To constrain the behavior of Mg isotopes during deep crustal processes and the Mg isotopic composition of the middle and lower continental crust, 30 composite samples from high-grade metamorphic ...terranes and 18 granulite xenoliths were investigated. The composites derive from eight different high-grade metamorphic terranes in the two largest Archean cratons of China, including 13 TTG gneisses, 5 amphibolites, 4 felsic, 4 intermediate, and 4 mafic granulites. They have variable bulk compositions with SiO2 ranging from 45.7 to 72.5%, representative of the middle crust beneath eastern China. The δ26Mg values of these samples vary from -0.40 to +0.12 ppm, reflecting heterogeneity of their protoliths, which could involve upper crustal sediments. The granulite xenoliths from the Cenozoic Hannuoba basalts also have a diversity of compositions with MgO ranging from 2.95 to 20.2%. These xenoliths equilibrated under high temperatures of 800-950°C, corresponding to depths of the lower continental crust (>30 km). They yield a large δ26Mg variation of -0.76 to -0.24 ppm. The light Mg isotopic compositions likely result from interactions with isotopically light metamorphic fluids, probably carbonate fluids. Together with previously reported data, the average δ26Mg values of the middle and lower continental crusts are estimated to be -0.21±0.07 ppm and -0.26±0.06 ppm, respectively. The bulk continental crust is estimated to have an average δ26Mg of -0.24±0.07 ppm, which is similar to the average of the mantle. The large Mg isotopic variation in the continental crust reflects the combination of several processes, such as continental weathering, involvement of supracrustal materials in the deep crust, and fluid metasomatism.

A size-segregated gas/particle partitioning coefficient KPi was proposed and evaluated in the predicting models on the basis of atmospheric polybrominated diphenyl ether (PBDE) field data comparing ...with the bulk coefficient KP. Results revealed that the characteristics of atmospheric PBDEs in southeast Shanghai rural area were generally consistent with previous investigations, suggesting that this investigation was representative to the present pollution status of atmospheric PBDEs. KPi was generally greater than bulk KP, indicating an overestimate of TSP (the mass concentration of total suspended particles) in the expression of bulk KP. In predicting models, KPi led to a significant shift in regression lines as compared to KP, thus it should be more cautious to investigate sorption mechanisms using the regression lines. The differences between the performances of KPi and KP were helpful to explain some phenomenon in predicting investigations, such as PL0 and KOA models overestimate the particle fractions of PBDEs and the models work better at high temperature than at low temperature. Our findings are important because they enabled an insight into the influence of particle size on predicting models.

Xylanases play a crucial role in the degradation of xylan in both terrestrial and marine environments. The endoxylanase XynB from the marine bacterium
KMM 241 is a modular enzyme comprising a long ...N-terminal domain (NTD) (E44 to T562) with xylan-binding ability and a catalytic domain (CD) (T563 to E912) of glycoside hydrolase family 8 (GH8). In this study, the long NTD is confirmed to contain three different functional regions, which are NTD1 (E44 to D136), NTD2 (Y137 to A193), and NTD3 (L194 to T562). NTD1, mainly composed of eight β-strands, functions as a new type of carbohydrate-binding module (CBM), which has xylan-binding ability but no sequence similarity to any known CBM. NTD2, mainly forming two α-helices, contains one of the α-helices of the catalytic domain's (α/α)
barrel and therefore is essential for the activity of XynB, although it is far away from the catalytic domain in sequence. NTD3, next to the catalytic domain in sequence, is shown to be helpful in maintaining the thermostability of XynB. Thus, XynB represents a kind of xylanase with a new domain architecture. There are four other predicted glycoside hydrolase sequences with the same domain architecture and high sequence identity (≥80%) with XynB, all of which are from marine bacteria. Phylogenetic analysis shows that XynB and these homologs form a new group in GH8, representing a new class of marine bacterial xylanases. Our results shed light on xylanases, especially marine xylanases.

