To determine if the distribution of transcallosal inhibition (TI) acting on proximal and distal upper extremity muscles is altered in chronic stroke.
We examined thirteen healthy controls and sixteen ...mildly to moderately impaired chronic stroke patients. We used transcranial magnetic stimulation (TMS) to probe TI from the contralesional onto ipsilesional hemisphere (assigned in controls). We recorded the ipsilateral silent period in the paretic biceps (BIC) and first dorsal interosseous (FDI). We measured TI strength, distribution gradient (TI difference between muscles), and motor impairment (Fugl-Meyer Assessment).
Both groups had stronger TI acting on their FDIs than BICs (p < 0.001). However, stroke patients also had stronger TI acting on their BICs than controls (p = 0.034), resulting in a flatter distribution of inhibition (p = 0.028). In patients, stronger FDI inhibition correlated with less hand impairment (p = 0.031); BIC inhibition was not correlated to impairment.
TI is more evenly distributed to the paretic FDI and BIC in chronic stroke. The relative increase in proximal inhibition does not relate to better function, as it does distally.
The results expand our knowledge about segment-specific neurophysiology and its relevance to impairment after stroke.
•Like healthy controls, chronic stroke patients have stronger transcallosal inhibition for a hand muscle than an arm muscle.•In stroke patients, inhibition acting on the arm muscle is also increased relative to healthy controls.•In stroke patients, the strength of transcallosal inhibition is related to better paretic hand function but is unrelated to arm function.

A ...multidisciplinary international Expert Panel was convened to update the clinical practice guideline recommendations informed by a systematic review of the medical literature.
The Expert Panel continues to recommend ER testing of invasive breast cancers by validated immunohistochemistry as the standard for predicting which patients may benefit from endocrine therapy, and no other assays are recommended for this purpose. Breast cancer samples with 1% to 100% of tumor nuclei positive should be interpreted as ER positive. However, the Expert Panel acknowledges that there are limited data on endocrine therapy benefit for cancers with 1% to 10% of cells staining ER positive. Samples with these results should be reported using a new reporting category, ER Low Positive, with a recommended comment. A sample is considered ER negative if < 1% or 0% of tumor cell nuclei are immunoreactive. Additional strategies recommended to promote optimal performance, interpretation, and reporting of cases with an initial low to no ER staining result include establishing a laboratory-specific standard operating procedure describing additional steps used by the laboratory to confirm/adjudicate results. The status of controls should be reported for cases with 0% to 10% staining. Similar principles apply to PgR testing, which is used primarily for prognostic purposes in the setting of an ER-positive cancer. Testing of ductal carcinoma in situ (DCIS) for ER is recommended to determine potential benefit of endocrine therapies to reduce risk of future breast cancer, while testing DCIS for PgR is considered optional. A protein extract was generated from the macroalga
, which was subsequently hydrolysed using the food-grade enzyme papain and angiotensin-converting Enzyme I and renin inhibitory peptides identified ...using a combination of enrichment strategies employing molecular weight cutoff filtration and mass spectrometry analysis. The generated hydrolysates with the most promising in vitro activity were further purified using preparative RP-HPLC and characterised. The 1 kDa hydrolysate (1 kDa-UFH), purified and collected by preparative RP-HPLC at minutes 41‒44 (Fr41‒44), displayed statistically higher ACE-I inhibitory activities ranging from 96.91% to 98.06%. A total of 48 novel peptides were identified from these four fractions by LC-MS/MS. A simulated gastrointestinal digestion of the identified peptide sequences was carried out using in silico enzyme cleavage simulation tools, resulting in 86 peptide sequences that were further assessed for their potential activity, toxicity and allergenicity using multiple predictive approaches. All the peptides obtained in this study were predicted to be non-toxic. However, 28 out of the 86 novel peptides released after the in silico gastrointestinal digestion were identified as potential allergens. The potential allergenicity of these peptides should be further explored to comply with the current labelling regulations in formulated food products containing
protein hydrolysates.
The primary function of dietary protein is to provide amino acids for protein synthesis. However, protein is also a source of latent bioactive peptides or cryptides with potential health benefits ...including the control and regulation of blood pressure. Hypertension or high blood pressure is one of the major, controllable risk factors in the development of cardiovascular disease (CVD), and it is also implicated in the development of myocardial infarction, heart failure, and end-stage diabetes. Cryptides can act on various systems of the body including the circulatory, gastrointestinal (GI), nervous, skeletal, and respiratory systems. A number of studies carried out to date have examined the health benefits of food protein isolates and hydrolysates. This review provides an overview of existing blood pressure regulating peptides and products derived from fish and other protein sources and hydrolysates. It discusses the methods used currently to generate and identify cryptides from these sources and their application in food and pharmaceutical products. It also looks at the current market for protein-derived peptides and peptide-containing products, legislation governing their use, and the future development of research in this area.
We find two chemically distinct populations separated relatively cleanly in the Fe/H-Mg/Fe plane, but also distinguished in other chemical planes, among metal-poor stars (primarily with metallicities ...) observed by the Apache Point Observatory Galactic Evolution Experiment (APOGEE) and analyzed for Data Release 13 (DR13) of the Sloan Digital Sky Survey. These two stellar populations show the most significant differences in their X/Fe ratios for the -elements, C+N, Al, and Ni. In addition to these populations having differing chemistry, the low metallicity high-Mg population (which we denote "the HMg population") exhibits a significant net Galactic rotation, whereas the low-Mg population (or "the LMg population") has halo-like kinematics with little to no net rotation. Based on its properties, the origin of the LMg population is likely an accreted population of stars. The HMg population shows chemistry (and to an extent kinematics) similar to the thick disk, and is likely associated with in situ formation. The distinction between the LMg and HMg populations mimics the differences between the populations of low- and high- halo stars found in previous studies, suggesting that these are samples of the same two populations.
Dry-cured ham by-products have been traditionally used in Mediterranean household cooking of broths and stews. The aim of this work was to evaluate the effect of cooking treatments and in vitro ...gastrointestinal digestion on the antioxidant activity of natural peptides found in bones from Spanish dry-cured hams. The antioxidant activity was tested using five different assays and results demonstrated that cooking using conventional household methods increased the antioxidant activity of ham by-products when assessed using different antioxidant assays with the exception of the ABTS radical scavenging measurement assay. Simulated gastrointestinal digestion showed no significant effect on the antioxidant activity of ham by-products and antioxidant activity decreased when assessed using the ORAC and β-carotene bleaching assays. Analysis by MALDI-TOF MS revealed a considerable breakdown of peptides due to the action of gastrointestinal enzymes, mainly in samples cooked at 100°C for 1h. In addition, 459 peptides derived from 57 proteins were identified and quantified using mass spectrometry in tandem, evidencing that peptides derived from collagen protein were responsible for the differences in antioxidant activities observed between the uncooked and cooked samples after digestion. The results show the potential of dry-cured ham bones as a source of antioxidant peptides that retain their bioactivity after household cooking preparations and gastrointestinal digestion.
