The BL Lacertae object OJ 287 (z = 0.306) has unique double-peaked optical outbursts every ∼12 yr, and it presents one of the best cases for a small-separation binary supermassive black hole (SMBH) ...system, with an extremely massive primary . However, the host galaxy is unresolved or only marginally detected in all optical studies so far, indicating a large deviation from the bulge mass-SMBH mass relation. We have obtained deep, high spatial resolution i-band and K-band images of OJ 287 when the target was in a low state, which enable us to detect the host galaxy. We find the broadband photometry of the host to be consistent with an early-type galaxy with MR = −22.5 and MK = −25.2, placing it in the middle of the host galaxy luminosity distribution of BL Lacertae objects. The central supermassive black hole is clearly overmassive for a host galaxy of that luminosity, but not unprecedented, given some recent findings of other "overmassive" black holes in nearby galaxies.
Gene therapy and its role in the medical field have evolved drastically in recent decades. Studies aim to define DNA-based medicine as well as encourage innovation and the further development of ...novel approaches. Gene therapy has been established as an alternative approach to treat a variety of diseases. Its range of mechanistic applicability is wide; gene therapy has the capacity to address the symptoms of disease, the body's ability to fight disease, and in some cases has the ability to cure disease, making it a more attractive intervention than some traditional approaches to treatment (i.e., medicine and surgery). Such versatility also suggests gene therapy has the potential to address a greater number of indications than conventional treatments. Many DNA-based therapies have shown promise in clinical trials, and several have been approved for use in humans. Whereas current treatment regimens for chronic disease often require frequent dosing, DNA-based therapies can produce robust and durable expression of therapeutic genes with fewer treatments. This benefit encourages the application of DNA-based gene therapy to manage chronic diseases, an area where improving efficiency of current treatments is urgent. Here, we provide an overview of two DNA-based gene therapies as well as their delivery methods: adeno associated virus (AAV)-based gene therapy and plasmid DNA (pDNA)-based gene therapy. We will focus on how these therapies have already been utilized to improve treatment of chronic disease, as well as how current literature supports the expansion of these therapies to treat additional chronic indications in the future.

This paper presents the results of a combined spectroscopic and photometric study of 20 contact binary systems: HV Aqr, OO Aql, FI Boo, TX Cnc, OT Cnc, EE Cet, RW Com, KR Com, V401 Cyg, ...V345 Gem, AK Her, V502 Oph, V566 Oph, V2612 Oph, V1363 Ori, V351 Peg, V357 Peg, Y Sex, V1123 Tau, and W UMa, which was conducted in the frame of the W UMa Project. Together with 51 already covered by the project and an additional 67 in the existing literature, these systems bring the total number of contact binaries with known combined spectroscopic and photometric solutions to 138. It was found that mass, radius, and luminosity of the components follow certain relations along the MS and new empirical power relations are extracted. We found that 30 per cent of the systems in the current sample show extreme values in their parameters, expressed in their mass ratio or fill-out factor. This study shows that, among the contact binary systems studied, some have an extremely low mass ratio (q < 0.1) or an ultrashort orbital period (Porb < 0.25 d), which are expected to show evidence of mass transfer progress. The evolutionary status of these components is discussed with the aid of correlation diagrams and their physical and orbital parameters compared to those in the entire sample of known contact binaries. The existence of very short orbital periods confirms the very slow nature of the merging process, which seems to explain why their components still exist as MS stars in contact configurations even after several Gyr of evolution.
ABSTRACT OJ 287 is a quasi-periodic quasar with roughly 12 year optical cycles. It displays prominent outbursts that are predictable in a binary black hole model. The model predicted a major optical ...outburst in 2015 December. We found that the outburst did occur within the expected time range, peaking on 2015 December 5 at magnitude 12.9 in the optical R-band. Based on Swift/XRT satellite measurements and optical polarization data, we find that it included a major thermal component. Its timing provides an accurate estimate for the spin of the primary black hole, . The present outburst also confirms the established general relativistic properties of the system such as the loss of orbital energy to gravitational radiation at the 2% accuracy level, and it opens up the possibility of testing the black hole no-hair theorem with 10% accuracy during the present decade.
Structure of interphase cell nuclei remains dynamic and can undergo various changes of shape and organisation, in health and disease. The double-membraned envelope that separates nuclear genetic ...material from the rest of the cell frequently includes deep, branching tubular invaginations that form a dynamic nucleoplasmic reticulum (NR). This study addresses mechanisms by which NR can form in interphase nuclei. We present a combination of Nanoscale Secondary Ion Mass Spectrometry (NanoSIMS) approach and light microscopy techniques to follow formation of NR by using pulse-chase experiments to examine protein and lipid delivery to nascent NR in cultured cells. Lamina protein incorporation was assessed using precursor accumulation (for lamin A) or a MAPLE3 photoconvertible tag (for lamin B1) and membrane phospholipid incorporation using stable isotope labelling with deuterated precursors followed by high resolution NanoSIMS. In all three cases, nascent molecules were selectively incorporated into newly forming NR tubules; thus strongly suggesting that NR formation is a regulated process involving a focal assembly machine, rather than simple physical perturbation of a pre-existing nuclear envelope.

