The quasar 3C 454.3 is a blazar known for its rapid and violent outbursts seen across the electromagnetic spectrum. Using γ-ray, X-ray, multiband optical, and very-long-baseline interferometric data ...we investigate the nature of two such events that occurred in 2013 and 2014 accompanied by strong variations in optical polarization, including a ∼230° electric vector position angle (EVPA) rotation. Our results suggest that a single disturbance was responsible for both flaring events. We interpret the disturbance as a shock propagating down the jet. Under this interpretation the 2013 flare originated most likely due to changes in the viewing angle caused by perhaps a bent or helical trajectory of the shock upstream of the radio core. The 2014 flare and optical polarization behavior are the result of the shock exiting the 43 GHz radio core, suggesting that shock crossings are one of the possible mechanisms for EVPA rotations.

The Coronavirus disease 2019 (COVID-19) is spreading around the world, representing a global pandemic. In this context, governments from around the world suspended almost all education, industry and business activities, alongside restricting the movement of people. Nevertheless, during this period, the activity of the law enforcement and forensic investigators never stopped.
At present, guidelines regarding forensic autopsies of SARS-CoV-2 virus-positive cases and the handling of potentially infected biological samples are available in literature. However, less attention has been given to the development of specific adjustments to the existing crime scene investigation protocols and procedures for this exceptional time.
This manuscript aims to share the methods and strategies adopted for the investigation of high priority criminal cases during the pandemic. Furthermore, other pandemic-related processes are critically explored, in order to propose adjustments for any forensic services to be prepared to face similar challenges in the future.
The overall goal of this manuscript is to provide a summary of the main measures and the procedures developed to make the operations possible, while safeguarding the technicians in the field and the activity in the forensic laboratory. In order to minimize the risk of infection for personnel, adjustments to the standard practice have been proposed for each of the different phases of crime scene management, i.e. CSI call policy, equipment preparation, working groups, procedure at the scene, chain of custody and analyses of the evidence at the forensic lab.
As this is a current study, based on limited cases and limited sources in the literature, changes and updates to the indications provided in this paper may be needed in the near future, according to new virological data epidemiological trends.
After several years of quiescence, the blazar CTA 102 underwent an exceptional outburst in 2012 September–October. The flare was tracked from γ-ray to near-infrared (NIR) frequencies, including Fermi ...and Swift data as well as photometric and polarimetric data from several observatories. An intensive Glast-Agile support programme of the Whole Earth Blazar Telescope (GASP–WEBT) collaboration campaign in optical and NIR bands, with an addition of previously unpublished archival data and extension through fall 2015, allows comparison of this outburst with the previous activity period of this blazar in 2004–2005. We find remarkable similarity between the optical and γ-ray behaviour of CTA 102 during the outburst, with a time lag between the two light curves of ≈1 h, indicative of cospatiality of the optical and γ-ray emission regions. The relation between the γ-ray and optical fluxes is consistent with the synchrotron self-Compton (SSC) mechanism, with a quadratic dependence of the SSC γ-ray flux on the synchrotron optical flux evident in the post-outburst stage. However, the γ-ray/optical relationship is linear during the outburst; we attribute this to changes in the Doppler factor. A strong harder-when-brighter spectral dependence is seen both the in γ-ray and optical non-thermal emission. This hardening can be explained by convexity of the UV–NIR spectrum that moves to higher frequencies owing to an increased Doppler shift as the viewing angle decreases during the outburst stage. The overall pattern of Stokes parameter variations agrees with a model of a radiating blob or shock wave that moves along a helical path down the jet.
The emergence of Russia as a major grain exporter is not only crucial for the world commercial agriculture and food security, but also for the country’s economy. Here we examine the past-to-future ...thermal suitability for winter wheat (Triticum aestivum, L. 1753) cultivation over Russia and compare it with the recent trends of wheat yields and harvested area. The analyses use a multi-model ensemble median of the most updated bias-corrected outputs from five CMIP5 Earth System Models (1950–2099) under two representative concentration pathways (RCP 4.5 and RCP 8.5) and the Era-Interim dataset (1979–2016).