Sugarcane molasses, which is a kind of microbial carbon source, is a viscous by-product of the refining of sugarcane into sugar. However, experiments were designed to ascertain the mechanism and ...kinetics of Cr(VI) reduction with sugarcane molasses without adding microbes in aqueous solution. Results indicated that sugarcane molasses can reduce Cr(VI) to Cr(III) at pH values that range from 2.0 to 6.1 when no bioreduction occurs in the reaction. Furthermore, the reaction mechanism was proven to be that Cr(VI) acts as an electrophile that readily accepts electrons from the phenolic hydroxyl group of plant polyphenol, and it is then reduced to Cr(III) and in the process oxidizes the phenolic hydroxyl group to a quinone. Meanwhile, the reaction could be described by the pseudo-first-order kinetic model with respect to Cr(VI) concentration. The reaction rate constants were 324.2, 65.9, 21.9, and 14.4 h

when pH values were 2.0, 3.5, 5.0, and 6.1, respectively, at 20 °C. The

Introgression of Erianthus arundinaceus has been the focus of several sugarcane breeding programs in the world, because the species has desirable traits such as high biomass production, vigour, ...ratooning ability and good resistance to environmental stresses and disease. In this study four genetic maps were constructed for two intergeneric populations. The first population (BC1) was generated from a cross between an Erianthus/Saccharum hybrid YC96-40 and a commercial sugarcane variety CP84-1198. The second population (BC2) was generated from a cross between YCE01-116, a progeny of the BC1 cross and NJ57-416, a commercial sugarcane cultivar. Markers across both populations were generated using 35 AFLP and 23 SSR primer pairs. A total of 756 and 728 polymorphic markers were scored in the BC1 and BC2 populations, respectively. In the BC1 population, a higher proportion of markers was derived from the Erianthus ancestor than those from the Saccharum ancestor Badila. In the BC2 population, both the number and proportion of markers derived from Erianthus were approximately half of those in the BC1 population. Linkage analysis led to the construction of 38, 57, 36 and 47 linkage groups (LGs) for YC96-40, CP84-1198, YCE01-116, and NJ57-416, encompassing 116, 174, 97 and 159 markers (including single dose, double dose and bi-parental markers), respectively. These LGs could be further placed into four, five, five and six homology groups (HGs), respectively, based on information from multi-allelic SSR markers and repulsion phase linkages detected between LGs. Analysis of repulsion phase linkage indicated that Erianthus behaved like a true autopolyploid.
Bile acid (BA) homeostasis is regulated by the extensive cross-talk between liver and intestine. Many bile-acid-activated signaling pathways have become attractive therapeutic targets for the ...treatment of metabolic disorders. In this study we investigated the regulatory mechanisms of BA in the intestine. We showed that the BA levels in the gallbladder and faeces were significantly increased, whereas serum BA levels decreased in systemic Krüppel-like factor 9 (Klf9) deficiency (Klf9
) mice. These phenotypes were also observed in the intestine-specific Klf9-deleted (Klf9
) mice. In contrast, BA levels in the gallbladder and faeces were reduced, whereas BA levels in the serum were increased in intestinal Klf9 transgenic (Klf9
) mice. By using a combination of biochemical, molecular and functional assays, we revealed that Klf9 promoted the expression of apical sodium-dependent bile acid transporter (Asbt) in the terminal ileum to enhance BA absorption in the intestine. Reabsorbed BA affected liver BA synthetic enzymes by regulating Fgf15 expression. This study has identified a previously neglected transcriptional pathway that regulates BA homeostasis.

Herein we report the discovery of a series of new KDM5A inhibitors. A three-dimensional (3D) structure model of KDM5A jumonji domain was firstly established based on homology ...modeling. Molecular docking-based virtual screening was then performed against commercial chemical databases. A number of hit compounds were retrieved. Further structural optimization and structure–activity relationship (SAR) analysis were carried out to the most active hit compound, 9 (IC50: 2.3μM), which led to the discovery of several new KDM5A inhibitors. Among them, compound 15e is the most potent one with an IC50 value of 0.22μM against KDM5A. This compound showed good selectivity for KDM5A and considerable ability to suppress the demethylation of H3K4me3 in intact cells. Compound 15e could be taken as a good lead compound for further studies.
