This study was focused on analyzing the effects of dietary inclusion of raw chickpea seed as a replacement of soybean meal as a primary protein source on bone structure in broiler chickens. Broiler ...chickens (n = 160) received in their diet either soybean meal (SBM) or raw chickpea seeds (CPS) as a primary protein source throughout the whole rearing period (n = 80 in each group). On the 42th day randomly selected chickens from each group (n = 8) were slaughtered. Collected tibiotarsus were subjected to examination of the biomechanical characteristics of bone mid-diaphysis, microstructure of the growth plate and articular cartilages; the analysis of mineral content and crystallinity of mineral phase, and the measurements of thermal stability of collagen in hyaline cartilage were also carried out. The inclusion of chickpea seeds resulted in increase of bone osteometric parameters (weight, length and mid-diaphysis cross-sectional area) and mechanical endurance (yield load, ultimate load, stiffness, Young modulus). However, when loads were adjusted to bone shape (yield and ultimate stress) both groups did not differ. Mineral density determined by means of densitometric measurements did not differ between groups, however the detailed analysis revealed the differences in the macro- and microelements composition. The results of FT-IR and XRD analyses showed no effect of diet type on mineral phase crystallinity and hydroxyapatite nanocrystallites size. In trabecular bone, the increase of real bone volume (BV/TV) and number of trabeculae was observed in the CPS group. Total thickness of articular cartilage was the same in both groups, save the transitional zone, which was thicker in the SBM group. The total thickness of the growth plate cartilage was significantly increased in the CPS group. The area of the most intense presence of proteoglycans was wider in the SBM group. The structural analysis of fibrous components of bone revealed the increase of fraction of thin, immature collagen content in articular cartilage, trabeculae and compact bone in the CPS group. The dietary inclusion of CPS affected the thermal stability of collagen, as decrease of net denaturation enthalpy was observed. This study showed a beneficial effect of CPS on the skeletal development, improving the overall bone development and the microarchitecture of cancellous bone. It suggests that CPS can be a promising replacement for SBM in broilers feeding in the aspect of animal welfare related to the development of the skeletal system.
Ni–P and Ni–P–Re coatings with various concentrations of P and Re were obtained on copper substrate by the electroless deposition from succinate baths. The influence of certain bath parameters on the ...alloys composition and the deposition rate is presented, followed by the relation between these process parameters as well as the rhenium addition on the surface morphology, microstructure, phase composition and thermal stability of the deposited films. The conditions for the formation of uniform, smooth and compact alloy films on copper substrate were defined. On the basis of X-ray diffraction analysis it can be stated that the alloy containing less than 5.5 wt.% of phosphorus exhibits fully crystalline microstructure for both studied cases: Ni–P and Ni–P–Re alloys. Differential scanning calorimetry analysis performed for the Ni-based alloy layers with and without rhenium additive and the same level of phosphorus are provided, proving, that the addition of rhenium to the Ni–P coatings results in the increase of the crystallization temperature. The increase of the crystallization temperature from 411 °C to 434 °C was noted for NiP5.0 alloy, where the addition of Re was only of 3.7 wt.%.
The most grievous complication of the COVID-19 is the acute respiratory distress syndrome. A specific, rescue treatment for rapidly deteriorating patients should emerge to improve respiratory ...function and help patients to survive the most challenging period. Drugs used in targeted therapy of pulmonary arterial hypertension (PAH) appears to be suitable for this task and this article describes their potential for treatment of severe cases of COVID-19.
The authors reviewed the following databases for randomized controlled trials, reviews and meta-analyses published up to July 2020: Pubmed, Scopus, Google Scholar, Cochrane Database and ClinicalKey. The authors included every study contributory to the assessment of the potential of drugs used in targeted PAH therapy in treatment of COVID-19.
Endothelin receptor antagonists, phosphodiesterase 5 inhibitors, riociguat and prostacyclin have proven ani-inflammatory effect and reduce pulmonary artery blood pressure, lung oedema and remodelling. Bosentan shows antiviral properties and sildenafil, as well as epoprostenol, inhibits apoptosis of lung epithelial cells. Among patients with lung lesions the decrease of pulmonary blood pressure can lead to increase of ventilation/perfusion mismatch and decrease of blood oxygenation.
Among all assessed drugs bosentan, sildenafil and epoprostenol appear to be most promising and a combination of these drugs should be considered due to synergism. The targeted PAH therapy in treatment of COVID-19 associated ARDS could be a useful tool saving lives of patients with severe SARS-CoV-2 infection, however, its introduction should be investigated and monitored very carefully as it can lead to transient deterioration of patient condition.

The use of biobased plastics is of great importance for many applications. Blending thermoplastic polylactide (PLA) with polyhydroxyalkanoate (PHA) enables the formulation of a more mechanically ...powerful material and this enables tailored biodegradation properties. In this study we demonstrate the 3D printing of a PLA/PHA blend as a potential candidate for biocompatible material applications. The filament for 3D printing consisted of PHA, which contains predominantly 3-hydroxybutyrate units and a small amount of 3-hydroxyvalerate units, as revealed by multistage mass spectrometry (ESI-MSn). This research found that the properties of 3D printed species before and during abiotic degradation are dependent on printing orientation. Furthermore, the 3D printed specimens exhibited good biocompatibility with HEK293 cells, indicating real promise as biological scaffolds for tissue engineering applications.
