We present a combined strong and weak lensing analysis of the J085007.6+360428 (J0850) field, which contains the massive cluster Zwicky 1953. This field was selected for its high projected ...concentration of luminous red galaxies. Using Subaru/Suprime-Cam imaging and MMT/Hectospec spectroscopy, we first perform a weak lensing shear analysis to constrain the mass distribution in this field, including the cluster at z = 0.3774 and a smaller foreground halo at z = 0.2713. We then add a strong lensing constraint from a multiply imaged galaxy in the imaging data with a photometric redshift of z 5.03. Unlike previous cluster-scale lens analyses, our technique accounts for the full three-dimensional mass structure in the beam, including galaxies along the line of sight. In contrast with past cluster analyses that used only lensed image positions as constraints, we use the full surface brightness distribution of the images. This method predicts that the source galaxy crosses a lensing caustic, such that one image is a highly magnified "fold arc" that could be used to probe the source galaxy's structure at ultra-high spatial resolution (<30 pc). We calculate the mass of the primary cluster to be with a concentration of , consistent with the mass-concentration relation of massive clusters at a similar redshift. The large mass of this cluster makes J0850 an excellent field for leveraging lensing magnification to search for high-redshift galaxies, competitive with and complementary to that of well-studied clusters such as the HST Frontier Fields.
We aimed to investigate the relationship between measures of HIV persistence with antiretroviral therapy (ART) and cigarette smoking, systemic markers of inflammation, and pulmonary function. ...Retrospective study of 82 people with HIV (PWH) on ART for a median of 6.9 years (5.6-7.8) and plasma HIV RNA levels <50 copies/mL. HIV DNA and cell-associated HIV RNA (CA-RNA) were measured in peripheral blood mononuclear cells (PBMC) and plasma HIV RNA was measured by single-copy assay (SCA). Plasma levels of 17 inflammatory mediators were measured by Bio-Plex, and standard pulmonary function tests (PFT) were performed in all participants. Median age was 52 years and 41% were women. Most had preserved CD4+ T cell counts (median (IQR) 580 (361-895) cells/mm3). Median plasma HIV RNA was 1.3 (0.7-4.6) copies/mL, and median levels of HIV DNA and CA-RNA in PBMC were 346 (140-541) copies and 19 (3.7-49) copies per 1 million PBMC, respectively. HIV DNA was higher in smokers than in nonsmokers (R = 0.3, P < 0.05), and smoking pack-years positively correlated with HIV DNA and CA-RNA (R = 0.3, P < 0.05 and R = 0.4, P < 0.01, respectively). HIV DNA, CA-RNA, and plasma HIV RNA were not significantly associated with any measure of pulmonary function or inflammation. Cigarette smoking was associated with HIV DNA and CA-RNA levels in blood, but measures of HIV persistence were not associated with pulmonary function or inflammation.
Lysophosphatidylcholine acyltransferase 1 (LPCAT1), the enzyme catalyzing the reaction in remodeling of phosphatidylcholine (PC) has been reported to express in prostate. However, its diagnostic and ...prognostic values remain unclear.
Immunohistochemistry (IHC) for LPCAT1was performed on the tissue microarray (TMA) slides containing 251samples from 148 patients with various prostatic disorders. The association of expression level of LPCAT1 with the progression of prostate cancer was analyzed.
LPCAT1 IHC mean score was the highest in metastatic prostate cancer (8.00±1.28), which was significantly higher than that in primary prostate cancer (4.63±3.00, p=9.73E-07), in high grade prostatic intraepithelial neoplasia (HGPIN, 2.72±2.47, p=1.02E-12), and in benign prostate (2.68, p=6.17E-12). The mean score in primary prostate cancer was significantly higher than that in HGPIN (p=4.09E-04) and in benign prostate (p=2.74E-04). There was no significant difference in the mean score between HGPIN and benign prostate (p=0.951). LPCAT1 IHC score also correlated to the tumor grade and stage of prostate cancer. Patients who underwent prostatectomy for prostate cancer and developed biochemical recurrence or clinical metastasis had higher LPCAT1 IHC score than those who underwent prostatectomy for prostate cancer and did not develop biochemical recurrence and clinical metastasis. The association of LPCAT1 with the progression of prostate cancer was independent of patient race and age, PSA level and positivity of surgical resection margins.
LPCAT1 correlates with the progression of prostate cancer and could be a new biomarker in diagnosis, prognosis and studying the pathogenesis of prostate cancer.