ABSTRACT
Flaring episodes in blazars represent one of the most violent processes observed in extra-galactic objects. Studies of such events shed light on the energetics of the physical processes ...occurring in the innermost regions of blazars, which cannot otherwise be resolved by any current instruments. In this work, we present some of the largest and most rapid flares captured in the optical band in the blazars 3C 279, OJ 49, S4 0954+658, TXS 1156+295, and PG 1553+113. The source flux was observed to increase by nearly ten times within a time-scale of a few weeks. We applied several methods of time series analysis and symmetry analysis. Moreover, we also performed searches for periodicity in the light curves of 3C 279, OJ 49 and PG 1553+113 using the Lomb–Scargle method and found plausible indications of quasi-periodic oscillations (QPOs). In particular, the 33- and 22-day periods found in 3C 279, i.e. a 3:2 ratio, are intriguing. These violent events might originate from magnetohydrodynamical instabilities near the base of the jets, triggered by processes modulated by the magnetic field of the accretion disc. We present a qualitative treatment as the possible explanation for the observed large amplitude flux changes in both the source-intrinsic and source-extrinsic scenarios.
The nuclei of cells may exhibit invaginations of the nuclear envelope under a variety of conditions. These invaginations form a branched network termed the nucleoplasmic reticulum (NR), which may be ...found in cells in pathological and physiological conditions. While an extensive NR is a hallmark of cellular senescence and shows associations with some cancers, very little is known about the formation of NR in physiological conditions, despite the presence of extensive nuclear invaginations in some cell types such as endometrial cells. Here we show that in these cells the NR is formed in response to reproductive hormones. We demonstrate that oestrogen and progesterone are sufficient to induce NR formation and that this process is reversible without cell division upon removal of the hormonal stimulus. Nascent lamins and phospholipids are incorporated into the invaginations suggesting that there is a dedicated machinery for its formation. The induction of NR in endometrial cells offers a new model to study NR formation and function in physiological conditions.
Tests of Einstein's general theory of relativity have mostly been carried out in weak gravitational fields where the space-time curvature effects are first-order deviations from Newton's theory. ...Binary pulsars provide a means of probing the strong gravitational field around a neutron star, but strong-field effects may be best tested in systems containing black holes. Here we report such a test in a close binary system of two candidate black holes in the quasar OJ 287. This quasar shows quasi-periodic optical outbursts at 12-year intervals, with two outburst peaks per interval. The latest outburst occurred in September 2007, within a day of the time predicted by the binary black-hole model and general relativity. The observations confirm the binary nature of the system and also provide evidence for the loss of orbital energy in agreement (within 10 per cent) with the emission of gravitational waves from the system. In the absence of gravitational wave emission the outburst would have happened 20 days later.
The nuclear lamina can bind and sequester transcription factors (TFs), a function lost if the lamina is abnormal, with missing or mutant lamin proteins. We now show that TF sequestration is not ...all-or-nothing, but a dynamic physiological response to external signals. We show that the binding of the ubiquitous TF, Oct-1, to lamin B1 was reversed under conditions of cellular stress caused, inter alia, by the chemical methylating agent methylmethanesulfonate (MMS). A search for lamin B1 post-translational modifications that might mediate changes in Oct-1 binding using kinase inhibitors uncovered a role for c-Jun N-terminal kinase (JNK). Phosphoproteomic and site-directed mutagenesis analyses of lamin B1 isolated from control and MMS-treated nuclei identified T575 as a JNK site phosphorylated after stress. A new phospho-T575 specific anti-peptide antibody confirmed increased interphase cellular T575 phosphorylation after cell exposure to certain stress conditions, enabling us to conclude that lamin B1 acts as an interphase kinase target, releasing Oct-1 to execute a protective response to stress.
Abstract
We present results from an observational campaign on the close binary system 2MASS J16211735+4412541 and a preliminary model based on the photometric data gathered during the quiescent and ...outburst levels. The modeling, done with the Wilson–Devinney code and its improvements, failed to reproduce the observational properties of the system. A secondary minimum obtained within the stellar model that is too shallow, as well as the evidence provided by the spectroscopic observations performed at outburst and quiescence, point toward an accretion disk surrounding one component, likely a white dwarf, as the cause of the outburst. Using a simple disk model, we modeled the observed multicolor light curves taken two (2016 August) and eight (2017 March) months after the outburst. We obtained a reasonable fit to the 2016 August light curves but those from 2017 March cannot be explained with the same parameters. We conclude that J1621 is an eclipsing cataclysmic binary, with an accretion disk still present almost a year after outburst, and not a contact-type system as previously classified. The binary is seen at an inclination of about 84° and there is evidence of changing accretion rates and disk parameters as a result of the outburst. Our results indicate that more cataclysmic variables may be hidden among contact binaries.