Our results show that the thermal suitability has increased by ∼10 Mha per decade since 1980. Consistently, winter wheat yields and harvested area have also increased over the last decade by ∼0.5 t/ha and ∼4 Mha, respectively. Moreover, a potential for the Russian wheat sector may still be exploited if we consider the abandoned land (∼27 Mha) after the collapse of the Soviet Union. Our results also show that the increase in heat availability and the reduction of the frost constraint will likely move the thermal suitability toward the north-western and the Far East regions. Conversely, increases of extreme heat events are projected in the southern regions of Russia, which currently represent the most productive and intensively managed wheat cultivation area. Our findings imply both opportunities and risks for the Russian wheat sector that calls for sustainable and farsighted land management strategies to comprehensively face the consequences of global warming.
We have performed new medium resolution spectroscopy, optical and near-infrared photometry to monitor the variability of the asymptotic giant branch (AGB) carbon star V 381 Lac. Our observations ...revealed rapid and deep changes in the spectrum and extreme variability in the optical and near-infrared bands. Most notably we observed the change of Na I D lines from deep absorption to emission, and the progressive growing of the NII doublet 6548-6584 A emission, strongly related to the simultaneous photometric fading. V381 Lac occupies regions of Two Micron All-Sky Survey and Wide-field Infrared Survey Explorer colour-colour diagrams typical of stars with dust formation in the envelope. The general framework emerging from the observations of V381 Lac is that of a cool AGB carbon star undergoing episodes of high mass ejection and severe occultation of the stellar photosphere reminiscent of those characterizing the RCB phenomenon. Comparing the spectral energy distribution obtained with the theoretical model for AGB evolution with dust in the circumstellar envelope, we can identify V381 Lac as the descendant of a star of initial mass ~2 M..., in the final AGB phases, evolved into a carbon star by repeated Third Dredge Up episodes. According to our model, the star is moderately obscured (t10 ~ 0.22) by dust, mainly formed by amorphous carbon (~80 per cent) and SiC (~20 per cent), with dust grain dimensions around ~0.2 and 0.08 mu m, respectively. Objectives
To evaluate the serum symmetric dimethylarginine (SDMA) and serum creatinine concentrations in a population of hypothyroid dogs at the time of diagnosis and after treatment.
Materials and ...Methods
Serum SDMA and serum creatinine were measured in serum samples of 24 healthy dogs and 24 hypothyroid dogs, at the time of diagnosis (T0) and after supplementation with levothyroxine (T1).
Results
The mean SDMA concentrations (reference intervals RI <18 μg/dL and <14 μg/dL depending on the source) were 11.7 ± 3.5 μg/dL, 13.8 ± 3.1 μg/dL and 11.83 ± 2.87 μg/dL in healthy dogs, and in the hypothyroid dogs at T0 and T1, respectively. The SDMA concentrations were higher in the hypothyroid dogs at T0 in comparison with the healthy dogs. Of the hypothyroid dogs, 1 out of 24 had an SDMA concentration above 18 μg/dL and 12 out of 24 above 14 μg/dL at T0. At T1, none of the hypothyroid dogs had SDMA concentrations above 18 μg/dL and two of them had SDMA concentrations above 14 μg/dL. The serum creatinine concentration was higher in the hypothyroid dogs at T0 as compared to the healthy dogs. At T0, 8 out of 24 hypothyroid dogs had serum creatinine concentrations above the RI (>1.4 mg/dL). In all but one dog, serum creatinine normalised after treatment.
Clinical Significance
The SDMA and serum creatinine concentrations were higher in hypothyroid dogs at diagnosis as compared to healthy dogs. Serum creatinine concentrations were increased in one‐third of the hypothyroid dogs and in the majority of cases normalised after levothyroxine supplementation. SDMA concentrations were rarely above the upper limit of the RI when the higest (<18 μg/dL) cut‐off was employed. The diagnostic accuracy of SDMA in dogs with thyroid dysfunction requires additional evaluation.
