2, 0.4, and 0.8 ppm as Cr yeast) and two AME levels (2,780 and 2,900 kcal AME kg−1). No significant effect of Cr yeast supplementation on feed intake, egg weight, egg production, intact eggs, egg mass, feed conversion ratio, or egg quality was observed. Egg quality parameters and Cr content in the yolk were not affected by dietary treatments. Plasma glucose and lipid levels were not influenced by either Cr yeast or AME levels used in this study. However, Cr yeast supplementation improved yolk percentage and hepatic glycogen content. The inclusion of Cr yeast at 0.2 and 0.4 ppm induced the highest hepatic glycogen content with the energy levels 2,900 and 2,780 kcal AME kg−1, respectively. Laying hens fed 2,900 kcal AME kg−1 showed the highest abdominal fat. The results observed in the present study support the hypothesis that the lack of positive effects of Cr yeast supplementation on production performance and egg quality may be related to good management practices, as the birds were not subjected to stressful conditions.
Abstract
The Open Targets Platform integrates evidence from genetics, genomics, transcriptomics, drugs, animal models and scientific literature to score and rank target-disease associations for drug ...target identification. The associations are displayed in an intuitive user interface (https://www.targetvalidation.org), and are available through a REST-API (https://api.opentargets.io/v3/platform/docs/swagger-ui) and a bulk download (https://www.targetvalidation.org/downloads/data). In addition to target-disease associations, we also aggregate and display data at the target and disease levels to aid target prioritisation. Since our first publication two years ago, we have made eight releases, added new data sources for target-disease associations, started including causal genetic variants from non genome-wide targeted arrays, added new target and disease annotations, launched new visualisations and improved existing ones and released a new web tool for batch search of up to 200 targets. We have a new URL for the Open Targets Platform REST-API, new REST endpoints and also removed the need for authorisation for API fair use. Here, we present the latest developments of the Open Targets Platform, expanding the evidence and target-disease associations with new and improved data sources, refining data quality, enhancing website usability, and increasing our user base with our training workshops, user support, social media and bioinformatics forum engagement.

First, an anatomical study of the
leaves was performed. Then, the extract (ESCS) was obtained and submitted to phytochemical analysis. Female rats were treated with a single dose of ESCS (2000 mg/kg) to assess acute toxicity. Other groups of female rats received an atherogenic diet for 60 days. After 30 days, the animals were treated orally with ESCS (30 and 300 mg/kg), rosuvastatin (5 mg/kg), or vehicle once daily for 30 days. Serum lipids oxidized low-density lipoprotein, soluble adhesion molecules, interleukins 1β and 6, and markers of renal and liver function were measured. Renal function, blood pressure, electrocardiography, and vascular reactivity were also evaluated. Arteries, heart, liver, and kidney were also collected to evaluate the tissue redox state and histopathological analysis.
Prolonged treatment with ESCS induces significant hypolipidemic and antioxidant effects, that prevent endothelial dysfunction and modulated the local inflammatory process, reducing the evolution of the atherosclerotic disease.
