Abstract
We analyze the physical conditions, chemical composition, and other properties of the photoionized Herbig–Haro (HH) object HH 204 through Very Large Telescope echelle spectroscopy and Hubble ...Space Telescope imaging. We kinematically isolate the high-velocity emission of HH 204 from the emission of the background nebula and study the subarcsecond distribution of physical conditions and ionic abundances across the HH object. We find that low- and intermediate-ionization emission arises exclusively from gas at photoionization equilibrium temperatures, whereas the weak high-ionization emission from HH 204 shows a significant contribution from higher-temperature shock-excited gas. We separately derive the ionic abundances of HH 204, the emission of the Orion Nebula, and the fainter diffuse blue layer. In HH 204, the O
+
abundance determined from collisional excited lines matches the one based on recombination lines, while the O
2+
abundance is very low, so that the oxygen abundance discrepancy is zero. The ionic abundances of Ni and Fe in HH 204 have similar ionization and depletion patterns, with total abundances that are a factor of 3.5 higher than in the rest of the Orion Nebula due to dust destruction in the bow shock. We show that a failure to resolve the kinematic components in our spectra would lead to significant error in the determination of chemical abundances (for instance, a 40% underestimate of O), mainly due to incorrect estimation of the electron density.
Rhizobia are recognized to establish N2-fixing symbiotic interactions with legume plants. Bradyrhizobium japonicum, the symbiont of soybeans, can denitrify and grow under free-living conditions with ...nitrate (NO3 (-)) or nitrite (NO2 (-)) as sole nitrogen source. Unlike related bacteria that assimilate NO3 (-), genes encoding the assimilatory NO3 (-) reductase (nasC) and NO2 (-) reductase (nirA) in B. japonicum are located at distinct chromosomal loci. The nasC gene is located with genes encoding an ABC-type NO3 (-) transporter, a major facilitator family NO3 (-)/NO2 (-) transporter (NarK), flavoprotein (Flp) and single-domain haemoglobin (termed Bjgb). However, nirA clusters with genes for a NO3 (-)/NO2 (-)-responsive regulator (NasS-NasT). In the present study, we demonstrate NasC and NirA are both key for NO3 (-) assimilation and that growth with NO3 (-), but not NO2 (-) requires flp, implying Flp may function as electron donor to NasC. In addition, bjgb and flp encode a nitric oxide (NO) detoxification system that functions to mitigate cytotoxic NO formed as a by-product of NO3 (-) assimilation. Additional experiments reveal NasT is required for NO3 (-)-responsive expression of the narK-bjgb-flp-nasC transcriptional unit and the nirA gene and that NasS is also involved in the regulatory control of this novel bipartite assimilatory NO3 (-)/NO2 (-) reductase pathway.

Abstract
Examination of emission lines in high-velocity-resolution optical spectra of the Orion Nebula confirms that the velocity component on the red wing of the main ionization front emission line ...is due to backscattering in the Photon Dominated Region. This scattered light component has a weak wavelength dependence that is consistent with either general interstellar medium particles or particles in the foreground of the Orion Nebula Cluster. An anomalous line-broadening component that has been known for 60+ years is characterized in unprecedented detail. Although this extra broadening may be due to turbulence along the line of sight of our spectra, we explore the possibility that it is due to Alfvén waves in conditions where the ratio of magnetic and thermal energies are about equal and constant throughout the ionized gas.
Reducible oxides have been shown to greatly improve the activity of water gas shift (WGS) catalysts. The precise mechanism for this effect is a matter of intense debate, but the dissociation of water ...is generally considered to be the key step in the reaction. We present here a study of the water activation on oxygen vacancies at the support as part of the mechanism of the WGS reaction on Pt supported on pure and gallium-doped ceria. Doping the ceria with gallium allows tuning the vacancies in the support while maintaining constant the metal dispersion. An inverse relationship was found between the catalytic activity to WGS and the amount of oxygen vacancies. In situ time-resolved X-ray diffraction, mass spectrometry, and diffuse reflectance infrared spectroscopy (DRIFT) showed that the oxygen vacancy filling by water is always fast in either Pt/CeO2 or Pt/CeGa. DFT calculation provides molecular insights to understand the pathway of water reaction with vacancies at the metal–oxide interface sites. Our results suggest that the activation of the water molecule in the WGS mechanism is not the rate-limiting step in these systems. Concentration-modulation spectroscopy in DRIFT mode under WGS reaction conditions allows the selective detection of key reaction intermediates, a monodentate formate (HCOO) and carboxylate (CO2 δ−) species, which suggests the prevalence of a carboxyl (HOCO) mechanism activated at the oxide–metal interface of the catalyst.
Geographic isolation substantially contributes to species endemism on oceanic islands when speciation involves the colonisation of a new island. However, less is understood about the drivers of ...speciation within islands. What is lacking is a general understanding of the geographic scale of gene flow limitation within islands, and thus the spatial scale and drivers of geographical speciation within insular contexts. Using a community of beetle species, we show that when dispersal ability and climate tolerance are restricted, microclimatic variation over distances of only a few kilometres can maintain strong geographic isolation extending back several millions of years. Further to this, we demonstrate congruent diversification with gene flow across species, mediated by Quaternary climate oscillations that have facilitated a dynamic of isolation and secondary contact. The unprecedented scale of parallel species responses to a common environmental driver for evolutionary change has profound consequences for understanding past and future species responses to climate variation.