We investigate the flow-channeling phenomenon caused by thermal drawdown in fractured geothermal reservoirs. A discrete fracture network-based, fully coupled thermal–hydrological–mechanical simulator ...is used to study the interactions between fluid flow, temperature change, and the associated rock deformation. The responses of a number of randomly generated 2D fracture networks that represent a variety of reservoir characteristics are simulated with various injection-production well distances. We find that flow channeling, namely flow concentration in cooled zones, is the inevitable fate of all the scenarios evaluated. We also identify a secondary geomechanical mechanism caused by the anisotropy in thermal stress that counteracts the primary mechanism of flow channeling. This new mechanism tends, to some extent, to result in a more diffuse flow distribution, although it is generally not strong enough to completely reverse flow channeling. We find that fracture intensity substantially affects the overall hydraulic impedance of the reservoir but increasing fracture intensity generally does not improve heat production performance. Increasing the injection-production well separation appears to be an effective means to prolong the production life of a reservoir.
Product design experts depend on online customer reviews as a source of insight to improve product design. Previous works used aspect-based sentiment analysis to extract insight from product reviews. ...However, their approaches for requirements elicitation are less flexible than traditional tools such as interviews and surveys. They require costly data labeling or pre-labeled datasets, lack domain knowledge integration, and focus more on sentiment classification than flexible aspect-opinion analysis. Related works lack effective mechanisms for probing the customer feedback of complex configurable products. This study proposes a generic graph-based opinion mining and analysis method for product design improvement. First, a customer feedback data preprocessing and annotation pipeline that can incorporate designer-specified domain knowledge is proposed. Second, an intuitive opinion-aware labeled property graph data model is designed to ingest preprocessed feedback data and perform ad hoc opinion analysis. Applying the generic model to a real-world dataset demonstrates superior functionality and flexibility compared to related works. A wider range of analyses is supported in a single model without repeating data preprocessing and modeling. Specifically, the proposed method supports regular and comparative aspect-opinion analysis, aspect satisfaction/influence ranking, opinion trend extraction, and targeted aspect-opinion summarization.
Depressive symptoms are prevalent in individuals living with sickle cell disease (SCD) and may exacerbate pain. This study examines whether higher depressive symptoms are associated with pain ...outcomes, pain catastrophizing, interference and potential opioid misuse in a large cohort of adults with SCD. The study utilized baseline data from the 'CaRISMA' trial, which involved 357 SCD adults with chronic pain. Baseline assessments included pain intensity, daily mood, the Patient Health Questionnaire (PHQ), the Generalized Anxiety Disorders scale, PROMIS Pain Interference, Pain Catastrophizing Scale, the Adult Sickle Cell Quality of Life Measurement Information System and the Current Opioid Misuse Measure. Participants were categorized into 'high' or 'low' depression groups based on PHQ scores. Higher depressive symptoms were significantly associated with increased daily pain intensity, negative daily mood, higher pain interference and catastrophizing, poorer quality of life and a higher likelihood of opioid misuse (all p < 0.01). SCD patients with more severe depressive symptoms experienced poorer pain outcomes, lower quality of life and increased risk of opioid misuse. Longitudinal data from this trial will determine whether addressing depressive symptoms may potentially reduce pain frequency and severity in SCD.

novel treatment for postmenopausal women with hormone-dependent ...breast cancer. Estrone and DHEA are formed by this sulfatase
pathway and can be converted to steroids (estradiol and androstenediol, respectively), which have potent estrogenic properties.
Experimental Design: STX64 (667 Coumate), a tricylic coumarin-based sulfamate that irreversibly inhibits STS activity, was selected for entry
into the first phase I trial of a STS inhibitor in postmenopausal women with breast cancer. STX64 was administered orally
(nine patients at 5 mg and five patients at 20 mg) as an initial dose followed 1 week later by 3 × 2 weekly cycles, with each
cycle comprising daily dosing for 5 days followed by 9 days off treatment. Blood and tumor tissue samples were collected for
the assessment of STS activity and serum was obtained for steroid hormone measurements before and after treatment.
Results: The median inhibition of STS activity by STX64 was 98% in peripheral blood lymphocytes (PBL) and 99% in breast tumor tissue
at the end of the 5-day dosing period. As expected, serum concentrations of estrone, estradiol, androstenediol, and DHEA all
decreased significantly from pretreatment levels. Unexpectedly, androstenedione and testosterone concentrations also decreased.
Four patients, all of whom had previously progressed on aromatase inhibitors, showed evidence of stable disease for 2.75 to
7 months. The drug was well tolerated with only minor drug-related adverse events recorded.
Conclusion: STX64 is a potent, well-tolerated STS inhibitor. It inhibits STS activity in PBLs and tumor tissues and causes significant
decreases in serum concentrations of steroids with estrogenic properties.

regions and supernova remnants (SNRs). Using low- and high-frequency surveys to constrain free–free and thermal dust emission contributions, we find evidence of excess flux density at 30 GHz in six regions, which we interpret as anomalous microwave emission. Furthermore we model ultracompact H
ii
contributions using data from the 5 GHz CORNISH catalog and reject these as the cause of the 30 GHz excess. Six known SNRs are detected at 30 GHz, and we measure spectral indices consistent with the literature or show evidence of steepening. The flux density of the SNR W44 at 30 GHz is consistent with a power-law extrapolation from lower frequencies with no indication of spectral steepening in contrast with recent results from the Sardinia Radio Telescope. We also extract five hydrogen radio recombination lines (RRLs) to map the warm ionized gas, which can be used to estimate electron temperatures or to constrain continuum free–free emission. Diffuse malignant gliomas, the most common type of brain tumor, carry a dire prognosis and are poorly responsive to initial treatment. The response to treatment is typically evaluated by measurements ...obtained from radiographic images several months after the start of treatment; therefore, an early biomarker of tumor response would be useful for making early treatment decisions and for prognostic information. Thirty-four patients with malignant glioma were examined by diffusion MRI before treatment and 3 weeks later. These images were coregistered, and differences in tumor-water diffusion values were calculated as functional diffusion maps (fDM), which were correlated with the radiographic response, time-to-progression (TTP), and overall survival (OS). Changes in fDM at 3 weeks were closely associated with the radiographic response at 10 weeks. The percentage of the tumor undergoing a significant change in the diffusion of water (VT) was different between patients with progressive disease (PD) vs. stable disease (SD) (P < 0.001). Patients classified as PD by fDM analysis at 3 weeks were found to have a shorter TTP compared with SD (median TTP, 4.3 vs. 7.3 months; P < 0.04). By using fDM, early patient stratification also was correlated with shorter OS in the PD group compared with SD patients (median survival, 8.0 vs. 18.2 months; P < 0.01). On the basis of fDM, tumor assessment provided an early biomarker for response, TTP, and OS in patients with malignant glioma. Further evaluation of this technique is warranted to determine whether it may be useful in the individualization of treatment or evaluation of the response in clinical protocols.