•Contact time with printer platform alters the thermal and mechanical properties of materials.•Filament arrangement from the algorithm used for printing affects material properties.•Processing conditions lead to different degradation profile of the investigated specimens.•3D printed specimens showed good biocompatibility with HEK293 cells.
Kynurenic acid (KYNA) is the end stage metabolite of tryptophan produced mainly by astrocytes in the central nervous system (CNS). It has neuroprotective activities but can be elevated in the ...neuropsychiatric disorders. Toxic effects of KYNA in the CNS are unknown. The aim of this study was to assess the effect of the subdural KYNA infusion on the spinal cord in adult rats.
A total of 42 healthy adult rats were randomly assigned into six groups and were infused for 7 days with PBS (control) or 0.0002 pmol/min, 0.01 nmol/min, 0.1 nmol/min, 1 nmol/min, and 10 nmol/min of KYNA per 7 days. The effect of KYNA on spinal cord was determined using histological and electron microscopy examination. Myelin oligodendrocyte glycoprotein (MOG) was measured in the blood serum to assess a degree of myelin damage.
In all rats continuous long-lasting subdural KYNA infusion was associated with myelin damage and myelin loss that was increasingly widespread in a dose-depended fashion in peripheral, sub-pial areas. Damage to myelin sheaths was uniquely related to the separation of lamellae at the intraperiod line. The damaged myelin sheaths and areas with complete loss of myelin were associated with limited loss of scattered axons while vast majority of axons in affected areas were morphologically intact. The myelin loss-causing effect of KYNA occurred with no necrosis of oligodendrocytes, with locally severe astrogliosis and no cellular inflammatory response. Additionally, subdural KYNA infusion increased blood MOG concentration. Moreover, the rats infused with the highest doses of KYNA (1 and 10 nmol/min) demonstrated adverse neurological signs including weakness and quadriplegia.
We suggest, that subdural infusion of high dose of KYNA can be used as an experimental tool for the study of mechanisms of myelin damage and regeneration. On the other hand, the administration of low, physiologically relevant doses of KYNA may help to discover the role of KYNA in control of physiological myelination process.
Introduction and objective: The purpose of this paper is a systematic review of articles and research in the context of the relationship between genetic determinants of migraine and thyroid ...dysfunction. Materials and methods: A non-systematic review of the scientific literature was carried out according to the following keywords: PubMed was searched and 76 articles published up to 2023 were analyzed. Description of the state of knowledge: Epidemiological studies confirm the co-occurrence of migraine and thyroid dysfunction, especially hypothyroidism, which are considered to be diseases with a strong genetic background. Thyroid function parameters such as TSH and fT4 are susceptible to genetic influences. However, there is a lack of consistent interpretation of the observational results, which highlights the need for further research. Analysis of gene overlaps and links between headache and thyroid function, based on GWAS data, could shed light on the complex genetic interactions underlying this association. Summary: The genetic associations revealed in the study enhance our comprehension of the intricate relationship between migraine and thyroid dysfunction. These findings not only offer the prospect of developing biomarkers for identifying migraine patients who might benefit from thyroid hormone therapy but also underscore the promising potential of future genetic studies in providing biological insights to advance clinical decision-making in understanding and addressing these interconnected phenomena.
A European Shorthair male cat, neutered, approximately 6 years of age, was presented to the veterinary clinic due to apathy and anorexia. The cat lived mostly outdoors and was fed raw chicken meat. ...After 3 days of diagnostic procedures and symptomatic treatment, respiratory distress and neurological signs developed and progressed into epileptic seizures, followed by respiratory and cardiac arrest within the next 3 days. Post-mortem examination revealed necrotic lesions in the liver, lungs, and intestines. Notably, the brain displayed perivascular infiltration of lymphocytes and histiocytes. Few foci of neuronal necrosis in the brain were also confirmed. Microscopic examination of the remaining internal organs was unremarkable. The A/H5N1 virus infection was confirmed using a one-step real-time reverse transcription polymerase chain reaction (RT-qPCR). The disease caused severe neurological and respiratory signs, evidence of consolidations and the presence of numerous B lines, which were detected on lung ultrasound examination; the postmortem findings and detection of A/H5N1 viral RNA in multiple tissues indicated a generalized A/H5N1 virus infection. Moreover, a multidrug-resistant strain of Enterococcus faecium was isolated in pure culture from several internal organs. The source of infection could be exposure to infected birds or their excrements, as well as contaminated raw poultry meat but, in this case, the source of infection could not be identified.
Data exchange in the multimedia system between the camera and the computer can be realized via the USB communication interface. Applying the USB port requires tuning the port parameters in such ...systems to ensure the quality of the services (multimedia) provided. The tuning of the communication system depends on such selection of port parameters to optimize the imposed criteria. The authors proposed a quality criterion based on the USB isochronous interval. The paper briefly presents the quality measure and the transmission model of the data package. Next, the problem of tuning the USB port was presented based on the selected parameters (MaxPacketSize and MaxBurst) of the endpoint of the peripheral device. To assess the impact of the selected parameter on the quality of services in the system, a statistical test for homoscedasticity of empirical distributions of isochronous intervals, was used. Besides the results of the conducted experiments for the selected parameters presented in the paper, the authors also tried to explain the obtained results in relation to the specification of the USB port.
