Tidal wetlands, such as tidal marshes and mangroves, are hotspots for carbon sequestration. The preservation of organic matter (OM) is a critical process by which tidal wetlands exert influence over ...the global carbon cycle and at the same time gain elevation to keep pace with sea-level rise (SLR). The present study assessed the effects of temperature and relative sea level on the decomposition rate and stabilization of OM in tidal wetlands worldwide, utilizing commercially available standardized litter. While effects on decomposition rate per se were minor, we show strong negative effects of temperature and relative sea level on stabilization, as based on the fraction of labile, rapidly hydrolyzable OM that becomes stabilized during deployment. Across study sites, OM stabilization was 29 % lower in low, more frequently flooded vs. high, less frequently flooded zones. Stabilization declined by ∼ 75 % over the studied temperature gradient from 10.9 to 28.5 ∘C. Additionally, data from the Plum Island long-term ecological research site in Massachusetts, USA, show a pronounced reduction in OM stabilization by > 70 % in response to simulated coastal eutrophication, confirming the potentially high sensitivity of OM stabilization to global change. We therefore provide evidence that rising temperature, accelerated SLR, and coastal eutrophication may decrease the future capacity of tidal wetlands to sequester carbon by affecting the initial transformations of recent OM inputs to soil OM.
Realistic appraisal of paleoclimatic information obtained from a particular location requires accurate knowledge of its paleolatitude defined relative to the Earth's spin-axis. This is crucial to, ...among others, correctly assess the amount of solar energy received at a location at the moment of sediment deposition. The paleolatitude of an arbitrary location can in principle be reconstructed from tectonic plate reconstructions that (1) restore the relative motions between plates based on (marine) magnetic anomalies, and (2) reconstruct all plates relative to the spin axis using a paleomagnetic reference frame based on a global apparent polar wander path. Whereas many studies do employ high-quality relative plate reconstructions, the necessity of using a paleomagnetic reference frame for climate studies rather than a mantle reference frame appears under-appreciated. In this paper, we briefly summarize the theory of plate tectonic reconstructions and their reference frames tailored towards applications of paleoclimate reconstruction, and show that using a mantle reference frame, which defines plate positions relative to the mantle, instead of a paleomagnetic reference frame may introduce errors in paleolatitude of more than 15° (>1500 km). This is because mantle reference frames cannot constrain, or are specifically corrected for the effects of true polar wander. We used the latest, state-of-the-art plate reconstructions to build a global plate circuit, and developed an online, user-friendly paleolatitude calculator for the last 200 million years by placing this plate circuit in three widely used global apparent polar wander paths. As a novelty, this calculator adds error bars to paleolatitude estimates that can be incorporated in climate modeling. The calculator is available at www.paleolatitude.org. We illustrate the use of the paleolatitude calculator by showing how an apparent wide spread in Eocene sea surface temperatures of southern high latitudes may be in part explained by a much wider paleolatitudinal distribution of sites than previously assumed.

Clinical measures in multiple sclerosis (MS) face limitations that may be overcome by utilising smartphone keyboard interactions acquired continuously and remotely during regular typing.
...Objective:
The aim of this study was to determine the reliability and validity of keystroke dynamics to assess clinical aspects of MS.
Methods:
In total, 102 MS patients and 24 controls were included in this observational study. Keyboard interactions were obtained with the Neurokeys keyboard app. Eight timing-related keystroke features were assessed for reliability with intraclass correlation coefficients (ICCs); construct validity by analysing group differences (in fatigue, gadolinium-enhancing lesions on magnetic resonance imaging (MRI), and patients vs controls); and concurrent validity by correlating with disability measures.
Results:
Reliability was moderate in two (ICC = 0.601 and 0.742) and good to excellent in the remaining six features (ICC = 0.760–0.965). Patients had significantly higher keystroke latencies than controls. Latency between key presses correlated the highest with Expanded Disability Status Scale (r = 0.407) and latency between key releases with Nine-Hole Peg Test and Symbol Digit Modalities Test (ρ = 0.503 and r = −0.553, respectively), ps < 0.001.
Conclusion:
Keystroke dynamics were reliable, distinguished patients and controls, and were associated with clinical disability measures. Consequently, keystroke dynamics are a promising valid surrogate marker for clinical disability in MS.

Objectives:
The aim of this study was to determine the reliability, construct and concurrent validity of a smartphone-adapted Symbol Digit Modalities Test (sSDMT).
Methods:
During a 28-day follow-up, 102 patients with MS and 24 healthy controls (HC) used the MS sherpa® app to perform the sSDMT every 3 days on their own smartphone. Patients performed the Brief International Cognitive Assessment for MS at baseline. Test–retest reliability (intraclass correlation coefficients, ICC), construct validity (group analyses between cognitively impaired (CI), cognitively preserved (CP) and HC for differences) and concurrent validity (correlation coefficients) were assessed.
