Abstract Porcine reproductive and respiratory disease virus (PRRSV) has the intrinsic ability to adapt and evolve. After 25 years of study, this persistent pathogen has continued to frustrate efforts ...to eliminate infection of herds through vaccination or other elimination strategies. The purpose of this review is to summarize the research on the virion structure, replication and recombination properties of PRRSV that have led to the extraordinary phenotype and genotype diversity that exists worldwide.
Madin-Darby Canine Kidney (MDCK) cells are widely utilized as a substrate for influenza virus isolation and propagation due to the high yields of virus. Here we compared the conventional MDCK cell ...line, MDCK-SIAT1 and MDCK-London for viral production, cell survival, and suitability in testing antivirals using six influenza strains including two H1N1 (pandemic and epidemic strains), three H3N2 and one influenza B strain. Overall our results suggest that MDCK-London cell line is superior for virus culturing and quantification, and hence an ideal platform to evaluate antiviral drug efficacy against multiple strains of influenza. Our data also suggests that while virus titers determined by the hemagglutination assay (HA) and neuraminidase activity (NA) are widely used to indicate viral load, there is a poor correlation between these measurements and the infectious titer obtained by plaque assay.
Objective was to compare estrous response, and first service and breeding season pregnancy rates in Angus cross beef heifers that received four progesterone (CIDR, vaginal insert) -based ...estrous-synchronization treatment regimens for timed artificial insemination (TAI) with or without short-term natural service (NS). Heifers (n = 1744; 4 locations) were assigned a reproductive tract score (RTS: 1 = immature, acyclic to 5 = mature, cyclic), body condition score (BCS: 1 = emaciated to 9 = obese) and temperament score (0 = calm to 1 = excitable). Within location, heifers were assigned to either of four treatment regimens. Heifers in Select-Synch + CIDR (C) + short-term NS (SSC + NS) treatment (n = 438) received 100 μg GnRH im + CIDR on Day −7 and CIDR removal + 25 mg PGF2α (im) + estrus-detection patches in the morning of Day 0 and were subjected to NS (bull to heifer ratio, 1:25) from the morning of Day 2 (48 h from CIDR removal) to evening of Day 8. Heifers in CO-Synch + C (COSC) +TAI + NS treatment (n = 433) received 100 μg GnRH + CIDR on Day −7 and CIDR removal + 25 mg PGF2α im + estrus-detection patches in the morning of Day 0 + 100 μg GnRH im + TAI on Day 2, 60 h from CIDR removal, and were subjected to natural service (bull: heifer, 1:25) from the evening of Day 2 immediately after AI through Day 8. Heifers in COSC + TAI treatment (n = 443) received 100 μg GnRH + CIDR on Day −7 and CIDR removal + 25 mg PGF2α im + estrus-detection patches in the morning of Day 0 + 100 μg GnRH im + TAI on Day 2, 60 h from CIDR removal. Heifers in SSC + split time AI (STAI) treatment (n = 430) received 100 μg GnRH + CIDR on Day −7 and CIDR removal + 25 mg PGF2α im + estrus-detection patches in the morning of Day 0. Heifers determined to be in estrus were inseminated at 64 h after PGF2α (25 mg, im) administration and non-estrous heifers determined to be in estrus 20 h later (at 84 h) were inseminated at that time, whereas remaining non-estrous heifers at 84 h received GnRH (100 μg) and were inseminated concomitantly. In SSC + NS and COSC + TAI + NS treatments, natural service bulls were removed and reintroduced (@ bull: heifer, 1:40) on Day 22 and kept until Day 63. In COSC + TAI and SSC + STAI bulls (bull: heifer, 1:40) were introduced on Day 14 and retained until Day 63. All heifers, irrespective of treatment regimens, were observed for estrus three times a day from Day 2 through Day 8. Pregnancy diagnosis was performed on Day 93, 30 days from end of breeding season. Accounting for temperament (P < 0.0001), BCS (P < 0.0001) and RTS (P < 0.0001), the breeding program influenced the estrous response (P < 0.0001). Heifers in COSC + TAI + NS (86.8%), SSC + STAI (84.9%), and SSC + NS (86.5%) treatments had greater estrous response compared with heifers in COSC + TAI (75.8%) treatment. Accounting for temperament (P < 0.01), BCS (P < 0.05) and RTS (P < 0.01), the treatment regimens influenced first service pregnancy rate (P < 0.05). Heifers in COSC + TAI + NS treatment had greater first service pregnancy rate (60.3%) compared with COSC + TAI (54.2%) (P < 0.05). First service pregnancy rate for heifers in SSC + STAI (59.3%) and SSC + NS (57.3%) treatments did not differ from COSC + TAI + NS and COSC + TAI treatments. Accounting for temperament (P < 0.001), BCS (P < 0.0001) and RTS (P < 0.0001), the treatment regimens influenced breeding season pregnancy rate (P < 0.05). Breeding season pregnancy rate for heifers in COSC + TAI + NS (95.4%) and COSC + STAI (94.5%) treatments were greater than that of heifers in COSC + TAI (90.8%) treatment (P < 0.05), and heifers in SSC + NS (94.1%%) treatment did not differ from heifers in other treatments. In conclusion, progesterone-based CO-Synch timed artificial insemination with short-term natural service treatment regimen resulted in proportionately more pregnancies than without short-term natural service treatment regimen. In addition, 64/84 h split-time AI or natural service following Select-Synch treatment regimen could be implemented as an alternative as these treatment regimens resulted in similar pregnancy rate as progesterone based CO-Synch timed artificial insemination with short-term natural service treatment regimen.

New optical sensors with a segmented photosensitive area are being developed for the next generation of neutrino telescopes at the South Pole. In addition to increasing sensitivity to high-energy ...astrophysical neutrinos, we show that this will also lead to a significant improvement in sensitivity to MeV neutrinos, such as those produced in core-collapse supernovae (CCSN). These low-energy neutrinos can provide a detailed picture of the events after stellar core collapse, testing our understanding of these violent explosions. We present studies on the event-based detection of MeV neutrinos with a segmented sensor and, for the first time, the potential of a corresponding detector in the deep ice at the South Pole for the detection of extra-galactic CCSN. Context. The emission and proper motion of the terminal hotspots of active galactic nucleus (AGN) jets can be used as a powerful probe of the intergalactic medium. However, measurements of hotspot ...advance speeds in active galaxies are difficult, especially in the young universe, because of the low angular velocities and the low brightness of distant radio galaxies. Aims. Our goal is to study the termination of an AGN jet in the young universe and to deduce physical parameters of the jet and the intergalactic medium. Methods. We used the LOw Frequency ARray (LOFAR) to image the long-wavelength radio emission of the high-redshift blazar S5 0836+710 on arcsecond scales between 120 MHz and 160 MHz. Results. The LOFAR image shows a compact unresolved core and a resolved emission region about 1.5 arcsec to the southwest of the radio core. This structure is in general agreement with previous higher-frequency radio observations with the Multi-Element Radio-Linked Interferometer Network (MERLIN) and the Very Large Array (VLA). The southern component shows a moderately steep spectrum with a spectral index of about ≳ − 1, and the spectral index of the core is flat to slightly inverted. In addition, we detect for the first time a resolved steep-spectrum halo with a spectral index of about −1 surrounding the core. Conclusions. The arcsecond-scale radio structure of S5 0836+710 can be understood as a Faranoff–Riley (FR) II radio galaxy observed at a small viewing angle. The southern component can be interpreted as the region of the approaching jet’s terminal hotspot, and the halo like diffuse component near the core can be interpreted as the counter-hotspot region. From the differential Doppler boosting of both features, we can derive the hotspot advance speed to (0.01 − 0.036) c. At a constant advance speed, the derived age of the source would exceed the total lifetime of such a powerful FR II radio galaxy substantially. Thus, the hotspot advance speed must have been higher in the past, in agreement with a scenario in which the originally highly relativistic jet has lost collimation as a result of instability growth and has transformed into an only mildly relativistic flow. Our data suggest that the density of the intergalactic medium around this distant (z = 2.22) AGN could be substantially higher than the values typically found in less distant FR II radio galaxies.