Microfluidic flow assays (MFA) that measure shear dependent platelet function have potential clinical applications in the diagnosis and treatment of bleeding and thrombotic disorders. As a step ...towards clinical application, the objective of this study was to measure how phenotypic and genetic factors, as well as experimental conditions, affect the variability of platelet accumulation on type 1 collagen within a MFA. Whole blood was perfused over type 1 fibrillar collagen at wall shear rates of 150, 300, 750 and 1500 s⁻¹ through four independent channels with a height of 50 µm and a width of 500 µm. The accumulation of platelets was characterized by the lag time to 1% platelet surface coverage (Lag(T)), the rate of platelet accumulation (V(PLT)), and platelet surface coverage (SC). A cohort of normal donors was tested and the results were correlated to plasma von Willebrand factor (VWF) levels, platelet count, hematocrit, sex, and collagen receptors genotypes. VWF levels were the strongest determinant of platelet accumulation. VWF levels were positively correlated to V(PLT) and SC at all wall shear rates. A longer Lag(T) for platelet accumulation at arterial shear rates compared to venous shear rates was attributed to the time required for plasma proteins to adsorb to collagen. There was no association between platelet accumulation and hematocrit or platelet count. Individuals with the AG genotype of the GP6 gene had lower platelet accumulation than individuals with the AA genotype at 150 s⁻¹ and 300 s⁻¹. Recalcified blood collected into sodium citrate and corn trypsin inhibitor (CTI) resulted in diminished platelet accumulation compared to CTI alone, suggesting that citrate irreversibly diminishes platelet function. This study the largest association study of MFA in healthy donors (n = 104) and will likely set up the basis for the determination of the normal range of platelet responses in this type of assay.
Variants in GBA are associated with Lewy Body (LB) pathology. We investigated whether variants in other lysosomal storage disorder (LSD) genes also contribute to disease pathogenesis.
We performed a ...genetic analysis of four LSD genes including GBA, HEXA, SMPD1, and MCOLN1 in 231 brain autopsies. Brain autopsies included neuropathologically defined LBD without Alzheimer Disease (AD) changes (n = 59), AD without significant LB pathology (n = 71), Alzheimer disease and lewy body variant (ADLBV) (n = 68), and control brains without LB or AD neuropathology (n = 33). Sequencing of HEXA, SMPD1, MCOLN1 and GBA followed by 'gene wise' genetic association analysis was performed. To determine the functional effect, a biochemical analysis of GBA in a subset of brains was also performed. GCase activity was measured in a subset of brain samples (n = 64) that included LBD brains, with or without GBA mutations, and control brains. A lipidomic analysis was also performed in brain autopsies (n = 67) which included LBD (n = 34), ADLBV (n = 3), AD (n = 4), PD (n = 9) and control brains (n = 17), comparing GBA mutation carriers to non-carriers.
In a 'gene-wise' analysis, variants in GBA, SMPD1 and MCOLN1 were significantly associated with LB pathology (p range: 0.03-4.14 x10(-5)). Overall, the mean levels of GCase activity were significantly lower in GBA mutation carriers compared to non-carriers (p<0.001). A significant increase and accumulation of several species for the lipid classes, ceramides and sphingolipids, was observed in LBD brains carrying GBA mutations compared to controls (p range: p<0.05-p<0.01).
Our study indicates that variants in GBA, SMPD1 and MCOLN1 are associated with LB pathology. Biochemical data comparing GBA mutation carrier to non-carriers support these findings, which have important implications for biomarker development and therapeutic strategies.
Clinical evidence suggests that individuals with factor VIII (FVIII) deficiency (hemophilia A) are protected against venous thrombosis, but treatment with recombinant proteins can increase their risk ...for thrombosis. In this study we examined the dynamics of thrombus formation in individuals with hemophilia A and their response to replacement and bypass therapies under venous flow conditions. Fibrin and platelet accumulation were measured in microfluidic flow assays on a TF-rich surface at a shear rate of 100 s⁻¹. Thrombin generation was calculated with a computational spatial-temporal model of thrombus formation. Mild FVIII deficiencies (5-30% normal levels) could support fibrin fiber formation, while severe (<1%) and moderate (1-5%) deficiencies could not. Based on these experimental observations, computational calculations estimate an average thrombin concentration of ∼10 nM is necessary to support fibrin formation under flow. There was no difference in fibrin formation between severe and moderate deficiencies, but platelet aggregate size was significantly larger for moderate deficiencies. Computational calculations estimate that the local thrombin concentration in moderate deficiencies is high enough to induce platelet activation (>1 nM), but too low to support fibrin formation (<10 nM). In the absence of platelets, fibrin formation was not supported even at normal FVIII levels, suggesting platelet adhesion is necessary for fibrin formation. Individuals treated by replacement therapy, recombinant FVIII, showed normalized fibrin formation. Individuals treated with bypass therapy, recombinant FVIIa, had a reduced lag time in fibrin formation, as well as elevated fibrin accumulation compared to healthy controls. Treatment of rFVIIa, but not rFVIII, resulted in significant changes in fibrin dynamics that could lead to a prothrombotic state.