Results:
Patients with MS and HC completed an average of 23.2 (SD = 10.0) and 18.3 (SD = 10.2) sSDMT, respectively. sSDMT demonstrated high test–retest reliability (ICCs > 0.8) with a smallest detectable change of 7 points. sSDMT scores were different between CI patients, CP patients and HC (all ps < 0.05). sSDMT correlated modestly with the clinical SDMT (highest r = 0.690), verbal (highest r = 0.516) and visuospatial memory (highest r = 0.599).
Conclusion:
Self-administered smartphone-adapted SDMT scores were reliable and different between patients who were CI, CP and HC and demonstrated concurrent validity in assessing information processing speed.
Dune development along highly dynamic land–sea boundaries is the result of interaction between vegetation and dune size with sedimentation and erosion processes. Disentangling the contribution of ...vegetation characteristics from that of dune size would improve predictions of nebkha dune development under a changing climate, but has proven difficult due to the scarcity of spatially continuous monitoring data. This study explored the contributions of vegetation and dune size to dune development for locations differing in shelter from the sea. We monitored a natural nebkha dune field of 8 ha, along the coast of the island Texel, the Netherlands, for 1 year using an unmanned aerial vehicle (UAV) with camera. After constructing a digital surface model and orthomosaic we derived for each dune (1) vegetation characteristics (species composition, vegetation density, and maximum vegetation height), (2) dune size (dune volume, area, and maximum height), (3) degree of shelter (proximity to other nebkha dunes and the sheltering by the foredune). Changes in dune volume over summer and winter were related to vegetation, dune size and degree of shelter. We found that a positive change in dune volume (dune growth) was linearly related to initial dune volume over summer but not over winter. Big dunes accumulated more sand than small dunes due to their larger surface area. Exposed dunes increased more in volume (0.81 % per dune per week) than sheltered dunes (0.2 % per dune per week) over summer, while the opposite occurred over winter. Vegetation characteristics did not significantly affect dune growth in summer, but did significantly affect dune growth in winter. Over winter, dunes dominated by Ammophila arenaria, a grass species with high vegetation density throughout the year, increased more in volume than dunes dominated by Elytrigia juncea, a grass species with lower vegetation density (0.43 vs. 0.42 (m3 m−3) week−1). The effect of species was irrespective of dune size or distance to the sea. Our results show that dune growth in summer is mainly determined by dune size, whereas in winter dune growth was determined by vegetation type. In our study area the growth of exposed dunes was likely restricted by storm erosion, whereas growth of sheltered dunes was restricted by sand supply. Our results can be used to improve models predicting coastal dune development.
Abstract Background In stroke and multiple sclerosis patients, gait is frequently hampered by a reduced ability to push-off with the ankle caused by weakness of the plantar-flexor muscles. To enhance ...ankle push-off and to decrease the high energy cost of walking, spring-like carbon-composite Ankle Foot Orthoses are frequently prescribed. However, it is unknown what Ankle Foot Orthoses stiffness should be used to obtain the most efficient gait. The aim of this simulation study was to gain insights into the effect of variation in Ankle Foot Orthosis stiffness on the amount of energy stored in the Ankle Foot Orthosis and the energy cost of walking. Methods We developed a two-dimensional forward-dynamic walking model with a passive spring at the ankle representing the Ankle Foot Orthosis and two constant torques at the hip for propulsion. We varied Ankle Foot Orthosis stiffness while keeping speed and step length constant. Findings We found an optimal stiffness, at which the energy delivered at the hip joint was minimal. Energy cost decreased with increasing energy storage in the ankle foot orthosis, but the most efficient gait did not occur with maximal energy storage. With maximum storage, push-off occurred too late to reduce the impact of the contralateral leg with the floor. Maximum return prior to foot strike was also suboptimal, as push-off occurred too early and its effects were subsequently counteracted by gravity. The optimal Ankle Foot Orthosis stiffness resulted in significant push-off timed just prior to foot strike and led to greater ankle plantar-flexion velocity just before contralateral foot strike. Interpretation Our results suggest that patient energy cost might be reduced by the proper choice of Ankle Foot Orthosis stiffness.
Micromagnetic tomography aims at reconstructing large numbers of individual magnetizations of magnetic particles from combining high‐resolution magnetic scanning techniques with micro X‐ray computed ...tomography (microCT). Previous work demonstrated that dipole moments can be robustly inferred, and mathematical analysis showed that the potential field of each particle is uniquely determined. Here, we describe a mathematical procedure to recover higher orders of the magnetic potential of the individual magnetic particles in terms of their spherical harmonic expansions (SHE). We test this approach on data from scanning superconducting quantum interference device microscopy and microCT of a reference sample. For particles with high signal‐to‐noise ratio of the magnetic scan we demonstrate that SHE up to order n = 3 can be robustly recovered. This additional level of detail restricts the possible internal magnetization structures of the particles and provides valuable rock magnetic information with respect to their stability and reliability as paleomagnetic remanence carriers. Micromagnetic tomography therefore enables a new approach for detailed rock magnetic studies on large ensembles of individual particles.

