Kinase inhibitors are important cancer therapeutics. Polypharmacology is commonly observed, requiring thorough target deconvolution to understand drug mechanism of action. Using chemical proteomics, ...we analyzed the target spectrum of 243 clinically evaluated kinase drugs. The data revealed previously unknown targets for established drugs, offered a perspective on the "druggable" kinome, highlighted (non)kinase off-targets, and suggested potential therapeutic applications. Integration of phosphoproteomic data refined drug-affected pathways, identified response markers, and strengthened rationale for combination treatments. We exemplify translational value by discovering SIK2 (salt-inducible kinase 2) inhibitors that modulate cytokine production in primary cells, by identifying drugs against the lung cancer survival marker MELK (maternal embryonic leucine zipper kinase), and by repurposing cabozantinib to treat FLT3-ITD-positive acute myeloid leukemia. This resource, available via the ProteomicsDB database, should facilitate basic, clinical, and drug discovery research and aid clinical decision-making.
Phosphorus retention in lowland soils depends on redox conditions. The aim of this study was to evaluate how the Fe(III) reduction degree affects phosphate adsorption and precipitation. Two similarly ...P‐saturated, ferric Fe‐rich lowland soils, a sandy and a peat soil, were incubated under anaerobic conditions. Mössbauer spectroscopy demonstrated that Fe(III) in the sandy soil was present as goethite and phyllosilicates, whereas Fe(III) in the peat soil was mainly present as polynuclear, Fe‐humic complexes. Following anoxic incubation, extensive formation of Fe(II) in the solids occurred. After 100 d, the Fe(II) production reached its maximum and 34% of the citrate–bicarbonate–dithionite extractable Fe (FeCBD) was reduced to Fe(II) in the sandy soil. The peat soil showed a much faster reduction of Fe(III) and the maximum reduction of 89% of FeCBD was reached after 200 d. Neoformation of a metavivianite/vivianite phase under anoxic conditions was identified by X‐ray diffraction in the peat. The sandy soil exhibited small changes in the point of zero net sorption (EPC0) and Pi desorption with increasing Fe(III) reduction, whereas in the peat soil Pi desorption increased from 80 to 3100 μmol kg−1 and EPC0 increased from 1.7 to 83 μM, after 322 d of anoxic incubation. The fast Fe(III) reduction made the peat soils particularly vulnerable to changes in redox conditions. However, the precipitation of vivianite/metavivianite minerals may control soluble Pi concentrations to between 2 and 3 μM in the long term if the soil is not disturbed.

STXBP2 encodes syntaxin binding protein 2 (Munc18-2), involved in the regulation of vesicle transport to the plasma membrane. We have identified syntaxin 11, a SNARE protein mutated in FHL-4, as an interaction partner of STXBP2. This interaction is eliminated by the missense mutations found in our FHL-5 patients, which leads to a decreased stability of both proteins, as shown in patient lymphocytes. Activity of natural killer and cytotoxic T cells was markedly reduced or absent, as determined by CD107 degranulation. Our findings thus identify a key role for STXBP2 in lytic granule exocytosis.

In the cirrus cloud regime, the surface wettability and pore abundance of soot particles are important determinants for their ice nucleation ability
via
pore condensation and freezing. In the atmosphere, soot particles can undergo various ageing processes which modify their surface chemistry and porosity, thus acting as ice nucleating particles with varying abilities as a function of ageing. In this study, size-selected soot particles were treated by thermal denuding at 573 K in a pure nitrogen (N
2
) or synthetic air (N
2
+ O
2
) flow and then exposed to varying relative humidity conditions at a fixed temperature in the range from 218 to 243 K, to investigate the role of volatile content in the ice nucleation ability. Both organic-lean and organic-rich propane (C
3
H
8
) flame soot particles, as well as two types of commercially available carbon black soot particles with high and low surface wettability, were tested. The size and mass distribution of soot aerosol were monitored during the ice nucleation experiments. Bulk soot samples also prepared in pure N
2
or synthetic air environments at 573 K were characterised by thermogravimetric analysis, Fourier transform infrared spectroscopy and dynamic vapour sorption measurements, to reveal the relation between denuding volatile content, associated soot particle property modifications and the ice nucleation ability. Our study shows that thermal denuding induces a change in soot particle porosity playing a dominant role in regulating its ice nucleation
via
the pore condensation and freezing mechanism. The enrichment in mesopore (2-50 nm) availability may enhance soot ice nucleation. The presence of O
2
in the thermal denuding process may introduce new active sites on soot particles for water interaction and increase soot surface wettability. However, these active sites only facilitate soot ice nucleation when mesopore structures are available. We conclude that a change in volatile content modifies both morphological properties and surface chemistry for soot particles, but porosity change plays the dominant role in regulating soot particle ice nucleation ability.
The removal of volatile content enhances the ice nucleation of soot particles by increasing the particle porosity and surface wettability.
Before the incorporation of the 13-valent pneumococcal conjugate vaccine (PCV13) into the childhood vaccination regimen in Greenland in 2010, Inuit populations experienced a substantial prevalence of ...invasive pneumococcal disease (IPD). The PCV13 introduction has been shown to markedly reduce the incidence of IPD. This current study estimated the impact of PCV13 introduction on IPD mortality in Greenland. This was a nationwide register-based study using all available data on IPD cases 1995-2020 in Greenland. Thirty-one-day IPD case fatality rates (CFR), and all-cause and mortality rates associated with IPD during the period before the introduction of PCV13 (January 1995 to September 2010) were compared with those observed in the post-PCV13 era (September 2010 to October 2020). Standardized mortality ratios (SMRs) expressed differences in mortality by sex, age, region, ethnicity, comorbidity, and serotype. IPD CFR decreased with 24.5% from the pre- to the post-PCV13 period. SMR in IPD patients decreased by 57% (95% CI, 36-75%), and a reduction occurred in all age groups. While SMR in IPD persons ≥60 years remained virtually unchanged, there were no IPD-related deaths in persons ≤39 years in the post-PCV13 period. In conclusion, IPD-related mortality has decreased in Greenland following PCV13 introduction in 2010 in the country.