The compatible solute trehalose is involved in the osmostress response of Rhizobium etli, the microsymbiont of Phaseolus vulgaris. In this work, we reconstructed trehalose metabolism in R. etli, and ...investigated its role in cellular adaptation and survival to heat and desiccation stress under free living conditions.
Besides trehalose as major compatible solute, R. etli CE3 also accumulated glutamate and, if present in the medium, mannitol. Putative genes for trehalose synthesis (otsAB/treS/treZY), uptake (aglEFGK/thuEFGK) and degradation (thuAB/treC) were scattered among the chromosome and plasmids p42a, p42c, p42e, and p42f, and in some instances found redundant. Two copies of the otsA gene, encoding trehalose-6-P-synthase, were located in the chromosome (otsAch) and plasmid p42a (otsAa), and the latter seemed to be acquired by horizontal transfer. High temperature alone did not influence growth of R. etli, but a combination of high temperature and osmotic stress was more deleterious for growth than osmotic stress alone. Although high temperature induced some trehalose synthesis by R. etli, trehalose biosynthesis was mainly triggered by osmotic stress. However, an otsAch mutant, unable to synthesize trehalose in minimal medium, showed impaired growth at high temperature, suggesting that trehalose plays a role in thermoprotection of R. etli. Desiccation tolerance by R. etli wild type cells was dependent of high trehalose production by osmotic pre-conditioned cells. Cells of the mutant strain otsAch showed ca. 3-fold lower survival levels than the wild type strain after drying, and a null viability after 4 days storage.
Our findings suggest a beneficial effect of osmotic stress in R. etli tolerance to desiccation, and an important role of trehalose on the response of R. etli to high temperature and desiccation stress.
Virtual fieldtrips in palaeontology and geology offer an expanded environment to make field sites more accessible for a broader audience. Based on extensive experience working in upper Neogene ...deposits in the province of Huelva, on the western edge of the Guadalquivir Basin, we have selected 10 sites for their regional palaeontological-geological interest (geosites). By applying geoinformatic tools, we have produced a virtual 3D tour of the georeferenced geosites, integrating multiple thematic digital layers (including geological maps, topographical maps, a digital terrain model and orthophotos). Each stop in the tour contains descriptive and graphic elements that can be viewed in free virtual globes (e.g., Google Earth), combined with diagrams, photographs and information sheets that quantitatively assess the cultural tourism, scientific and educational value of the geosite. From this digital 3D geodatabase, we propose a virtual flight itinerary that can be displayed in compatible video formats and using new technologies, such as smartphones, tablets or iPads.
Flood estimation and flood management have traditionally been the domain of hydrologists, water resources engineers and statisticians, and disciplinary approaches abound. Dominant views have been ...shaped; one example is the catchment perspective: floods are formed and influenced by the interaction of local, catchment-specific characteristics, such as meteorology, topography and geology. These traditional views have been beneficial, but they have a narrow framing. In this paper we contrast traditional views with broader perspectives that are emerging from an improved understanding of the climatic context of floods. We come to the following conclusions: (1) extending the traditional system boundaries (local catchment, recent decades, hydrological/hydraulic processes) opens up exciting possibilities for better understanding and improved tools for flood risk assessment and management. (2) Statistical approaches in flood estimation need to be complemented by the search for the causal mechanisms and dominant processes in the atmosphere, catchment and river system that leave their fingerprints on flood characteristics. (3) Natural climate variability leads to time-varying flood characteristics, and this variation may be partially quantifiable and predictable, with the perspective of dynamic, climate-informed flood risk management. (4) Efforts are needed to fully account for factors that contribute to changes in all three risk components (hazard, exposure, vulnerability) and to better understand the interactions between society and floods. (5) Given the global scale and societal importance, we call for the organization of an international multidisciplinary collaboration and data-sharing initiative to further understand the links between climate and flooding and to advance flood research.
Bradyrhizobium japonicum RegSR regulatory proteins belong to the family of two-component regulatory systems, and orthologs are present in many Proteobacteria where they globally control gene ...expression mostly in a redox-responsive manner. In this work, we have performed a transcriptional profiling of wild-type and regR mutant cells grown under anoxic denitrifying conditions. The comparative analyses of wild-type and regR strains revealed that almost 620 genes induced in the wild type under denitrifying conditions were regulated (directly or indirectly) by RegR, pointing out the important role of this protein as a global regulator of denitrification. Genes controlled by RegR included nor and nos structural genes encoding nitric oxide and nitrous oxide reductase, respectively, genes encoding electron transport proteins such as cycA (blr7544) or cy2 (bll2388), and genes involved in nitric oxide detoxification (blr2806-09) and copper homeostasis (copCAB), as well as two regulatory genes (bll3466, bll4130). Purified RegR interacted with the promoters of norC (blr3214), nosR (blr0314), a fixK-like gene (bll3466), and bll4130, which encodes a LysR-type regulator. By using fluorescently labeled oligonucleotide extension (FLOE), we were able to identify two transcriptional start sites located at about 35 (P1) and 22 (P2) bp upstream of the putative translational start codon of norC. P1 matched with the previously mapped 5'end of norC mRNA which we demonstrate in this work to be under FixK2 control. P2 is a start site modulated by RegR and specific for anoxic conditions. Moreover, qRT-PCR experiments, expression studies with a norC-lacZ fusion, and heme c-staining analyses revealed that anoxia and nitrate are required for RegR-dependent induction of nor genes, and that this control is independent of the sensor protein RegS.