Neurodegeneration, rather than inflammation, plays a key role in the progressive phase of multiple sclerosis (MS). Current disease modifying treatment options for people with progressive MS (PMS) do ...not specifically target neurodegeneration. Preliminary evidence suggests that exercise therapy might have neuroprotective effects. However, neuroprotective effect studies of exercise interventions in PMS are scarce and the possible mode of action underlying neuroprotective effects of exercise are unknown and need to be elucidated. The main aim of this phase II trial is to assess whether progressive resistance training (PRT) and high intensity interval training (HIIT), can slow down neurodegeneration in people with PMS.
In a single-blinded phase II clinical trial with an extended baseline period, 60 people with PMS will be randomly assigned to PRT or HIIT. The participants should have had a relapse onset of MS with confirmed disease progression, however still ambulatory. The duration of the study is 48 weeks, consisting of 16 weeks baseline period (no intervention), 16 weeks intervention and 16 weeks follow-up. Patient-tailored training will be performed 3 times per week for one hour in groups, led by an experienced physiotherapist. The primary outcome measure is neurodegeneration, measured as whole brain atrophy on magnetic resonance imaging (MRI). Secondary outcome parameters will include other biomarkers associated with neurodegeneration (i.e. regional brain atrophy, lesion load, white matter integrity, resting state functional connectivity, blood biomarkers (brain derived neurotrophic factor (BDNF) and serum neurofilament light (sNFL)), patient functioning (physical and cognitive) and cardiovascular risk factors.
Besides the primary outcome measures, this study will examine a large variety of biomarkers associated with neurodegeneration after an exercise intervention. Combining outcome parameters may help to elucidate the mode of action underlying neuroprotective effects of exercise.

Abstract The mechanical characteristics of ankle foot orthoses (AFOs), such as the stiffness and neutral angle around the ankle and metatarsal-phalangeal (MTP) joints, are rarely quantified. ...Paradoxically, it is expected that these characteristics determine the function of the AFO in pathological gait. Therefore a device to determine these AFO characteristics named BRUCE was designed based on multidisciplinary consensus. The design is based on a replicated human leg that is manually driven and continuously registers joint configuration and force exerted by the AFO onto the device. From this information, neutral angles and stiffnesses around the ankle and MTP joints are determined using a linear fit. The reliability of the stiffnesses and neutral angles was studied by repeatedly measuring the mechanical characteristics of four different AFOs, and evaluating the inter-session, intra-session, and inter-observer errors. The reliability study revealed that ankle and MTP stiffness could be measured with very high reliability (ICC = 0.98–1.00). Ankle and MTP neutral angles showed reasonable reliability (ICC = 0.79–0.92). Measurement error in the neutral angles could mainly be attributed to the difference in testers. With a fixed tester excellent reliability was obtained (ICC = 0.99–0.99). The results derived using BRUCE can help to gain insight into the role of the mechanical characteristics of AFOs in correcting pathological gait. Objective information of AFO characteristics is expected to lead to a better founded prescription of AFOs, resulting in optimal functional benefit for the patient.
Genetic monitoring using noninvasive samples provides a complement or alternative to traditional population monitoring methods. However, next‐generation sequencing approaches to monitoring typically ...require high quality DNA and the use of noninvasive samples (e.g., scat) is often challenged by poor DNA quality and contamination by nontarget species. One promising solution is a highly multiplexed sequencing approach called genotyping‐in‐thousands by sequencing (GT‐seq), which can enable cost‐efficient genomics‐based monitoring for populations based on noninvasively collected samples. Here, we develop and validate a GT‐seq panel of 324 single nucleotide polymorphisms (SNPs) optimized for genotyping of polar bears based on DNA from noninvasively collected faecal samples. We demonstrate (1) successful GT‐seq genotyping of DNA from a range of sample sources, including successful genotyping (>50% loci) of 62.9% of noninvasively collected faecal samples determined to contain polar bear DNA; and (2) that we can reliably differentiate individuals, ascertain sex, assess relatedness, and resolve population structure of Canadian polar bear subpopulations based on a GT‐seq panel of 324 SNPs. Our GT‐seq data reveal spatial‐genetic patterns similar to previous polar bear studies but at lesser cost per sample and through use of noninvasively collected samples, indicating the potential of this approach for population monitoring. This GT‐seq panel provides the foundation for a noninvasive toolkit for polar bear monitoring and can contribute to community‐based programmes – a framework which may serve as a model for wildlife conservation and management for species worldwide.