We have previously documented a high incidence of tuberculosis (TB) (>1000 per 100 000 inhabitants) and considerable Mycobacterium tuberculosis transmission in East Greenland despite population-wide ...screenings implemented by the Greenland health authorities 1–5. In Greenland, all contacts of TB patients are routinely screened for M. tuberculosis infection and TB. Individuals with newly acquired M. tuberculosis infection are offered 6 months of preventive isoniazid treatment. Despite the considerable resources spent on clinical examinations and preventive treatment over the years, TB incidence rates have remained high in East Greenland 1.
The use of clay nanofillers offers a potential route to improved barrier properties in polylactide films. Magnesium–aluminium layered double hydroxides (LDHs) are interesting in this respect and we ...therefore explored synthesis of PLA-LDH nanocomposites by ring-opening polymerization. This method is attractive because it should ensure good dispersion of LDH in the polymer. The effect of adding either LDH carbonate (LDH-CO
3) or laurate-modified LDH (LDH-C
12) was investigated. X-ray diffraction, scanning electron microscopy, and transmission electron microscopy revealed that exfoliated nanocomposites were obtained when using LDH-C
12 but that LDH-CO
3 gave a partly phase-separated morphology. Thermogravimetric analysis showed that PLA-LDH combinations exhibited higher degradation onset temperatures and differential scanning calorimetry confirmed that LDHs can act as nucleating agents. However, PLA molecular weight was significantly reduced when
in-situ polymerization was conducted in the presence of the LDHs and we suggest that chain termination via LDH surface hydroxyl groups and/or metal-catalyzed degradation could be responsible.
The 13-valent pneumococcal conjugate vaccine (PCV13) was introduced in 2010 to the childhood vaccination program in Greenland. This study aimed to estimate the effectiveness of the PCV13 on the ...incidence of invasive pneumococcal disease (IPD) in children and in adults in Greenland. IPD cases from the pre-PCV13 period (January 1995–September 2010) were compared with the post-PCV13 period (September 2010–October 2020). Register data were collected from laboratory records, IPD reports, the national registry on admissions, and medical files. A total of 295 IPD cases were identified in the study period. Overall IPD incidence rate (IR) declined from the pre-PCV13 period to the post-PCV13 period (IR 23.3 to 15.3 per 100,000 person years). Overall IPD incidence among children decreased significantly, whereas overall IPD incidence among the elderly increased significantly. During the post-PCV13 period, the incidence of vaccine serotype (VT) IPD decreased in all ages, while the incidence of non-vaccine serotype (NVT) IPD increased. This increase was most substantial among elderly ≥60 years. In conclusion, the PCV13 has reduced incidence rates of IPD in Greenland. However, the increase in NVT IPD among the elderly is noteworthy, and sup-ports continued surveillance of IPD in the population of Greenland.
Chronic recalcitrant dermatophytoses, due to Trichophyton (T.) mentagrophytes Type VIII are on the rise in India and are noteworthy for their predominance. It would not be wrong to assume that travel ...and migration would be responsible for the spread of T. mentagrophytes Type VIII from India, with many strains resistant to terbinafine, to other parts of the world. From September 2016 until March 2020, a total of 29 strains of T. mentagrophytes Type VIII (India) were isolated. All patients were residents of Germany: 12 females, 15 males and the gender of the remaining two was not assignable. Patients originated from India (11), Pakistan (two), Bangladesh (one), Iraq (two), Bahrain (one), Libya (one) and other unspecified countries (10). At least two patients were German-born residents. Most samples (21) were collected in 2019 and 2020. All 29 T. mentagrophytes isolates were sequenced (internal transcribed spacer (ITS) and translation elongation factor 1-α gene (TEF1-α)). All were identified as genotype VIII (India) of T. mentagrophytes. In vitro resistance testing revealed 13/29 strains (45%) to be terbinafine-resistant with minimum inhibitory concentration (MIC) breakpoints ≥0.2 µg/mL. The remaining 16 strains (55%) were terbinafine-sensitive. Point mutation analysis revealed that 10/13 resistant strains exhibited Phe397Leu amino acid substitution of squalene epoxidase (SQLE), indicative for in vitro resistance to terbinafine. Two resistant strains showed combined Phe397Leu and Ala448Thr amino acid substitutions, and one strain a single Leu393Phe amino acid substitution. Out of 16 terbinafine-sensitive strains, in eight Ala448Thr, and in one Ala448Thr +, new Val444 Ile amino acid substitutions were detected. Resistance to both itraconazole and voriconazole was observed in three out of 13 analyzed strains. Treatment included topical ciclopirox olamine plus topical miconazole or sertaconazole. Oral itraconazole 200 mg twice daily for four to eight weeks was found to be adequate. Terbinafine-resistant T. mentagrophytes Type VIII are being increasingly isolated. In Germany, transmission of T. mentagrophytes Type VIII from the Indian subcontinent to Europe should be viewed as a significant public health issue.