► We tracked seabirds of the tropical Indian Ocean to identify major foraging hotspots. ► Five large oceanic areas have both high seabird densities and high seabird diversity. ► All of them overlap ...with potential threats showing that seabirds may be at risk when foraging. ► These information are crucial to identify potential Marine Protected Areas in the Indian Ocean.
We conducted a regional tracking program on seabirds in order to identify major forging hotspots and potential Marine Protected Areas in the tropical western Indian Ocean. Thirty-one species of seabirds breed in the region, totaling 7.4 million pairs. The main breeding grounds are in the Seychelles, in the Mozambique Channel and in the Mascarene. Seven pelagic species have been tracked so far from eight different islands of the region. Using count per sector analysis we identified five major oceanic foraging hotspots, among which three include the breeding colonies and two are oceanic areas not connected to a breeding island. We found important overlaps between most of these seabird foraging hotspots and potential threats (industrial fishery targeting surface dwelling tunas and marine pollution due to maritime routes) suggesting that in these regions seabirds may be at risk when foraging. Although this analysis is based on a limited number of tracking studies, the knowledge on seabird distribution at sea has increased tremendously in the last 6years in the tropical western Indian Ocean, and this trend will continue, as research is ongoing. The data, we present here for the first time in a single synthesis show clear spatial patterns that identify high priority locations for designation as Marine Protected Areas in the tropical western Indian Ocean.
IceCube is the first representative of the km3 class of neutrino telescopes and currently the most sensitive detector to high-energy neutrinos. Its main mission is to search for Galactic and ...extragalactic sources of high-energy neutrinos, but it is also an excellent detector for the investigation of a variety of other highly topical astrophysics and particle physics topics like supernovae, dark matter and neutrino oscillations. After an introduction to neutrino astronomy and neutrino telescopes, this article presents a selection of latest results from the IceCube neutrino detector with respect to searches for cosmic high-energy neutrino sources.
A goal of seabird ecology is to relate the physiology, population dynamics, distribution and behaviour of seabirds with their biotic and abiotic environments. One of the most challenging aspects is ...to understand how seabirds interact with their environment when direct observations are not always possible or practical. In the present paper, we reviewed 218 published studies that examined associations between seabird distribution, behaviour and their environment, in order to assess current trends, weaknesses and the future directions of research. Based on the number of publications, it is evident that the field is growing rapidly and that methods for evaluating seabird distribution are becoming increasingly more sophisticated and are changing from Eulerian (grid-like) to Lagrangian (particle-like) data types. This has been accompanied by a reduction in the spatial and temporal scale of observation, where, in most cases, no behavioural information is inferred from Lagrangian data; instead they are often used as if they were Eulerian data. In parallel, environmental remote sensing is becoming more common; however, we did not record significant changes in the statistical approaches used to describe seabird distributions and used to link them with oceanographic variables. In particular, despite the spatially explicit nature of the data, spatial statistics have rarely been used. The vast majority of studies used environmental variables that described water masses (descriptive approach), whereas a few studies determined oceanographic features that enhance prey availability to seabirds (process-based approach). Future studies could enhance their ecological interpretation of seabird–environment interactions by making greater use of ad hoc statistical approaches that facilitate appropriate pattern detection (e.g. area-restricted searching pattern for birds, mesoscale patterns for environment). Furthermore, appropriate hypothesis testing and modelling that accounts for the spatially explicit, multiscale and multivariate nature of the interaction between seabirds and their habitats is recommended. Although quantitative methods currently exist (but are rarely used), further application could greatly improve our understanding of the processes linking seabird distribution to their environment.
